Project description:RNA-seq libraries were generated on thymic epithelial cell (TEC) subsets from thymic samples (11 days to 3 months of age). Cells were sorted to isolate cortical TEC (cTEC), MHC low medullary TEC (mTEClo) and MHC high medullary TEC (mTEChi). Between 7,575 and 50,000 cells were isolated for each sample. TEC were isolated using CD45 MACS depletion followed by the sorting protocol described in Stoeckler et al. J Vis Exp 2013 (PMID 24084687; doi: 10.3791/50951). The study has been granted ethical approval and is publicly listed (IRAS ID 156910, CPMS 19587).

Project description:Accumulation of visceral (VIS) as opposed to subcutaneous (SUB) fat is a predictor of metabolic disorders, insulin resistance, and cardiovascular disease. This is due in part to the limited capacity of VIS fat to buffer lipids allowing them to deposit in insulin-sensitive tissues, thus impairing insulin action. In addition, VIS adipose tissue is more prone to fibrosis and inflammation, which in turn contributes to the pathogenesis of metabolic disease. Mechanisms underlying selective hypertrophic growth and tissue remodeling properties of VIS fat are not well understood. We and others identified subsets of adipose progenitors (APs) unique to VIS fat with differential adipogenic capacity. We named these CD34+ APs VIS low and VIS high based on low or high CD34 expression respectively. Despite sharing the same developmental origin, VIS low APs are adipogenic whereas VIS high APs are not. Furthermore, VIS high APs inhibit adipogenic differentiation of SUB and VIS low APs in vitro through the secretion of soluble inhibitory factor(s) including insulin-like growth factor binding protein 2 (Igfbp2). The number of VIS high APs increased with VIS adipose tissue expansion and their abundance in vivo causes hypertrophic growth, fibrosis, inflammation and impaired glucose and insulin tolerance. This study unveil the presence of APs unique to VIS fat involved in the paracrine regulation of adipogenesis and tissue remodeling.

Project description:Erythropoiesis from pluripotent stem cells is very complex, and mirrors that observed in developmental erythropoiesis, with primitive and definitive waves. For a better understanding of their potential for their use in blood transfusion, they were compared to erythropoiesis from cord derived CD34 haematopoietic stem cells. We used microarrays to detail the global programme of gene expression underlying different stages of erythropoiesis from each source, pluripotent or haematopoietic stem cells.

Project description:Studies of human erythropoiesis have relied, for the most part, on the in vitro differentiation of hematopoietic stem and progenitor cells (HSPC) from different sources. Here, we report that despite the common core erythroid program that exists between cord blood- and peripheral blood-HSPC induced towards erythroid differentiation in vitro, significant functional differences exist. We undertook a comparative analysis of human erythropoiesis using these two different sources of HSPC and differentiated them in vitro. We observed that cells derived from cord blood proliferate 4.5 times more than cells derived from peripheral blood. However, these cells present a delay in their differentiation pattern due to increased quantities of progenitors, notably CFU-E. Using our method of immunophenotyping for the study of erythroid progenitors, we document the presence and maintenance of a specific population in peripheral blood-derived erythroid progenitors. This population, defined as IL3R-GPA-CD34+CD36+, has the ability to form both BFU-E and CFU-E colonies in colony-forming assays, reflecting a higher potential. To further understand the differences between cord blood- and peripheral blood- HSPC, we sorted all stages of erythropoiesis from both sources and compared their transcriptome. We document differences at the CD34, BFU-E, poly- and orthochromatic stages. Among the genes presenting the highest differences in expression, many are involved in the regulation of the cell cycle and autophagy. Altogether, our studies provide a qualitative and quantitative comparative analysis of human erythropoiesis and highlight functional differences, critical to our understanding of the impact of the developmental origin of HSPCs on erythroid differentiation.

Project description:We investigated the miRNA expression in ex vivo human erythroid cultures from umbilical cord blood (UCB)-derived CD34 cells. Hypothetically, the decline of certain miRNAs may promote erythropoiesis by unblocking expression of key functional proteins while the up-regulation of other miRNAs may block commitment to non-erythroid lineages. Therefore, discovering the patterns and sequence of miRNA expression during hematopoietic differentiation will provide insights about the functional roles of these tiny noncoding genes. Human CD34+ cells were cultured for 48 hours in the erythroid differentiation media described above with the addition of 40 ng/mL of FMS-like tyrosine kinase 3 (flt-3; Miltenyi Biotech) and 15 ng/mL of Granulocyte colony-stimulating factor (G-CSF; Amgen). Cells were cultured in the presence of the drugs of interest for 24 hours. RNA was purified using Trizol (Invitrogen). RNA was amplified and labeled by in vitro transcription and hybridized to the Affymetrix HT Human Genome U133A Array. Three replicates per condition.
