Comparative analysis of gene expression in 9 BXH RI strains and progenitors (B6 and C3) and BXHF1.

ABSTRACT: We hypothesize that gene expression in the lungs of this RI panel are divergent thus contributing to the disparity in their phenotypes. More specifically, (1) B6 mice tend to have a much smaller lung volume than the C3 mice while the F1 appears to be like the B6 in lung structure and function. We postulate that their gene expression differences would reflect this. (2) The diversity in the phenotypes of this RI panel could be explained by a few genes whose expression spectrum across this panel could explain the physiological spectrum. In order to test these hypotheses at the gene expression level, we utilized microarray analysis to examine transcriptional differences among lungs of these strains of mice. Project description:Ribonuclease Inhibitor (RI also known as Rnh1) is a 50 kDa, ubiquitously expressed leucine-rich repeat (LRR) protein. It is localized in cytosol and binds to pancreatic-type ribonucleases and inhibit their function. However the entire biological role for Rnh1 is unknown. We generated Rnh1 knock out mice by homologous recombination. Here we studied differential gene expression from wild type (Rnh1 +/+), Heterozygous (Rnh1+/-) and Knock out (Rnh1-/-) yolk sacs isolated from embryonic day 9.5 (E9.5). We used microarrays to study global gene expression regulated by Rnh1 in yolk sacs.

Project description:To identify differences in the gene regulation between BCL6+/+ and BCL6-/- CML cells a gene expression analysis has been performed. We investigated the gene expression pattern in BCL6+/+ cells in the presence or absence of Imatinib and a combination of Imatinib and RI-BPI (a novel retro-inverso BCL6 peptide inhibitor). In BCL6-/- CML cells, we investigated the gene expression pattern in the presence or absence of Imatinib. BCR-ABL1 transformed myeloid cells from BCL6+/+ mice were cultured in the presence or absence of 10µM Imatinib or 10µM Imatinib and 20µM RI-BPI for 16 hours. BCR-ABL1 transformed myeloid cells from BCL6-/- mice were cultured in the presence or absence of 10µM Imatinib. Two samples for each condition were processed.
