Morphology and transcriptome of Atlantic salmon skin after seawater transfer, comparison of a semi-closed containment system and open net-pen

ABSTRACT: Development of semi-closed containment systems is a promising strategy to further expand Atlantic salmon production. The technology operates with higher production intensities where skin integrity is recognized as a central factor for animal health and welfare. This study investigate the skin development and its immune status in healthy Atlantic salmon growers reared in a semi-closed containment system and a traditional open net-pen system (control).

Effect of photoperiod and seawater tranfer on transcriptome of saccus vasculosus from Atlantic salmon

Project description:Atlantic salmon (Salmo salar) move from fresh- to seawater environments following a seasonally timed preparative transition called smoltification, which takes place under photoperiodic control in the freshwater environment. In masu salmon (Oncorhynchus masou), coordination of photoperiodic sexual maturation is proposed to involve in a fish-specific circumventricular organ, the saccus vasculosus (SV), through its intrinsic opsin-based light sensitivity, thyrotrophin secretion and modulation of deiodinase activity (TSH-DIO cascade). The saccus vasculosus is a highly vascularized structure located on the ventral side of the hypothalamus and its interface between the blood and cerebrospinal fluid also hints at a role in ionic balance of the cerebrospinal fluid (CSF). Both the potential photoperiodic and ionic functions of the SV led us to perform transcriptome analysis of the SV in smoltification in Atlantic salmon. Our data show that SV response to seawater exposure is highly dependent on photoperiodic history and identifies ependymin as a major secretory output of the SV, consistent with a role in control of CSF composition. Conversely, we could not detect crucial elements of the opsin-TSH-DIO cascade suggesting that the photoperiodic history-dependence of the SV to seawater exposure is unlikely to stem from SV-intrinsic responses to photoperiod.

Effects of yeast species and processing on transcriptomic profile and intestinal health of Atlantic salmon (Salmo salar) fed soybean meal-based diets in seawater

Project description:The objective of the current study was to examine the effects of yeasts on intestinal health and transcriptomic profile of Atlantic salmon fed SBM-based diets in seawater. Cyberlindnera jadinii (CJ) and Wickerhamomyces anomalus (WA) yeasts were produced in-house and processed by direct heat-inactivation with spray-drying (ICJ and IWA) or autolyzed at 50 ºC for 16 h (ACJ and AWA), followed by spray-drying. Six diets were formulated, one based on fishmeal (FM), a challenging diet with 30% soybean meal (SBM) and four other diets containing 30% SBM and 10% of each of the four yeast fractions (i.e., ICJ, ACJ, IWA and AWA). The results showed that the inclusion of CJ yeasts reduced loss of supranuclear vacuolization, along with reduction in population of CD8α positive cells present in the lamina propria of fish fed the SBM diet. The CJ yeasts controlled the inflammatory profile of fish fed SBM through up-regulation of pathways related to wound healing and taurine metabolism. Additionally, the WA yeasts dampened the inflammatory profile of fish fed SBM through down-regulation of pathways related to toll-like receptor signaling, C-lectin receptor, cytokine receptor and signal transduction. The results suggest that yeasts could be used as protein ingredients with functional properties in diets for Atlantic salmon.

Differential transcriptomic response of Atlantic salmon to fermented sunflower meal under commercial seawater farming conditions

Project description:The study focused on the differential transcriptome of immune organs (i.e., head kidney and spleen) from farmed Atlantic salmon (Salmo salar) fed fermented sunflower meal (FSFM) at 5% and 10% in two commercial farms (CS: coast and FD: fjord location). Data showed that both head kidney and spleen (from fish fed FSFM5% at CS) up-regulated mRNA biogenesis (linked to lipid metabolism and mRNA surveillance) as well as glycosyltransferases (associated with vitamin B6 and lipoic acid metabolism, or pantothenate and CoA biosynthesis). Regarding FD, head kidney from FSFM10% group had up-regulated enzymes (related to lysosome, glutathione metabolism, ribosomes and fatty acid elongation) and the downregulation of transcription machinery and DNA repair/recombination proteins (involved in nucleocytoplasmic transport or spliceosome)

Protective immune trajectories in early viral containment of non-pneumonic SARS-CoV-2 infection

Project description:The antiviral immune response to SARS-CoV-2 infection can limit viral spread and prevent development of pneumonic COVID-19. However, the protective immunological response associated with successful viral containment in the upper airways remain unclear. Here, we combine a multiomics approach with longitudinal sampling to reveal temporally resolved protective immune signatures in non-pneumonic and ambulatory SARS-CoV-2 infected patients and associate specific immune trajectories with upper airway viral containment. We see a distinct systemic rather than local immune state associated with viral containment, characterized by interferon stimulated gene (ISG) upregulation across circulating immune cell subsets in non-pneumonic SARS-CoV2 infection. We report reduced cytotoxic potential of Natural Killer (NK) and T cells, and an immune-modulatory monocyte phenotype with protective immunity in COVID-19. Together, we show protective immune trajectories in SARS-CoV2 infection, which have important implications for patient prognosis and the development of immunomodulatory therapies.
