MRNA and lncRNA profiling of radiation-induced gastric injury in mouse model

ABSTRACT: In this study, we measured mRNA and lncRNA profiles of gastric tissues following 0, 6 and 12 Gy irradiation. The stomach of C57 mice were exposed to 0, 6 and 12 Gy X-ray irradiation at a dose rate of 2 Gy/min. The mice were sacrificed 7 days after irradiation and gastric tissues were collected. RNA was extracted and quantified. mRNA and lncRNA profilings of each groups were analyzed by microarray.

Project description:Purpose: The goal of this study was to compare the lncRNA and mRNA expression profile of human fibroblast WS1 cells between 0 Gy and 5 Gy X-ray irradiation. Methods: lncRNA and mRNA-sequencing of WS1 cells were generated using Illuminal Hiseq4000, RNA-seq reads were mapped to human genome hg19 by hisat2 software. Cuffdiff software was used to analysis the differentially expressed levels of LncRNA and mRNA FPKM. Results: We identified different expressed levels of 88780 lnRNA and 20311 mRNA transcripts in WS1 cells between 0 Gy and 5 Gy X-ay irradiation. Conclusion: Our study first revealed the detail of lncRNA and mRNA expression profiles of WS1 cells after 5Gy of X-Ray irradiation

Project description:This study aims to compare mRNA expression between radiated and non-radiated human keratinocyte HaCaT cells by microarray analysis. Human keratinocyte HaCaT cells were divided into two groups, each group has three repeats. The cells were irradiated with a single dose of 0 or 20Gy of X-ray irradiation. 48 hours post radiation, cells from 0 or 20 Gy groups were collected and subjected to microarray analysis. mRNA and lncRNA profilings of each group were analyzed by microarray.

Genome-wide analysis of the therapeutic effect of NeuroD on irradiation-induced intestinal injury

Project description:The objective was to investigate the therapeutic effect of NeuroD on irradiation-induced intestinal injury at gene expression level. Gene expression profilings between NeuroD and EGFP treated mice which were received 9 Gy total body irradiation were compared. All mice were given 9 Gy radiation and divided into two groups (3 mice per group). After radiation, mice were immediately administered with an intraperitoneal injection of 0.1 mM EGFP or NeuroD-EGFP. EGFP treated mice were served as controls. 12 hours post radiation, mice jejunum tissues were isolated. Total RNA obtained from jejunum tissues and subjected to gene expression profiling assays.

Next Generation Sequencing Facilitates Quantitative Analysis of 0 and 5 Gy HuR binding RNAs

Project description:Purpose: Next-generation sequencing (NGS) has revolutionized systems-based analysis of RNA. The goals of this study are to compare HuR binding RNAs between 0 or 5 Gy irradiation, and analyze its effect on RNA expression. Methods: Human skin fibroblast cell line WS1 cells were treated with 0 or 5 Gy irradiation. After 24 h, RNA expression was measured using using Illumina HiSeq4000. Meanwhile, RIP was performed using HuR antibody. HuR bound mRNAs after 0 or 5 Gy irradiation was measured using using Illumina HiSeq4000. Results: RIP-sequencing reveals HuR binding RNAs between 0 or 5 Gy irradiation in WS1 cells. RNA sequencing reveals dyeregulated RNAs between 0 and 5 Gy irradiation. Conclusions: Our study represents the first detailed analysis of HuR binding RNAs between 0 and 5 Gy irradiation generated by RNA-seq technology. This study contributes to our understanding of the role of HuR inresponse to radiation.

Characterization of protoporphytin IX effects on X-ray irradiation in Hela cells in vitro

Project description:Hela cells were used to evaluate the protoporphytin IX (PpIX) effects on X-ray irradiation. Treatment conditions were (1) control group (untreated cells); (2) cells treated with 1 Œºg /ml PpIX for 6 hours; (3) cells irradiated with 3Gy X-rays; (4) cells treated with 1 Œºg /ml PpIX for 6 hours prior to irradiation with 3Gy X-rays. Hela cells gene expressions in 4 groups were measured at 24 hours after exposure to 0 and 3 Gy X-rays plus treatment with PpIX prior to irradiation.
