Allelic variation at a major QTL for cell wall related traits and coordinated changes in gene expression in maize

ABSTRACT: ngs2015_12_zeawall2-zeawall2-Address the hypothesis that allelic variation at a major QTL for cell wall related traits gives rise to coordinated changes in gene expression in maize-Two pairs of BC2S2 maize lines carrying F271 or F288 alleles in an 18-20 Mb interval of chromosome 6 in an otherwise F271 genetic background were grown in the field at Mauguio (Hérault, France) in 2015 with two row replicates. Irrigation was applied during the summer to prevent water stress. The bottom third of upstream-ear internodes (without nodes) of three representative plants in each of the two replicates were harvested two days before silking. All samples were immediately frozen in liquid nitrogen and stored at -80°C.

Gene expression has been considered as a way in identification of candidate gene underlying QTL. Five lines chosen for their allelic pattern at four main QTL were considered for expression studies, two lines with all four favorable alleles (RIL99, RIL54), two lines with the unfavorable allele at bin 6.06 QTL position (RIL39 , RIL71), and one line with the unfavorable allele at bin 9.02 QTL position (RIL118). Expression study will be investigated in ear internode at two stages emerging tassel and early silking stages with two replicates. Plants were cropped at INRA Lusignan (Vienne, France) in 2009 in a bloc design with two four-row replicates. Ltd (www.biozeron.com). Selecte three stem segments of plants REPEAT 1, 2 and 3 with good and similar growth to use: 2nd-3rd internodes (poplar stem top: PST1, PST2, PST3); 9th-10th internodes (poplar stem middle: PSM1, PSM2, PSM3); 14th-15th internodes (poplar stem bottom: PSB1, PSB2, PSB3). Project description:Transcriptional profiling of three mexican maize landraces under 10, and 17 days stress and recovery irrigation A dye balanced modified loop design was implemented. Two biological replicates (pooling five representative plants) representing each sampling point for each genotype were obtained for purified RNA from 120 randomly chosen seedlings. This experiment involved a total of forty-eight (24 sets) of microarray hybridizations, including direct and dye swap comparisons between treatments as well as across the three landraces. This design allowed us to determine differences in gene expression between the three different landraces under drought stress (10 and 17 days) and at recovery irrigation compared to irrigated controls.

Ethephon has been reported to shorten the internodes elongation and increase the harvestable grain yield of maize. In the present study, we characterized that internodes phenotypic responses to ethephon treatment induced should due to ethylene released inhibited the longitudinal growth of cells and promoted its the lateral growth. We used microarrays to detail the global programme of gene differentially expression underlying maize and identified distinct classes of up or down-regulated genes during this process.

Comparative Proteomic Analysis to Reveal Key Maize Low Nitrogen Responsive Genes

Project description:Two maize hybrid cultivars contrasting in low nitrogen tolerance (low nitrogen-tolerant XY335 and low nitrogen-sensitive HN178) were used in this study . The experiment was carried out at Xinji Experimental Station (43º31′N, 124º48′E) of Hebei Agricultural University. The top 0-20 cm of the soil used contained organic matter 17.79 g·kg-1, total nitrogen 1.21 g·kg-1, alkali hydrolyzed nitrogen 64.9 mg.kg-1, available phosphorus 23.8 mg·kg-1, and available potassium 120.6 mg·kg-1. The experiment adopted a split plot design, with varieties as the main plot and nitrogen fertilizer as the sub-plot. There were 2 varieties for testing: XY335 and HN138. Two levels of nitrogen supply: N0 (0 kg N ha-1) and N240 (240 kg N ha-1), replicated three times. Each plot had 6 rows, with the row length measuring 20 m, and the row spacing of 60 cm, giving the plot area of 72 m2. The planting density was 67,500 plants ha-1. Nitrogen fertilizer used was urea (46% N), and 50% was applied before sowing and at the flared stage, respectively. During the grain filling stage, leaf tissues of three biological replicates were collected from control and treatment conditions, and immediately frozen in liquid nitrogen for subsequent proteomics analysis.
