Expression data of sheep exposed to Mycobacterium avium subsp. Paratuberculosis

ABSTRACT: In this study, consistent gene expression changes were analysed to identify gene-to-gene interaction and functional pathway relationships associated with the MAP exposure outcomes of either a paucibacillary or a multibacillary disease form or sheep resilient to disease. The results suggest that although groups of genes are differentially changed in the MAP exposed sheep regardless of clinical outcome, there are distinct variations in gene expression patterns between the paucibacillary and multibacillary forms of disease and of particular note was the finding that MAP exposed animals with evidence of recovery or resilience respond to MAP exposure with a pattern of gene and molecular pathway interactions distinct from those animals that progress to clinical paratuberculosis.

Project description:Sheep total RNA was extracted from the Ileo-caecal valve lymph node (ICLN) of sheep with multibacillary paratuberculosis compared to ICLN of uninfected controls. Sequencing libaries were prepared from RNA using the Illumina TruSeq RNA Sample Preparation Kit v2. Sequencing with 101 base paired end reads was performed on an Illumina HiSeq 2500 at Edinburgh Genomics.

Project description:Mycobacterium avium subsp. paratuberculosis (MAP) causes Johne’s disease that is a chronic wasting disease in ruminants. Spread of MAP is mainly provoked by a long subclinical stage during which MAP is shed into the environment with feces; accordingly, detection of subclinical animals is very important to its control. Therefore, the current diagnostic methods are not suitable for detection of subclinical animals. In this study, we conducted to develop a diagnostic method for analysis of the expression of genes of prognostic potential biomarker candidates in cattle naturally infected with MAP. We conducted miRNA-Seq to evaluate potential biomarker candidates. Experimental animals were divided into four groups based on fecal MAP PCR, serum ELISA, and clinical sign.

Identification of immunoreactive proteins of Mycobacterium avium subsp. Paratuberculosis

Project description:Mycobacterium avium subsp. paratuberculosis (MAP) is the cause of a chronic enteritis of ruminants (bovine paratuberculosis-Johne disease) that is associated with enormous worldwide economic losses for the zootechnical industries. Diagnosis is based on observation of clinical signs, on the detection of antibodies in milk or serum or on evaluation of bacterial culture from feces. The limit of these methods is that they are not able to detect the disease in the subclinical stage and are applicable only when the disease is already in an advanced status. For this reason the main purpose of this study is to use the MAP proteome to detect novel immunoreactive proteins that may be helpful for paratuberculosis diagnoses. 2D electrophoresis and 2D immunoblotting of MAP proteins were performed using sera of control cattle and paratuberculosis infected cattle in order to highlight the specific immunoreactive proteins. Among the assigned identifiers to immunoreactive spots it was found that most of them correspond to surface-located proteins while three of them have never been described before as antigens. The identification of these proteins improves scientific knowledge that could be useful for paratuberculosis diagnoses. The sequence of the identified protein can be used for the synthesis of immunoreactive peptides that could be screened for their immunoreaction against bovine sera infected with MAP.

Project description:Bovine paratuberculosis is a serious chronic disease of the gastrointestinal tract caused by Mycobacterium avium subspecies paratuberculosis (MAP). The aim of this study was to detect proteomic changes in peripheral blood mononuclear cells (PBMC) from cows of the same herd with different MAP infection status after co-incubation with viable MAP in vitro using label-free LC-MS/MS. Our data contribute to a better understanding of the bovine immune response and mechanisms of susceptibility to MAP.

Characterization of the inflammatory phenotype of Mycobacterium avium subspecies paratuberculosis

Project description:Understanding the pathogenic mechanisms and host responses to Johne's disease, caused by Mycboacterium avium subspecies paratuberculosis (MAP), is complicated by a number of challenges of the disease progression and lack of ex vivo models to study the disease. We describe novel cell culture passage model which mimics the course of infection in vivo. Using this model we determined that cell culture passaged MAP develops an inflammatory phenotype initiating higher levels of pro-inflammatory cytokines and chemokines compared to levels initiated by non-passaged MAP. To verify that the MAP phenotypes were changing, we conducted transcriptome analysis of each population during the passage model and identified a change in gene expression, specifically and upregulation of genes involved in the biosynthesis of lipid components within the cell. Lipidomic analysis showed dramatically different lipid profiles between the 2 populations. These transcriptome changes were validated by the identification of high levels of upregulated transcripts from MAP-infected cattle ileal tissue. In total, this study described a novel model used to show that MAP changes phenotype during intracellular passage, thus changing its lipid profile and triggering an inflammatory response in the host leading to the severe clinical phage of Johne's disease.
