Project description:Floral organ identities are specified by few transcription factors which act as master regulators. Subsequently, specification of organ axes programs the distribution of distinct tissue types within the organs that themselves develop unique identities. The C-class, AGAMOUS-clade MADS box genes are primary promoters of the gynoecium which is divided into a distal style and a subtending ovary along the apical-basal axis. We show that members of a clade of B3-domain transcription factors, NGATHA1 to NGA4 (NGA1-4), are expressed distally in all lateral organs, and all four have a redundant and essential role in style development. Loss of all four genes results in gynoecia where style is replaced by valve-like projections and a reduction in style-specific SHATTERPROOF1 (SHP1) expression. In agreement, floral misexpression of NGA1 promotes ectopic style and SHP1 expression. STYLISH1, an auxin biosynthesis inducer, conditionally activated NGA genes, which in turn, promoted distal expression of other STY genes in a putative positive feed back loop. Inhibited auxin transport or lack of YABBY1 gene activities resulted in a basally expanded style domain and broader expression of NGA genes. We speculate that early gynoecium factors delimit NGA gene response to an auxin-based signal, elicited by STY gene activity, to restrict the activation of style program to a late and distal carpel domain.

RNA was extracted from inflorescences of 30 day old plants using Qiagen RNAEasy kit. cRNA was synthesized and hybridized to Affymetrix ATH1 array according to manufacture recommendation. Seven repeats of wild type, two repeats of nga quadruple mutants and two repeats of 35S:amiR-NGA164a were collected. Signal values were obtained and normalized using the GC-RMA protocol, using R 2.7.2 (www.r-project.org) and Bioconductor 2.2 (www.bioconductor.org/). Average correlation values between repeats was 0.985, while the average correlation between wild type and mutant samples was only 0.972. As the average correlation between samples the nga quadruple mutants and 35S:amiR-NGA164a plants was 0.984, and given the identical phenotype, we grouped the two genotypes together.

Five independent samples of floral bud tissues with bud scales removes were collected from the upper crown of a sexually mature male tree and female tree. RNA was extracted from tissues and hybridized on Affymetrix Genechip Poplar Genome Array.

Project description:The carpel number (CN) is an important fruit trait affecting fruit shape, size, and internal quality in cucumber. CsCLAVATA3 (CsCLV3) was previously showed to be the simply inherited gene responsible for carpel number variation in cucumber, but the molecular mechanism of CsCLV3 regulating carpel number remains elusive. Here, we found that the expression of CsCLV3 was negatively correlated with carpel number variation in different cucumber lines. Knock down of CsCLV3 by RNAi led to increased number of petals and carpels, suggesting that CsCLV3 functions as a negative regulator for floral organ number in cucumber. WUSCHEL (WUS) has been well characterized to promote CLV3-expressing stem cell activity in a non-cell autonomous manner to regulate meristem maintenance and floral organ number. However, here we found the expression region of CsCLV3 overlaps with CsWUS in the basal domain of meristem, and CsCLV3 interact with CsWUS at the protein level through binding to the WUS-box motif. Overexpression of CsFUL1, a FRUITFULL-like MADS-box gene involved in fruit length regulation, resulted in increased number of floral organs in cucumber. Biochemical analyses indicated that CsFUL1 can directly bind to CsWUS promoter to stimulate its expression. Further, we found that auxin participates in carpel number variation in cucumber through physical interaction of AUXIN RESPONSE FACTOR 14 (CsARF14) and CsWUS. Therefore, CsFUL1 and CsARF14 are two new players in the WUS-CLV pathway in determining carpel number variation in cucumber.

Project description:Floral organs are extremely sensitive to stress during anthesis and lead to severe yield loss. Rice anthers and pollinated pistils of two cultivars with contrasting tolerance to heat and drought stress under variable conditions, including control, heat, combined heat and drought stress, were used to explore gene expression pattern in male and female reproductive organs during anthesis under control and stress conditions. More gene regulation was induced by combined drought and heat stress than heat in anthers of both cultivars. N22 showed less regulation under combined stress than Moroberekan. The overlap of regulated genes between two cultivars was rather low, indicated the distinct molecular stress responses. We used whole genome microarrays to explore gene expression pattern and molecular mechanisms in male and female reproductive organs during anthesis under control and stress conditions in two rice cultivars, sought to identify the key transcripts that play roles in inducing heat and drought tolerance during reproduction in rice.
