SW480 cells undergoes postmitotic neuron-like differentiation in response to EZH2 knockdown

ABSTRACT: Formerly we found that combined inhibition of class I HDACs, DNMTs, EZH2 and LSD1 with chemical inhibitors in a variety of cancer cells caused neuronal differentiation. Our recent study demonstrated that functional knockdown of EZH2 alone in SW480 cells can also lead to significant neuronal differentiation. We used microarray to analyze global gene expression change in SW480 cells after neuronal differentiation induced by EZH2 knockdown.

Project description:Cancer cells are not fully differentiated cells. Re-differentiation of cancer cells is expected to drive the loss of cancer cell malignancy. Project description:SW480 is a colorectal cancer cell line. We have shown that cancer cells are characteristic of neural stem cells and neural stemness contributes to cell tumorigenicity. We found that during serial transplantation of SW480 cells into nude mice via subcutaneous injection (in vivo passaging), the cells enhanced their neural stemness, tumorigenicity and pluripotent differentiation potential.
