Project description:Monocytes are central for atherosclerotic vascular inflammation. The human non-classical, patrolling subtype, which expresses high levels of CD16 and fractalkine receptor CX3CR1, strongly associates with cardiovascular events. Compared to classical CD14+ monocytes or transwell cocultures, CD16+ monocytes enhanced endothelial STAT1 and NFκB p65 phosphorylation, upregulated expression of CX3CL1 and IL-1β, numerous CCL and CXCL chemokines and molecules promoting leukocyte patrolling and adhesion such as ICAM1 and VCAM1.

Project description:Nlrp6-/- lamina propria Ly6C-hi monocytes in response to AOM/DSS have deficient TNFα production, but increased production of other pro-inflammatory cytokines as compared to WT NLRP6 is a member of the Nod-like receptor family, whose members are involved in the recognition of microbes and/or tissue injury. NLRP6 was previously demonstrated to regulate the production of IL-18 and is important for protecting mice against chemically-induced intestinal injury and colitis-associated colon cancer. However, the cellular mechanisms by which NLRP6 reduces susceptibility to colonic inflammation remain unclear. Here, we determined that NLRP6 expression is specifically upregulated in Ly6Chi inflammatory monocytes that infiltrate into the colon during dextran sulfate sodium (DSS)-induced inflammation. Adoptive transfer of WT Ly6Chi inflammatory monocytes into Nlrp6-/- mice was sufficient to protect them from mortality, significantly reducing intestinal permeability and damage. NLRP6-deficient inflammatory monocytes were specifically defective in TNFα production, which was important for reducing DSS-induced mortality and dependent on autocrine IL-18 signaling by inflammatory monocytes. Our data reveal a previously unappreciated role for NLRP6 in inflammatory monocytes, which are recruited during intestinal injury to promote barrier function and limit bacteria-driven inflammation. This study also highlights the importance of early cytokine responses, particularly NLRP6-dependent and IL-18-dependent TNFα production in preventing chronic dysregulated inflammation.

Project description:Chronic inflammation promotes cancer progression by affecting the tumor cells and their microenvironment. Here, we demonstrate that the continuous stimulation (6 weeks) of triple-negative breast tumor cells (TNBC) by the potent pro-inflammatory cytokines tumor necrosis factor α (TNFα) and interleukin 1β (IL-1β) had a robust effect on gene expression, demonstrating that the cancer cells have been skewed to strong pro-inflammatory phenotype. Moreover, continuous TNFα + IL-1β stimulation has reduced cell-to-cell contacts in the cancer cells and has induced metabolic plasticity in TNBC cells: this was exemplified by increase in active mitochondria area and elevations in the glycolytic as well as mitochondrial respiratory potential of the cancer cells (OXPHOS). Glycolysis has induced p65 activation and has led to increased transcription and expression of pro-metastatic chemokines such as CXCL8, CXCL1 and CCL2 and sICAM-1; agreeing with the ability of these chemokines to induce the recruitment of deleterious myeloid cells to tumors, mainly inhibition of glycolysis has given rise to reduced monocytic cell migration, in vitro and in vivo, in response to cancer cell supernatants obtained following continuous TNFα + IL-1β stimulation. Such inflammation-induced metabolic plasticity, which promotes metastatic cascades in TNBC, may have important clinical implications in the treatment of TNBC patients

Project description:Macrophage population in most mammalian organs consists of cells of different origin, with the exception of the central nervous system and the liver, where macrophages of monocytic origin are almost completely absent. The reasons for such distribution and the phenomenon of coexistence of the two separate macrophage lineages with different origin in mammals remain poorly understood. In present study we compared Kupffer cells and monocytes by the immunophenotype, gene expression profile, proteome and pool of mircoRNA. Observed differences do not allow to consider the resident liver macrophages as purely M2 macrophages or monocytes have purely M1 features. Two populations of macrophages of monocytic origin and resident macrophages have fundamentally different roles in maintaining homeostasis. Monocytic macrophages are involved in the regulation of inflammation, and resident macrophages are involved in the regulation of specific organ functions (nitrogen metabolism, complement system protein synthesis). However, if other indicators would be considered as markers of macrophage activity, it is worth noting that Kupffer cells possess some features of M2 macrophages. This is indicated by their expression profile of let-7b/c/d/e miRNAs, a high content of proteins associated with oxidative phosphorylation, as well as an increased level of synthesis of Arg1, IL10.

Project description:Myocardial ischemia-reperfusion (I/R) injury represents a major clinical challenge with limited therapeutic options. This study examines the role of Olfactory Receptor 2 (Olfr2) in monocytes during myocardial I/R injury. We utilized RNA sequencing to analyze monocytes isolated from the spleens of wild-type (WT) mice, investigating their response to low-dose LPS priming and subsequent treatment with octanal (Oct), a specific agonist of Olfr2. Principal component analysis (PCA) revealed distinct clustering among experimental groups, highlighting significant gene expression changes in pathways related to cAMP/PKA signaling and inflammasome activation. Notably, the low-dose LPS+Oct treatment induced substantial upregulation of genes such as Adcy, Pka, Nlrp3, Pycard, Casp4, Gsdmd, and Nr4a1. Moreover, monocytes subjected to low-dose LPS+Oct exhibited increased expression of pro-inflammatory cytokines (IL-1β, IL-12a, IL-18, and TNF), chemokines (CCL3, CCL4, CXCL1, CXCL2, and CXCL3), and demonstrated enrichment in pathways associated with oxidative stress, positive chemotaxis, and the production and secretion of inflammatory cytokines (IL-1β, IL-8, TNF).

Project description:Objective The vascular endothelium provides a unique interaction plane for plasma proteins and leukocytes in inflammation. It has been established that particular proteins, including ICAM1, VCAM1 and SELE, are upregulated on the endothelial cell surface in the presence of pro-inflammatory cytokines Tumor Necrosis Factor α (TNFα) and Interleukin 1β (IL-1β). We now map cytokine-induced proteomic alterations to gain further insight into endothelial inflammation. Approach and results We evaluated alterations in the in-depth proteome, cell surface proteome and phosphoproteome after 24 hours of cytokine stimulation. In addition, protein expression profiles were measured after 4, 8, 16 and 24 hours of stimulation. We found that cytokine-stimulation induced a two-step response. After 4 hours, initial protein alterations were detected, including upregulation of ICAM1 and SELE, followed by a second burst of regulation after 8 to 16 hours, which included proteins involved in antigen processing, cytokine production, virus defense and ECM-interaction. Furthermore, we found that TNFα and IL-1β provoke a highly similar endothelial response with few specific alterations. Using a novel cell surface proteomics approach, we observed highly similar changes on the cell surface. Two surface proteins displayed altered labeling of specific epitopes, suggesting that these sites are modified or involved in interactions. Combined, our integrated proteomic data describes a full array of affected processes: leukocyte interaction, self-antigen presentation, cytokine production, nitric oxide production and extracellular matrix interaction. Conclusions Our study provides a detailed quantitative proteomic analysis of endothelial inflammation in which novel inflammation-responsive proteins were uncovered, and a two-step cytokine response was identified.
