The results showed that there were different expression of miRNAs in WT and Cbl-b-/- CD4+ T cells when activated or not.

mRNA expression of Qa2+ and Qa2- CD4+ thymocytes isolated from WT and miR-142-/- mice

Project description:Developing CD4+ T-cells in the thymus mature from CD69+Qa2- cells to CD69-Qa2+ T-cells. This maturation is accombanied by increased expression of CD62L and S1PR1 in Qa2+ cells, which allows mature T-cells to emigrate from the thymus to the periphery. However, miR-142-/- mice are characterized by an altered thymocytes homeostasis, characterized by reduced CD62L protein levels on their cell surface. To characterize the underlying molecular mechanisms, which lead to reduced CD62L expression in miR-142-/- CD4+ thymocytes, we investigated the mRNA levels in immature Qa2- as well as mature Qa2+ thymocytes isolated ex vivo from WT and miR-142 deficient Bl6 mice. Thymi were isolated from a pool of 4-5 mice per genotype and cells were isolated by FACS sorting. After sorting, total mRNA was isolated and expression levels were investigated with an Affymetrix microarrage Gene 1.0ST chip.

Silencing miR-31 in naive CD4+ T cells by antagomiRNA

Project description:To describe the global profile of genes regulated by miR-31, we compared the transcriptomics between miR-31 silenced and antagomir control treated naive CD4+ T cells. Interestingly, the cells could be separated into two subsets according FCS and SSC by flow cytometer after the silence of miR-31, one (~65%) was small sized cells with low granularity, the other (~30%) was large cells with high granularity, whereas the antagoNC treated naïve CD4+ T cells only contain a marginal fraction of large cells. We pooled “antagoNC”, “antagomiR31S” and “antagomiR31L” naïve CD4+ T cells from 5 healthy donors, respectively, and analyzed the differential genes among those three subsets.

Loss of hepatic miR-33 improves metabolic homeostasis and liver function without altering body weight or atherosclerosis

Project description:miR-33 is an intronic microRNA within the gene encoding the SREBP2 transcription factor. Like its host gene, miR-33 has been shown to be an important regulator of lipid metabolism. Inhibition of miR-33 has been shown to promote cholesterol efflux in macrophages by targeting the cholesterol transporter ABCA1, thus reducing atherosclerotic plaque burden. Inhibition of miR-33 has also been shown to improve high-density lipoprotein (HDL) biogenesis in the liver and increase circulating HDL-C levels in both rodents and nonhuman primates. However, evaluating the extent to which these changes in HDL metabolism contribute to atherogenesis has been hindered by the obesity and metabolic dysfunction observed in whole-body miR-33–knockout mice. To determine the impact of hepatic miR-33 deficiency on obesity, metabolic function, and atherosclerosis, we have generated a conditional knockout mouse model that lacks miR-33 only in the liver. Characterization of this model demonstrates that loss of miR-33 in the liver does not lead to increased body weight or adiposity. Hepatic miR-33 deficiency actually improves regulation of glucose homeostasis and impedes the development of fibrosis and inflammation. We further demonstrate that hepatic miR-33 deficiency increases circulating HDL-C levels and reverse cholesterol transport capacity in mice fed a chow diet, but these changes are not sufficient to reduce atherosclerotic plaque size under hyperlipidemic conditions. By elucidating the role of miR-33 in the liver and the impact of hepatic miR-33 deficiency on obesity and atherosclerosis, this work will help inform ongoing efforts to develop novel targeted therapies against cardiometabolic diseases.
