Gene expression regulated by warm temperature in C. elegans

ABSTRACT: Stress resistance induced by mild environmental stress protects animals against aging-associated cellular damages and promotes longevity. Here, we reveal that mild heat triggers innate immune and detoxification responses that confers longevity at warm temperatures. We compared the transcriptome of 25°C-cultivated animals to that of 15°C controls through RNA-seq analysis. A total of 1958 transcripts were differentially regulated, with 1143 up-regulations and 815 down-regulations.

Project description:The model plant Arabidopsis thaliana responds to mild high temperature by increased elongation growth of organs to enhance cooling capacity, in a process called thermomorphogenesis. Our understanding of the genetic regulation of thermomorphogenesis has increased in recent years. However, hardly anything is known about molecular mechanisms outside A. thaliana and cellular signaling pathways have been underexplored. Therefore, we mapped changes in protein phosphorylation in A. thaliana and crops exposed to warm temperature. Based on these results, we identified and characterized a novel, functionally conserved signaling complex of MITOGEN-ACTIVATED PROTEIN KINASE KINASE KINASE KINASEs (MAP4KS) in warm temperature-mediated growth regulation in plants. This contributes to our understanding of warm temperature signaling, and can help guarantee food security under a changing climate.

Project description:MiRNA regulate the maintenance, differentiation and function of stem cells and progenitor cells. miRNA expression of progenitor cells located in the adventital layer of arterial vessels has not been characterized in either animal or human models. Further it is unknown if local arterial miRNA expression profiles change after injury of end organs supplied blood by these arterial conduits. CD34+/CD105- cells were extracted and analyzed for changes in miRNA expression after kidney specific ischemic injury. Cardiac-dead mouse lungs may remain transplantable at 37ºC for 4 hours if gas exchange is preserved

Project description:Despite a potentially huge number, uncontrolled donation after circulatory death contributed little to alleviating donor lung shortage due to rapidly progressive warm ischemia. Many methods have been studied in animals, but the tolerable warm ischemic time (WIT) remains less than 90 minutes. Using a refined mouse model of pulmonary artery ligation (PAL), we firstly determined the maximum tolerable WIT. 4-hour PAL caused mild lung infiltration without dysfunction upon reperfusion, whereas 5-hour PAL triggered arterial endothelium injury and more significant infiltration with dysfunction. Transcriptional profiling showed a myeloid-dominant inflammation with mild injury in 4-hour PAL. The maximum WIT was then adapted in a clinically relevant scenario. Donor mice died of circulatory arrest without heparization and remained at 37ºC for 4 hours, followed by isogenic transplantation. As observed in 4-hour PAL, nonhypoxic warm ischemia-reperfusion hardly affected graft function and histology, no matter if warm ischemic lung preserved gas exchange by spontaneously breathing or by postmortem protective ventilation. If the dead donors were left untouched, however, the grafted lungs suffered severe hypoxic warm ischemia-reperfusion injury, varying from partially aerated totally lost. Taken together, the retrieval time can be extended to 4 hours at 37ºC by preventing cardiac-dead donor lung hypoxia.

Facing warm temperatures during migration M-bM-^@M-^S cardiac mRNA responses of two adult sockeye salmon Oncorhynchus nerka populations to warming and swimming challenges

Project description:The main findings of the current study were that exposing adult sockeye salmon Oncorhynchus nerka to a warm temperature that they regularly encounter during their river migration induced an mRNA-level heat shock response that is exacerbated with swimming. Similar immune defense-related responses were also observed. Microarray analyses revealed that 347 genes were differentially expressed between the cold (12-13M-BM-0 C) and warm (18-19M-BM-0 C) treated fish (P < 0.01), with stress response (GO:0006950; P = 0.014) and response to fungus (GO:0009620; P = 0.003) elevated with warm treatment, while expression for genes involved in oxidative phosphorylation (GO:0006119; P = 0.0019) and electron transport chain (GO:0022900; P = 0.00043) increased in cold-treated fish. By studying single genes with RT-qPCR, warm treatment fish from the Chilko population of O. nerka induced expression of heat shock protein (hsp) 90M-NM-1, hsp90M-NM-2 and hsp30, as well as interferon-inducible protein (P < 0.05). A Nechako population of O. nerka with a narrower thermal tolerance window than the Chilko population showed even more pronounced responses to the warm treatment. In conclusion, it appears that during their once-in-the-lifetime migration these adult sockeye salmon encounter conditions that induce several cellular defense mechanisms. As river temperatures continue to increase, it remains to be seen whether or not these cellular defenses provide enough protection for all sockeye salmon populations. Comparative transcriptomic analyses of Spring OilSeed Rape (SOSR)accessions in response to warm temperature.

Project description:Elevated growth temperatures are negatively affecting crop productivity and increasing yield losses. Root traits associated with improved adaptation to rising temperatures are a promising approach to generate new varieties better suited to face the environmental constrains caused by climate change. In this study, we identified various Brassica napus roots traits altered in response to warm temperature. Thus, different combination of changes in specific root traits results in an extended and deeper root system. This overall root growth expansion facilitates root adaptation by maximizing root-soil surface interaction and increasing its ability to explore extended soil areas. We associated these traits to coordinated cellular events, including changes in cell division and elongation rates, that drive the increase in root growth triggered by warm temperature. Comparative genome wide transcriptomic analysis revealed the main genetic determinants of these RSA changes and uncovered the necessity of a tight regulation of the heat shock stress response to adjust root growth to warm temperature. Our work provides a phenotypic, cellular and genetic framework of root response to warming temperatures that will help to harness root adaptation mechanisms for crop yield improvement under the future climatic scenario.
