RNA-sequencing of inguinal adipose tissue from wild type and adipocyte specific HIF2a knock-out mice upon thermoneutral and cold exposure

Project description:Adipose tissue plays an important role in the regulation of wholebody energy homeostasis, depending on external environments. Upon cold acclimation, brown and beige adipocytes dissipate mitochondrial proton gradient generating heat via uncoupling protein 1. Due to increasing demands for oxygen during thermogenesis, adipose tissue could become hypoxic upon cold exposure. Hypoxia-inducible factors (HIFs) are one of the major transcription factors which are involved in various cellular responses under hypoxic condition. Although HIFs regulate diverse metabolic processes with selective target gene expression, their roles in thermogenic adipocytes are still elusive. In this study, we demonstrate that adipocyte HIF2a regulate thermogenic activity upon cold stimuli. To investigate molecular mechanisms of adipocyte HIF2a upon thermoneutral and cold exposure, we report the transcriptomes of inguinal adipose tissue from WT and adipocyte specific HIF2a knock-out mice.

Expression data in inguinal adipose tissue at different circadian times

Project description:Peripheral circadian clocks regulate many aspects of physiology. In this study we deleted the core circadian clock component Bmal1 specifically in mouse adipocytes in order to study the role of the adipocyte clock in energy homeostasis and body weight. We used microarrays to indentify changes in gene expression in the adipose tissue of mice lacking a functional adipocyte circadian clock and identified a small number of up- and down- regulated genes. Adipose tissues were isolated from inguinal adipose fat pads at four different times of the diurnal cycle under constant darkness (circadian time, CT) for RNA extraction and hybridization on Affymetrix microarrays. Adipocyte-specific Bmal1 KO (Ad-Bmal1-/-) and control mice were used for isolation of tissues at CT0, CT6, CT12, CT18 where CT0 the beginning of the subjective day.

Interaction between PPARγ and mTORC1 in the regulation of inguinal white adipose tissue transcriptome

Project description:Purpose: To investigate the involvement of mTORC1 as a mediator of the actions of the PPARγ ligand rosiglitazone in subcutaneous inguinal white adipose tissue transcriptome; Methods: Mice bearing regulatory associated protein of mTOR (Raptor) deletion and therefore mTORC1 deficiency exclusively in adipocytes (adiponectin Cre recombinase) and littermate controls were fed a high-fat diet supplemented or not with the PPARγ agonist rosiglitazone (30 mg/kg/day) for 8 weeks and evaluated for inguinal white adipose tissue transcriptome (Rnaseq); Results: 3,2425 genes had their correspondent mRNA levels altered by either adipocyte Raptor deficiency or rosiglitazone administration or their combination. Among those, 408 genes modulated by rosiglitazone required mTORC1. Conclusion: PPARγ and mTORC1 are essential partners in the regulation of a cluster of genes in inguinal white adipose tissue.

Project description:In order to select mRNA transcripts strongly enriched in murine white adipocytes versus brown adipocytes or stromal-vascular fraction, gene expression data of the adipocyte and stromal-vascular fractions of the interscapular brown, inguinal subcutaneous as well as visceral epididymal adipose tissue depots of young adult male C57BL/6 mice housed at constant 23Â°C ambient temperature were obtained. 18 samples: 3 different adipose tissues separated into stromal-vascular fraction and adipocytes, analyzed in biological triplicates.

The Transcription Factor ATF7 Controls Adipocyte Differentiation and Thermogenic Gene Programming

Project description:The adipocytes functions as a central organ in the regulation of metabolic homeostasis. Factors which contribute to the adipocyte differentiation and function would be the promising targets to combat the obesity and associated metabolic disorders. The activating transcription factor 7 (ATF7), a stress-responsive chromatin regulator, has recently been shown to be involved in the energy metabolism; however, the underlying mechanisms are still unknown. Here, we show that ATF7 is required for adipocyte differentiation and it interacts with histone dimethyltransferase G9a in adipocyte to repress the interferon-stimulated genes (ISGs) expression, which suppresses adipogenesis. Ablation of ATF7 promotes the beige biogenesis and browning of inguinal white adipose tissue (iWAT). ATF7 binds to the transcription regulatory regions of Ucp1 gene, and silences it by maintaining the histone H3K9 dimethylation level. These results establish the multifunction of ATF7 in adipocyte and provide molecular insights into the epigenetic control of development and function of adipose tissues. We used microarrays to identify the differentatial expression genes in the inguinal white adipose tissue of ATF7 KO mice.
