Project description:Hepatic cirrhosis or advanced fibrosis caused by chronic hepatitis may be the main risk factor contributing to the occurrence of HCC. Growing clinical evidence show that patients with hepatic cirrhosis or advanced fibrosis may have a 7-fold higher incidence rate of HCC than those without, suggesting a crucial role of inflammation-carcinogenesis malignant transformation in HCC development and progression. Project description:ChREBP was stably overexpressed in the liver of C57BL/6J mice through the use of the sleeping beauty transposon system. ChREBP overexpression resulted in HCC tumor development in 12 months. We used microarray analysis to detail the global program of gene expression in ChREBP overexpressing tumors compared to non-tumoral adjacent tissue or compared with normal liver samples from C57BL/6J mice. WE then identified distinct classes of up and down regulated genes.

Applied Biosystems Human TaqMan Low Density Array (TLDA) in HCC tumor and adjacent non-tumor tissues

Project description:MicroRNAs (miRNAs) are aberrant expressed in hepatocellular carcinoma (HCC) tissue and play a central role in diverse biological processes. We conducted a genome-wide miRNAs screening in 10 pairs of HCC tumor and adjacent non-tumor tissues to test the hypothesis that dysregulation of miRNAs in HCC tumor tissue are partially due to aberrant methylation in relevant miRNAs host genes. Taqman low density arrays were used to examine miRNA profiles in paired HCC tissues, and quantitative RT-PCR was used to validate candidate miRNAs for both discovery and validation sets. A cross-sectional study was conducted in 10 HCC tumor tissues and 10 adjacent non-tumor tissues in Columbia University Medical Center (CUMC), which is approved by the Institutional Review Board.

Methylation in Taiwanese HCC tumor and adjacent tissues

Project description:We analyzed tumor and adjacent non-tumor tissues from 62 Taiwanese HCC cases using Illumina HumanMethylation 27 chips. After Bonferroni adjustment, a total of 2,324 CpG sites significantly differed in methylation level, with 684 CpG sites significantly hypermethylated and 1,640 hypomethylated in tumor compared to non-tumor tissues. Transcriptome analysis in hepatocellular carcinoma (HCC) tissues

Project description:We report the application of RNA sequencing technology for high-throughput profiling in HCC tissues. In the present study, to explore novel biomarkers of HCC, we firstly explored the expression profiles of mRNAs and miRNAs in three pairs of HCC patients (tumor tissues and non-tumorous tissues) by high-throughput RNA sequencing. A total of 1024 mRNAs were found to be significantly dysregulated (636 up-regulated and 388 down-regulated), 50 miRNAs were found to be significantly dysregulated (27 up-regulated and 23 down-regulated) in the HCC tissues compared with the non-tumorous tissues.Then validae the differentially expressed miRNAs by qRT-PCR in HCC tissues and serum. This study provides a framework for the application of miRNAs to be ideal biomarkers for HCC.

SNP-based LOH of hepatocellular carcinoma (HCC)

Project description:High frequency of loss of heterozygosity (LOH) at locus D4S2964 was found in hepatocellular carcinoma (HCC) by our previous studies and others’. The present study was designed to explore genes around the locus affected by LOH and their clinical implications. 440 SNPs located at 49 genes around D4S2964 were selected from NCBI website for the SNPs microarray fabrication. LOH of these SNPs markers in 112 cases of HCC tissues and paired adjacent liver tissues were investigated by the SNP microarray. A map of LOH of SNPs in genes around D4S2964 was constructed. 112 pairs of HCC tissues and corresponding non-tumor tissues was genotyped. LOH and its clinical implication was analyzed.
