Characterization of the Endometrial Transcriptome in Early Diestrus Influencing Pregnancy Establishment in Dairy Cattle after Transfer of In-Vitro Produced Embryos

ABSTRACT: In this study, we sequenced the RNA content of endometrial biopsies from Holstein cows at day 6-8 to identify candidate genes and describe biological mechanisms for endometrium receptivity and pregnancy establishment upon transfer of IVP embryos.

Transcriptomics of the HPG axis from in vitro or in vivo-produced dairy male calves

Project description:In cattle, two of the most common technologies for embryo production are “in vivo” (MOET) or “in vitro” (IVP). IVP can sometimes impact fetal growth, resulting in large offspring, although most IVP calves are born apparently healthy. However, gene regulation changes in the organs involved in the developmental process in phenotypically normal postnatal calves are unknown and could indicate physiological programming. The objective of this study was to compare the transcriptomic modifications in organs of the hypothalamus-pituitary-gonadal and adrenal (HPGA) axes between IVP and MOET male calves of three months of age. RNA extracted from the HPGA organs samples (n=4 per group) were sequenced through RNAseq. The raw files were subjected to a bioinformatic pipeline to determine differentially expressed genes (FDR<0.05) in IVP versus MOET samples and the KEGG pathways enriched by these genes (FDR < 0.1). Pathways related to the reproductive process (GnRH secretion in hypothalamus, GnRH signaling in pituitary and steroidogenesis in testicle) were stimulated in IVP calves, suggesting an early activation of the HPG axis in male IVP calves when compared to MOET counterparts.

The objective was to compare the epigenomic and transcriptomic modifications in the liver, muscle, and organs of the hypothalamus-pituitary-gonadal and adrenal (HPGA) axes between IVP and MOET male calves of three months of age. Pregnancies were monitored until delivery. Male calves were maintained in similar conditions during the postnatal period until euthanasia at around 100 days of age. There were no differences regarding body weight at birth and growth rate among the animals in both groups at this age.

Transcriptomics of the liver and the muscle from in vitro or in vivo-produced dairy male calves

Project description:In cattle, the in vitro production (IVP) of embryos is becoming more relevant than embryos produced in vivo, i.e., after ovarian stimulation and embryo transfer (MOET). However, the effects of IVP on the developmental programming of specific organs in the postnatal calves are yet unknown. The objective of this study was to compare the hepatic and muscular transcriptomic modifications between IVP and MOET male calves of three months of age (n=4 per group). Tissue samples from liver and semitendinosus muscle were obtained at 3 months of age, and the extracted RNA was sequenced through RNAseq to determine differentially expressed genes (DEG; FDR<0.05). KEGG pathways enrichment analysis showed that DEG up-regulated in the liver and the muscle of the IVP calves were involved in oxidative phosphorylation and the tricarboxylic acid cycle. In contrast, ribosome and translation were up-regulated in the liver but down-regulated in the muscle of the IVP calves compared to the MOET calves (FDR<0.05). In conclusion, our findings indicated an altered hepatic and muscular energy regulation in phenotypically normal IVP calves compared to MOET calves.

Association between progesterone concentration in early diestrus and endometrial gene expression in grazing dairy cows

Project description:The objective of this study was to evaluate the association between progesterone concentration (P4) at day 4 of the estrous cycle (beginning of diestrus) and endometrial global gene expression at day 9 (middle diestrus) in lactating grazing dairy cows. Blood samples were obtained on days 0, 4 and 9 for P4 measurement by chemiluminescence. Cows were asignated to one of the following groups (n=3/group): cows with low physiological P4 at day 4 (PLP4), cows in anestrous (ANE), cows with high physiological P4 at day 4 (HPP4) and superovulated cows (SO). Endometrial biopsy samples were obtained on day 9 for RNA sequencing. Quality control and determination of differentially expressed genes (DEG; FDR<0.05) were determined using the edgeR package for R. The number of DEG between groups were: HPP4 vs LPP4: 453; HPP4 vs ANE: 623; SO vs LPP4: 603; SO vs ANE: 1102; LPP4 vs ANE: 261 and SO vs HPP4: 0. Functional analysis using the DAVID database revealed that endometrial genes upregulated by low P4 are enriched in the immune system and inflammatory response. Conversely, genes upregulated by high P4 are enriched inmodulation of uterine relaxation-contraction, GnRH signaling pathway, focal adhesion and EGF-like related terms. In conclusion, our results support the concept of changes in P4 at the beginning of the luteal phase could be associated with the endometrial expression of critical genes involved in the preparation of the uterine environment for embryo implantation.

Whole genome bisulfite sequencing of bovine morula derived from metabolically divergent dairy cows

Project description:Lactating cows experience transient metabolic stresses during postpartum which results in abnormal concentrations of non-esterified fatty acids and beta-hydroxybutyrate that can affect embryo-maternal communication and ultimately reproductive success. We hypothesize that metabolic challenges in lactating cows influence DNA methylation changes of the embryos prior to implantation affecting genes involved in embryo developmental competency. Therefore, we compared whole genome bisulfite sequencing (wgbs) of morulas derived from in vitro produced 2-4 cell embryos and transferred in the oviduct of metabolically profiled lactating cows until day 7. Similar stage of in vitro production (IVP) embryos were transferred to the oviduct of nulliparous heifers as a metabolically unchallenged control. Bisulfite-Seq DNA libraries were generated from groups of five morulas using an EZ DNA methylation-direct kit and Pico-Methyl seq library preparation kit (Zymo Research), and parallel sequenced using the illumine HiSeq2500 system at the institute of Genome Biology, Leibniz-Institute for Farm Animal Biology (FBN), Dummerstorf, Germany.

Yet, the miRNA gene regulatory network for the synthesis of milk protein in mammary is poorly understood. In this study, the hypothesis was that miRNAs have potential roles in bovine milk protein production. Using miRNA-seq and RNA-seq, we investigated the miRNAs profiles of mammary glands from 12 Chinese Holstein cows with six cows at peak of lactation and six in non-lactating period, from which three cows were in high and three in low milk protein percentage.
