Project description:Slow-growing Korat chicken (KR) is an alternative to broiler chickens that has been used as a national tool to support smallholder farmers due to a higher selling price of KR meat. However, the individual variability of feed efficiency (FE) within a KR stockbreeding results in a lack of competitiveness. Therefore, improvement of FE of KR is of major importance to improve the profitability of livestock production enterprises. Here, we selected two groups of KR with divergent feed conversion ratios (FCR). We performed RNA-sequencing in order to profile KR jejunal transcriptome and to identify the transcriptional variations and biological pathways implied in response to divergent FCR. The biological pathways involved were revealed by enrichment of the Gene Ontology (GO) terms, and the Kyoto Encyclopedia of Gene and Genome (KEGG) pathways. The results showed that main pathways involved in KR FCR divergence were related to immune response, glutathione metabolism, vitamin transport and metabolism, lipid metabolism, and maturation, development and growth. This is the first study to investigate the molecular genetic mechanisms affecting the FCR values in jejunum of slow-growing chicken. This study will be useful in the line-breeding programs for slow growing chickens to improve FE in the stockbreeding and its profitability.

Project description:PurposePhytochemicals of ethno medicines are being developed as effective drugs with minimum or no toxic side effects. Dhanwantaram Kashayam (DK) is a polyherbal formulation used as a potent general health tonic and is found to have antioxidant activities, but there are no proper scientific studies on its possible benefits. In this study we investigated the antioxidant and antidiabetic properties of DK and trying to explore the possibility of employing DK for the treatment of diseases like diabetes mellitus caused by the imbalance in Reactive Oxygen Species (ROS).MethodsWe investigated the effect of DK on normal and rat model of diabetes. Rats were fed with DK for 21 days. Fasting blood glucose level, haemoglobin, glycosylated hemoglobin, non enzymatic antioxidants like vitamin C, vitamin E, reduced glutathione, and MDA were evaluated.ResultsDK was found to ameliorate the disease symptoms of diabetes. A significant decrease in non enzymatic antioxidants - vitamin C, vitamin E, reduced glutathione and an increase in MDA and fasting blood glucose level was observed in diabetic rats. Administration of DK resulted significant increase in non enzymatic antioxidant levels both in diabetic and normal rats and a decrease in MDA, fasting blood glucose, glycosylated hemoglobin and HOMA-IR index was observed.ConclusionThis study reveals the protective role of DK in diabetic condition through its antioxidant and antihyperglycemic activity. Our results strongly indicate the possibility of DK being developed as a potent antidiabetic drug. DK could be an alternative in the treatment of diseases with ROS imbalance to expensive and toxic synthetic 'medicines'.

Project description:The functional differentiation of the granulosa cell (GC) layer plays an essential role in follicular development and oocyte maturation in avian. In the current study, single-cell RNA sequencing (scRNA-seq) was employed to identify GC types, uncover heterogeneity, and construct developmental trajectories of chicken GCs in two developmental stages of hierarchical follicles (HF-GCs) and prehierarchical (PHF-GCs). The findings enhance our understanding of the cellular biology of GC layer of ovarian follicles and will assist studies of the molecular mechanisms of follicle development and selection in chicken.

Project description:Improvement of feed efficiency would increase profitability of the poultry industries by decreasing the amount of feed required for production. Korat (KR) chicken is a new alternative meat-type chicken breed which its meat is recognized for its high protein, low fat and low purine content, whereas its low feed efficiency leads to high cost of production. Deeper understanding on how feed efficiency influences meat quality is poorly elucidated. To fulfill deeper understand molecular key that point the variation in feed efficiency and meat quality, the aim of this study was to investigate molecular pathways and genes involved in feed efficiency and meat quality in thigh of slow-growing KR chicken. A total of 75 males KR chicken were reared in individual cage until 10 weeks of age. Individual feed intake and body weight were collected weekly to calculate Feed Conversion Ratio (FCR) and Residual Feed Intake (RFI). Meat quality parameters were measured in thigh muscles such as ultimate pH (pHu), water-holding capacity (WHC), drip loss (DL), nucleotides content and several biomolecules (amide, …). Base on extreme values of FCR at 10 weeks of ages, 12 birds from the high FCR group (HFCR) and 9 birds from the low FCR group (LFCR) were selected for investigating their transcriptome using an 8×60K Agilent chicken microarray. In addition, a weighted gene coexpression network analysis was performed to detect the relationship between modules of co-expressed genes and feed efficiency, meat quality in thigh muscle. The result in this study indicated that selection on feed efficiency (FCR, RFI) would affect flavor precursor, lipid and protein content in thigh muscle. Based on WGCNA and functional enrichment analysis, results suggested that the key molecular pathways relate to FCR, RFI and meat quality (WHC, DL, IMP, AMP and inosine) in thigh muscle were the pathways of regulation of biological process, biological regulation and regulation of metabolic. Moreover, we revealed four genes there are assembly competence domain (ACD) gene, baculoviral IAP repeat containing 5 (BIRC5) gene, cytochrome c oxidase assembly factor 3 (COA3) gene and myosin light chain 9 (MYL9) gene that might be biomarker gene in feed efficiency and meat quality in thigh muscle. The hypothesis of the current study was alteration feed efficiency in slow-growing chicken will impact meat quality especially in term of texture and flavor.

Project description:Type 2 Diabetes (T2D) is a polygenic disease requiring a multipronged therapeutic approach. In the current scenario, the use of polyherbals is increasing among the diabetics. MD-1, a poly herbal formulation is constituted as a mixture of six popular anti diabetic herbs, used in the management of Diabetes mellitus (DM). The physicochemical, biochemical and in vitro efficacy studies have been carried out to ascertain the possible mechanisms underlying the anti-diabetic action of MD-1.MD-1 was evaluated for residual toxins as per Ayurvedic Pharmacoepia of India (API) procedures. The hydro alcoholic extract of the formulation (HAEF) was evaluated for anti oxidant activity against 2, 2-diphenyl-1-picrylhydrazil (DPPH) and nitric oxide radicals in vitro. The effect of HAEF on carbohydrate digestive enzymes ?-glucosidase and ?-amylase was studied using biochemical assays. HAEF was studied for its glucose lowering potential in L6 myotubes and 3T3L1 preadipocytes, using 2-deoxy-D-[1-3H] glucose (2-DG) uptake assay. Effect of MD-1 on adipogenesis was evaluated in 3T3L1 adipocytes using oil O red staining. The effect of HAEF on mRNA expression of peroxisome proliferator activated receptor gamma (PPAR?) and glucose transporter 4 (GLUT4) in 3T3L1 adiocytes was investigated by reverse transcriptase polymerase chain reaction (RT-PCR). Statistical analysis was performed by student t-test, ANOVA.Residual toxins present within the API limits and HAEF demonstrated strong antioxidant potential and significantly inhibited the ?-glucosidase (IC50 63.6?±?0.46 ?g/mL) and ?-amylase (IC50 242.81?±?1.26 ?g/mL) activity. HAEF significantly (p?<?0.05) enhanced the insulin stimulated glucose uptake in both the cell lines studied. Unlike standard pioglitazone (PGZ), HAEF modulated the mRNA expression of PPAR? and GLUT4 (p?<?0.0001) in 3T3L1 adipocytes, without inducing adipogenesis.Physicochemical parameters established in the study may serve as reference standards in regular quality control. Absence of residual toxins underpins the safety. The enhanced glucose uptake and favorable modulation of insulin sensitivity through a plausible weak PPAR? agonism is similar to the distinct PPAR? activation pattern of several reported natural compound agonists. The differential binding modes of such dynamic combinatorial ligands within the formulation unlike synthetic ligands like thiozolidinediones (TZD) can be linked to the safe mitigation of diabetic complications by MD-1.
