Rca-T78 vs, Rca-S78

ABSTRACT: Transcription profiiling of arabidopsis transgenic Rca-T78S comparing Control WT plant (Rca-T78). Rca is Rubisco activase and Threonine78 residue of Rca is phosphorylated only in the dark. However, Serine substituted Rca (T78S) was phosphorylated even in the light. Thus, Rca-T78 or Rca-T78S expressing plants which were exposed light for an hour were harvested to check gene expression pattern

mRNA Expression Data from Murine Hearts of Shiftwork MI, Normal MI and Sham Wildtype Mice at 24-hours Post-MI

Project description:Male C57Bl/6 mice were randomized to undergo 5 days of i) a shiftwork protocol (10-hour light: 10-hour dark cycle) before myocardial infarction (MI) surgery, ii) a normal 12-hour light: 12-hour dark environment before MI surgery, iii) a normal 12-hour light: 12-hour dark environment and used as sham controls, or iv) a shiftwork protocol (10-hour light: 10-hour dark cycle) and used as sham controls. MI surgery was performed on the 5th day, after which all mice were returned to a normal 12-hour light: 12-hour dark cycle. Hearts were collected 24-hours post-MI at ZT06. The microarray approach allows the investigation of transcriptome-wide gene expression changes in hearts from mice on a shiftwork cycle or on a regular light:dark cycle before MI.

Single cell transcriptomic studies of mouse colonic lamina propria cells from wildtype or RORgtS182A mutant mice

Project description:The evolutionarily conserved serine 182 residue on RORγt is phosphorylated. Here, we report that this residue is dispensable for thymic T cell development, but essential for maintaining proper balance of Th17-Treg populations in the colon. Single-cell RNA-seq (scRNA-seq) revealed that serine 182 residue on RORγt is critical for restricting IL-1b-mediated Th17 activities and promoting anti-inflammation cytokine IL-10 production in Lymphoid tissue (LT)-like RORγt+Foxp3+ regulatory T cells in inflamed tissues.

A database of circadian and diel rhythmic gene expression in the yellow fever mosquito Aedes aegypti

Project description:The mosquito Ae. aegypti is responsible for the transmission of many diseases including yellow fever and Dengue fever. This species exhibits many behaviors that are under diel and circadian control. However, there has been little reporting on gene expression rhythmicity. To study how gene expression is globally regulated by diel and circadian mechanisms, we have undertaken a DNA microarray analysis of Ae. aegypti head and bodies under 12:12 light:dark cycle (LD) and constant dark (DD, free-running) conditions. Zeitgeber Time (ZT) with ZT12 defined as the initiation of the one hour dusk period under the light:dark cycle, and ZT0 defined as beginning of the one hour dawn period. Circadian Time (CT) with CT0 defined as subjective dawn, inferred from ZT0 of the previous light:dark cycle.

Project description:The serine 31 (S31) residue of histone H3.3 is phosphorylated during mitosis and this modification is enriched at repetitive DNA. ChIP-seq of H3.3 S31Ph was carried out in a mitotic population of synchronised mouse ES cells. We find that H3.3 S31Ph is enriched at telomeres and IAP repeats in mitotic mouse ES cells.
