Project description:Small non-coding RNAs (sRNAs) regulate gene expression of many biological processes. During growth, some myxobacteria produce an sRNA — Pxr — that blocks fruiting-body development, an aggregative multicellular process typically triggered by starvation. Deleting the pxr gene allows Myxococcus xanthus to develop despite nutrient availability, but Pxr binding targets and the genes regulated by Pxr remain unknown. Here, after showing that Pxr controls the temporal dynamics of development, we compare the transcriptomes of vegetative M. xanthus cells possessing vs lacking pxr. Over half of the genes impacted by pxr deletion are linked to development, including known and previously undiscovered critical regulators. Pxr also positively regulates genes associated with general metabolic processes. Our study discovers phenotypic effects of Pxr regulation with ecological importance, identifies the suite of genes this sRNA controls during vegetative growth and reveals a previously unknown developmental regulator. These findings provide insights into the molecular mechanism controlling myxobacterial development.

Project description:Tiller angle is a key factor determining rice plant architecture, planting density, light interception, photosynthetic efficiency, disease resistance, and grain yield. The distribution of auxin and shoot gravitropism play important roles in regulating tiller angles of rice. Several tiller angle-associated genes have been cloned. However, the mechanisms underlying tiller angle control are far from clear. In this study, we isolate bta1-1, a mutant with an enlarged tiller angle throughout its life cycle. A detailed analysis reveals that BTA1 has multiple functions because several major agronomic traits, including tiller and panicle number, biomass production, secondary branch number per panicle, panicle weight, grain size, and grain weight, are increased in bta1-1 plants. Moreover, BTA1 is a positive regulator of shoot gravitropism in rice. Shoot responses to gravistimulation are disrupted in bta1-1 under both light and dark conditions. Gene cloning reveals that bta1-1 is a novel mutant allele of LA1. LA1 is able to rescue the tiller angle and shoot gravitropism defects observed in bta1-1. BTA1/LA1 is required to regulate the expression of auxin transporters and signaling factors that control shoot gravitropism and tiller angle. High-throughput mRNA sequencing is performed to elucidate the molecular and cellular functions of BTA1/LA1. The results show that BTA1/LA1 may have multiple functions in regulating nucleosome and chromatin assembly, and protein and DNA interactions. Our results provide new insight into the mechanisms whereby BTA1/LA1 controls shoot gravitropism and tiller angle in rice.

Project description:Abstract: Multiple nodes and dwarf mutants in barley are a valuable resource for identifying genes that control shoot branching, vegetative growth, and development. In this study, physiological, microscopic and genetic analylsis were conducted to characterize and fine-map the underling gene of A a barley mutant with Multiple Stem Nodes and Spikes and Dwarf (msnsd) , which was selected from EMS- and 60Co-treated barley cv. Edamai 934 (E934) to characterize the mutant and fine-map its underlying gene. The msnsd mutant had more stem nodes and , lower plant height and a shorter plastochron than Edamai 934the wild type. Moreover, the mutant had two or more spikes on each tiller., emerging from the lower nodes or the base of the top spike, and a shorter plastochron. Microscopic analysis showed that the dwarf phenotype of msnsd resulted from reduced cell lengths and cell numbers in the stem. Further physiological analysis showed that msnsd was GA3-deficient, with its plant height increasing after external GA3 application. Genetic analysis revealed that a single recessive nuclear gene, namely, HvMSNSD, controlled the msnsd phenotype. Using a segregating population derived from Harrington and the msnsd mutant, HvMSNSD was fine-mapped on chromosome 5H in a 200 kb interval using bulked segregant analysis (BSA) coupled with RNA-sequencing (BSR-seq), with a C-T substitution in the exon of HvTCP25 co-segregating with the msnsd phenotype. RNA-seq analysis showed that a gene encoding gibberellin 2-oxidase 8, a negative regulator of GA biosynthesis, was upregulated in the msnsd mutant. Transcriptomic analysis identified several Several known genes related to inflorescence development that were also upregulated and enriched in the msnsd mutant. Collectively, we propose that HvMSNSD regulates the plastochron and morphology of reproductive organs, likely by coordinating GA homeostasis and changed expression of floral development related genes in barley. this This study offers valuable insights into the molecular regulation of barley plant architecture and inflorescence development

Project description:Analysis of different leaves from transgenic plants over expressing microRNA miR156. Time-dependent gene expression is essential for developmental timing control. MiR156 was characterized as a time-dependent expressed microRNA during leaf development. The level of miR156 is positive correlated to developmental time during leaf growth. We used microarray to identify genes regulated by miR156 in a time-dependent manner during leaf development. Results indicate that most time-negative genes were activated in mature leaves of miR156 over expressed plants compared with wild type. Keywords: gene expression array-based (RNA / in situ oligonucleotide) To identify the temporal regulation role of miR156 in different leaves, all leaves in the main tiller were collected separately from Md/Mh (miR156 over expressed lines) and wild type (WT) plants. The level of miR156 were checked in these samples, then three biological repeats were selected and these RNA samples of the oldest (L1) and youngest leaf (L4 or L5) were applied for microarray hybridization.
