Circulating exosomal miRNA signature in pregnancies with gestational diabetes mellitus across gestation

Project description:Pancreatic β-cell function impairment is a key mechanism for developing gestational diabetes mellitus (GDM). Maternal and placental exosomes regulate maternal and placental responses during hyperglycemia. Studies have associated exosomal micro RNAs (miRNAs) with GDM development. To date, no studies have been reported that evaluate the profile of miRNAs present in maternal and placental exosomes in the early stages of gestation from pregnancies that develop GDM. We used microarrays to assess whether early pregnancy maternal and placental exosomal miRNA profiles vary according to pancreatic β-cell function in women who will develop GDM (preGDM).

Plasma exosomal microRNAs associated with metabolism as early predictor of gestational diabetes mellitus

Project description:The microRNA (miRNA) expression profile of plasma exosome in pregnant women complicated with gestational diabetes mellitus (GDM) has not been fully clarified. In this study, differentially expressed miRNAs in plasma exosomes were identified by high-throughput small RNA sequencing in 12 GDM and 12 normal glucose tolerance (NGT) pregnant women and validated in 102 GDM and 101 NGT pregnant women. A total of 22 exosomal miRNAs were found and five of them were verified by qRT-PCR. Exosomal miR-423-5p was upregulated, while miR-99a-5p, miR-192-5p, miR-148a-3p, and miR-122-5p were downregulated in pregnant women complicated with GDM.

Exosomal RNA expression profiles and their prediction performance in gestational diabetes mellitus patients with macrosomia

Project description:Exosomal RNAs in cord blood may allow intercellular communication between maternal and fetus. We aimed to establish exosomal RNA expression profiles in cord blood exosomes from gestational diabetes mellitus (GDM) patients with macrosomia.We used microarray technology to establish the differential mRNA, lncRNA and circRNA expression profiles in cord blood exosomes from GDM patients with macrosomia compared with normal controls. A total of 98 mRNAs, 372 lncRNAs and 452 circRNAs were differentially expressed in cord blood exosomes from GDM patients with macrosomia. Pathway analysis showed the differential genes were associated with PI3K-Akt signaling pathway, JAK-STAT signaling pathway, TGF-beta signaling pathway, insulin resistance, glycerolipid metabolism, fatty acid degradation and mTOR signaling pathway. These results showed that exosomal RNAs are aberrantly expressed in the cord blood of GDM patients with macrosomia.

Circulating circRNA signature in pregnancies with gestational diabetes mellitus

Project description:Circular RNA can regulate blood glucose levels by targeting mRNA expression, but the role of circRNA in GDM is still unknown. Therefore, a joint microarray analysis of circRNAs and their targeting mRNAs using the peripheral blood of GDM patients and healthy pregnant women was carried out for the first time. In our study, high-throughput microarray sequencing technique was used to analyze the expression profile of circRNA and transcripts mRNA in the peripheral blood of GDM patients, in order to comprehensively evaluate the role of circRNAs targets and their parents genes in the signal pathways related to the pathogenesis of GDM. Some of the discovered circRNAs and their linear transcript mRNAs related to T cell receptor signaling pathway were further verified in larger samples by droplet digital PCR(ddPCR) and quantitative real-time PCR(qRT-PCR), respectively. The verification results confirmed the initial microarray results.

Project description:The objective of this study was to investigate whether placental exosomes in gestational diabetes mellitus (GDM) carries a specific set of miRNAs associated with skeletal muscle insulin sensitivity. Exosomes were isolated from chorionic villi-conditioned media and plasma from normal and GDM pregnancies. A specific set of miRNAs was identified to be selectively enriched within exosomes when compared to their cells of origin indicating specific packaging of miRNAs into exosomes. In addition, miRNA expression varies in a consistent pattern in placenta, placental-derived exosomes, circulating exosomes and skeletal muscle in GDM.

Exosome proteomics reveals the deregulation of coagulation, complement and lipid metabolism proteins in gestational diabetes mellitus

Project description:Exosomes are the smallest extracellular vesicles which are released during pregnancy by the placenta, umbilical cord, amniotic fluid and various cell membranes in the extracellular space. The content of exosomes during pregnancy is strongly related to various conditions such as gestational diabetes mellitus (GDM). Although it is well-known that GDM is characterized by different levels of chronic low-grade inflammation, complement system dysregulation, vascular dysfunction and platelet activation, there is little data characterizing the serum exosomal protein cargo of GDM patients and their associations to these processes. The aim of this study was to analyze the serum exosomal proteome of GDM patients, with focus on the platelet activation and the complement proteins and their relationship to serum biochemical parameters and other protein markers of lipid metabolism and prothrombotic factors.

Gestational diabetes mellitus alters placental structure, efficiency, and plasticity

Project description:The hemochorial placenta provides a critical barrier at the maternal-fetal interface to modulate maternal immune tolerance and enable gas and nutrient exchange between mother and conceptus. Pregnancy outcomes are adversely affected by gestational diabetes mellitus (GDM); however, the effects of GDM on placental formation, and subsequently fetal development, are not fully understood. In this report, streptozotocin was used to induce hyperglycemia in pregnant rats for the purpose of investigating the impact of GDM on placental formation and fetal development. GDM caused placentomegaly and placenta malformation, decreasing placental efficiency and fetal size. Elevated glucose disrupted rat trophoblast stem (TS) cell differentiation in vitro. Evidence of altered trophoblast differentiation was also observed in vivo, as hyperglycemia affected the junctional zone transcriptome and interfered with intrauterine trophoblast invasion and uterine spiral artery remodeling. When exposed to hypoxia, rats with GDM showed decreased proliferation and ectoplacental cone development on gestation day (gd) 9.5 and complete pregnancy loss by gd 13.5. Furthermore, elevated glucose concentrations inhibited TS cell responses to hypoxia in vitro. Overall, these results indicate that alterations in placental development, efficiency, and plasticity could contribute to the suboptimal fetal outcomes in offspring from pregnancies complicated by GDM.
