Comparative analysis of gene expression of SLC3A2 monoallelic knockout and wild type clones

ABSTRACT: By employing a global gene expression profiling, we performed analysis of the molecular pathways which are deregulated in SLC3A2 monoallelic knockout clone generated from the head-and-neck cancer cell line Cal33 via CRISPR/Cas9 approach. Both non-irradiated and irradiated with 4Gy samples were compared.

Cancer SLC3A2-mediated lysine uptake restricts T cell immunity against hepatocellular carcinoma

Project description:Our research demonstrates that in hepatocellular carcinoma (HCC), tumor cells outcompete T cells for lysine (Lys) by overexpressing the SLC3A2 transporter, thereby reducing Lys availability within the tumor microenvironment. This lysine deprivation results in decreased STAT3 levels in T cells, inhibiting their proliferation and effector functions, which ultimately accelerates tumor progression. Additionally, we identified a c-Myc-SLC3A2 regulatory axis activated by Lenvatinib, enhancing the sensitivity of HCC to the combined treatment with Lenvatinib and Lys.

Random monoallelic expression in the mouse: SNP array data

Project description:Random monoallelic expression is defined by the allele-specific expression of genes, and by the fact that for an individual cell this monoallelic expression is neither obligate nor necessarily coordinated with the allelic expression in other cells. In order to find novel examples of random monoallelic expression in mouse, we did a transcriptome-wide survey of allele-specific gene expression in two different immortalized cell types. Lymphoblast cell lines and fibroblast cell lines were established (both clonal and nonclonal) and were used as a source of both nuclear RNA and genomic DNA. These samples were assessed for allele-specific gene expression using a custom-designed Mouse SNP Chip. A large number of genes (over 10% of those that were assessed in lymphoblast clones) displayed random monoallelic expression. For each cell line, two replicate samples of ds-cDNA were assessed for monoallelic expression, while genomic DNA was assessed as a control for possible LOH events. Nonclonal samples were used as controls for cis-acting allelic bias.

Liquid-liquid phase separation of ZHX2 protects DLBCL cells against ferroptosis through induction of SLC3A2

Project description:Diffuse large B-cell lymphoma (DLBCL), the most common B-cell non-Hodgkin lymphoma (B-NHL), is characterized by strong aggression, high heterogeneity and poor prognosis. Consequently, there is an urgent need to identify crucial therapeutic targets. Here, we found that the transcription factor Zinc-finger and homeoboxes 2 (ZHX2) is highly expressed in DLBCL. Subsequently, ZHX2 was proven to be critical for promoting DLBCL cells proliferation by inhibiting ferroptosis. Mechanistically, ZHX2 binds to the promoter region of the solute carrier family 3-member 2 (SLC3A2) gene through liquid-liquid phase separation (LLPS) and activates its function to negatively regulate ferroptosis. Furthermore, we constructed lipid nanoparticles Zhx2-siRNA@LNP targeting DLBCL, which effectively inhibited the growth of the tumor in vivo. In summary, our study indicated that LLPS of ZHX2 protects DLBCL against ferroptosis through induction of SLC3A2, and disturbing it with Zhx2-siRNA@LNP could significantly repress DLBCL, providing a promising therapeutic strategy for DLBCL.

Mass spectrometry imaging (MSI) combined with LC-MS/MS data from head and neck squamous cell carcinoma (HNSCC) xenograft samples to map peptide information

Project description:Mass spectrometry imaging (MSI) was used to investigate different tumors regions from five head and neck squamous cell carcinoma (HNSCC) xenograft samples of tumor model CAL33. Specifically, the authors investigated if measurements could be attributed to tumor hypoxia. Hypoxic regions were defined using consecutive tissue slices which were stained with anti-pimonidazole polyclonal antibody. M/z values which were associated to hypoxic regions were converted to masses and mapped with masses from LC-MS/MS experiment.
