Project description:MCF-7 TET Off cells (MCF-7 wt) were used to produce stable clones expressing ER-beta tagged with TAP-tag at the C-term (C-TAP-ER-beta). All cells were grown in Dulbecco's modified Eagle's medium (DMEM), starved by using DMEM w/o phenol red and 5% Dextran Coated Charcoal stripped serum (DCC-FBS) for 5 days. Cells were transfected wiyh hsa-miR-30a-5p Mimics and with tha negative control Scramble both at concentration of 5 and 10 nM. Biological replicate were lysed and RNA extracted were pooled 24 hrs after transfectiions. For mRNA expression profiling, 500 ng total RNA were reverse transcribed and used for synthesis of cDNA and biotinylated cRNA. Finally cRNA were hybridized for 19 hours on Illumina HumanHT-12 v4.0 BeadChips and after scanning, data analysis was performed.

Project description:Exposures to environmental endocrine disruptors is growing and human contact in industrialized countries has become signficant and constant. We studied the effect of chronic exposure to two endocrine disrupting compound, bisphenol A and genistein, in an in vitro cell culture system. MCF7 cells were cultured for greater than 70 passages under normal (MCF7-F) conditions or with the addition of 50 nM BPA (MCF7-B) or GEN (MCF7-G). We performed transcriptome analysis of the all three cell lines in the absence of any estrogenic compounds and in the presence of 10 nM estradiol for 3 hours. The MCF7-F, B, G cell lines were starved of all estrogenic compounds by culturing the cells for 72 hours in phenol red free DMEM containing 5% charcoal/dextran treated FBS. Cells were then treated with either ethanol vehicle or 10 nM 17b-estradiol for 3 hours. Total RNA was harvested and utilized for whole genome analysis on the Affymetrix Human Trascriptome Array 2.0. Project description:This study provides a comprehensive evaluation of changes in gene expression during treatment with Genistein in vitro. Ishikawa cells were maintained and grown (to confluency or at a desired cell density between 500,000 - 1 million cells/mL) in DMEM/F-12 Medium (supplemented with 10% Fetal Bovine Serum + 1X penicillin/streptomycin) in xenoestrogen-free Corning plasticware. Cells were gently washed in warm PBS and transferred to phenol red-free DMEM/F-12 Medium (supplemented with 10% Charcoal-Stripped Fetal Bovine Serum + 1X penicillin/streptomycin) in Corning Cell Culture Cluster wells overnight and then challenged with 10pM (very low, vL), 1 nM (low, L), 10nM (high, H), and 1uM (very high, vH) levels of Genistein. Prior to collection, cells were washed in warm PBS, resuspended & briefly incubated in TRI-Reagent, & finally collected (in quintuplicate replicates) at time each specific time point: 8 hours, 24 hours, and 48 hours. Following RNA isolation, the best RNA yields for each quadruplicate set was selected for target preparation and GeneChip processing.
