ABSTRACT: A customized targeted oligoarray was used to monitor the expression levels of 1000 genes, representative of the immature kernel transcriptome. Using this oligoarray we compared transcripts from 10 DAP kernels of two susceptible and two drought tolerant genotypes. Peroxidase B6T173 (ZmPrx35) from insect-resistant maize (Zea mays, p84C3) kernels

Project description:Peroxidase (POD) activity was found to be positively correlated with insect resistance in maize kernels. Thus, we purified active PODs from (Zea mays, p84C3) kernels and subjected to LC-MS/MS analyses. To verify the identified putative plant PODs, we produced them by recombinant expression in E. coli. Whereas B4FFK9 did not display in vitro activity, B6T173 (ZmPrx35) was active and also exhibits a heme cofactor. B6T173 accounts for about 80% of the POD activity in the kernel, according to densitometric evaluation.

Transcriptional profiles uncover Aspergillus flavus induced resistance in mature maize kernels

Project description:Aflatoxin contamination caused by the opportunistic pathogen A. flavus is a major concern in maize production prior to harvest and during storage. Previous studies indicate that both constitutive and induced resistance are involved in maize kernel defense against A. flavus infection, little is known about molecular mechanisms of mature kernels in response to fungal infection. The purpose of this study is to determine gene expression differences in maize kernels between resistant and susceptible lines in response to A. flavus challenge. To avoid the environmental effects in the field inoculation, a laboratory based inoculation technique Kernel Screening Assay (KSA) was used to challenge kernels with A. flavus. After 72 hours incubation of inoculated and noninculated mature kernels, gene expression profiles of two Near Isogenic Lines (NIL) of Eyl25 (A. flavus resistant) and Eyl31 (A. flavus susceptible) were compared using oligonucleotide array.

Transcriptional changes in Aspergillus flavus-infected maize kernels

Project description:Maize kernels are susceptible to infection by the opportunistic pathogen Aspergillus flavus. Infection results in reduction of grain quality and contamination of kernels with the highly carcinogenic mycotoxin, aflatoxin. To understand host response to infection by the fungus, transcription of approximately 9,000 maize genes were monitored during the host-pathogen interaction with a custom-designed Affymetrix GeneChip® DNA array. More than 1,000 maize genes were found differentially expressed at a fold change of 2 or greater. This included the up regulation of defense-related genes and signaling pathways. Transcriptional changes also were observed in primary metabolism genes. Starch biosynthetic genes were down regulated during infection, while genes encoding maize hydrolytic enzymes, presumably involved in the degradation of host reserves, were up regulated. These data indicate that infection of the maize kernel A. flavus induced metabolic changes in the kernel, including the production of a defense response, as well as a disruption in kernel development.

Maize kernels were mock inoculated at the blister (R2) or dough (R4) stage or inoculated with A. flavus at the blister (R2), milk (R3), dough (R4), or dent (R5) stage, and harvested 4 days later. Each treatment consisted of three biological replications. For each biological replication, 8 kernels were ground and RNA was isolated and further processed.

Totally 555 significantly differentially expressed genes (DEGs) were identified according to these RNA-Seq data (q-value <0.05, log2(fold change) >= 1.5), with 317 down- and 238 up-regulated genes.

Metabolic pathways operating in deteriorating versus germination-competent cyanide-primed walnut kernels

Project description:Incubation of physiologically dormant walnut kernels at warm and moist conditions leads to reduced germination and lower viability associated with triacylglycerol depletion, lipase activation and fatty acid accumulation. Cyanide however, releases kernel dormancy and improves germination accompanied with enhanced gluconeogenesis. The hypothesis that the viability loss in aging kernels is mainly due to lipid catabolism-induced oxidative stress and higher respiration was tested in a proteomic study. We used kernels with 15% moisture content aged by controlled deterioration, physiologically dormant non-aged and cyanide-treated germination primed; having low, medium and high germination potentials, respectively. We revealed 155 differentially abundant proteins out of 930 identified ones compared to dry (6% moisture content) kernels. Proteins accumulated in deteriorated kernels mainly belonged to protein folding, translation and degradation, defense, stress response and detoxification, glycolysis and fermentation but less abundant ones were related to gluconeogenesis and amino acid metabolism. Contrastingly, accumulated proteins belonged to gluconeogenesis and oxidative pentose phosphate pathway in germination-primed kernels. Deteriorated kernels also showed increased levels of H2O2, lipid peroxidation, free fatty acids, protein carbonylation, and amino acids, particularly proline but, a lower amount of sucrose and enhanced lipase, catalase, peroxidase and glutamate dehydrogenase activities. Of note, we revealed accumulation of various chaperons and proteins related to detoxification in deteriorated kernels, unlike reports on other aging seeds. Suppressed amino acid metabolism and gluconeogenesis enhanced respiration and oxidative stress in deteriorated kernels, while optimized carbon/energy metabolism promoted germination of primed kernels.

Developmental changes in transcriptomes of two key transport tissues in maize kernels: The Maternal Transfer Zone (MATZ) and Basal Endosperm Transfer Layer (BETL)

Project description:Transcriptome profiles of MATZ and BETL tissues are compared across three stages of development. Sugars and other nutrients unloaded from vascular tissues in the MATZ are imported by the BETL for utilization by the developing endosperm. Pronounced changes in gene expression occur in both tissues during kernel development. RNAseq data were obtained from duplicate tissue samples isolated by cryomicrodissection of developing maize kernels at three developmental time points; 8 days post-pollination (DAP), 14 DAP and 20 DAP.
