RNA-Seq Analysis of Wild Type and LGMN-/- Peritoneal Macrophage Transcriptomes.

ABSTRACT: Research on the interaction between tumour-associated macrophages (TAMs) and tumour cells has generated great interest in the potential use of macrophages as a therapeutic strategy against tumour cells. Here, we show that deletion of legumain, a conversed proteolytic enzyme in macrophages, promotes senescence of macrophages and tumour cells. Peritoneal macrophages isolated from wide type and lgmn-/- mice were co-cultured with EO771 breast cancer cells for 4 days. Total RNA was extracted for RNA-Seq analysis, cDNA library construction and sequencing were performed using BGISEQ-500 platform.

pH-gated nanoparticles selectively regulate lysosomal function of tumour-associated macrophages for cancer immunotherapy

Project description:Tumour-associated macrophages (TAMs), as one of the most abundant and phagocytic tumour-infiltrating immune cells, play a pivotal role in tumour antigen clearance and immune suppression. M2-like TAMs present a heightened lysosomal acidity and protease activity, which limits the function of antigen cross-presentation. How to selectively reprogram the antigen-destroying TAMs to a restorative phenotype for efficient anti-tumour immunity is challenging. Here, we report a pH-gated nanoadjuvant (PGN) that selectively targets the lysosomes of M2-like TAMs in tumours rather than the corresponding organelles from macrophages in healthy tissues. Enabled by the PGN nanotechnology, M2-like TAMs are specifically switched to M1-like phenotypes with tuned-down lysosomal function featured by attenuated lysosomal acidity and cathepsin activity for improved antigen cross-presentation, thus provoking adaptive immune response and sustained tumour regression. Our findings provide new insights into how to specifically regulate lysosomal function of TAMs for efficient cancer immunotherapy.

Characterisation of human in vitro tumour-associated macrophage models to define translational relevance

Project description:Tumour-associated macrophages (TAMs) are key components of the tumour microenvironment with a demonstrated ability to modulate anti-tumour T-cell responses and immunotherapy outcomes. With increasing realisation that the M1/M2 paradigm does not reflect the complexity of macrophage phenotypes in cancer patients, an urgent need has arisen to develop improved, translatable in vitro models for human TAMs. To address this gap, we have screened conditioned media from a panel of tumour cell lines for their ability to induce suppressive marker upregulation on human monocyte-derived macrophages (hMDMs), as well as active T-cell immunosuppression. We performed secretome characterization of these tumour-conditioned media (TCM) to shed light on cancer cell-derived soluble factors that may contribute to TAM polarisation. Furthermore, we characterized the proteomic and transcriptomic signatures of macrophages exposed to either TCM or primary ascites fluid from ovarian cancer patients and performed bioinformatics analysis to determine the most translationally relevant models of TAMs. In summary, our work provides mechanistic insights on tumour-macrophage crosstalk in the context of establishing suppressive TAM phenotypes and addresses the long-standing gap of defining translationally relevant human in vitro TAM models.

Expression data from in vitro-induced non-hypoxic tumour associated macrophages (TAMs)

Project description:Active immunotherapy is a promising strategy for anti-angiogenic cancer therapy. Recently, we have reported that a vaccine using human umbilical vein endothelial cells (HUVECs) induced specific anti-endothelial immune responses in the most of immunized patients, and resulted in tumor regression in some patients with recurrent malignant brain tumors, whereas not in colorectal cancer patients. In this study, we hypothesized that non-hypoxic perivascular tumor associated macrophages (TAMs) in colorectal cancer, but not in glioblastoma, might negatively alter the therapeutic efficacy of anti-angiogenic active immunotherapy. To test this hypothesis, we examined global gene expression profiles of non-hypoxic macrophages stimulated in vitro by soluble factors released from tumor cells of human glioblastoma U-87MG (‘brain TAMs’) or colorectal adenocarcinoma HT-29 (‘colon TAMs’). Murine non-hypoxic TAMs were induced in vitro by incubation with soluble factors released from human cancer cell lines U-87MG ('brain TAMs') or HT-29 ('colon TAMs'), for RNA extraction and subsequent hybridization on Affymetrix microarrays. To evaluate homogeneous macrophage populations at different tumour developmental stages, RNA aliquots of control macrophages and TAMs obtained at five different time-points, i.e. 8h, 16h, 24h, 32h and 40h, were pooled and used for screening of differentially expressed genes. The experiments for TAMs as well as for control unstimulated macrophages were performed in triplicates.

M1hot tumour associated macrophages are positively associated with tissue-resident memory T cell responses and survival outcomes in human lung cancer (bulk).

Project description:Tumour-associated macrophages (TAMs) are classified as M1 (“anti-tumour”) and M2 (“pro-tumour”), but their balance has not yet been accurately established in human cancer tissue. Here, we present bulk and single-cell transcriptomic data from human TAMs isolated from lung cancer compared with autologous non-tumour involved lung tissue macrophages (N-TAM). TAMs have a profoundly different transcriptomic profile from N-TAMs and displayed a broad M2 pro-tumour signature that was homogeneous across all the lung cancer patients studied. However, in some tumours, we found TAM populations that simultaneously displayed strong M1 gene expression pattern. These M1hot TAMs appeared to contribute to effective anti-tumour immunity via expression of T cell chemo-attractants (CXCL9, 10 and 11). We found that M1hot TAMs expressed low levels of FABP3, 4 and 5, take up less fatty acids and showed “metabolic generosity” to tissue resident memory T cells (TRM) by allowing them access to this essential nutrient resource that is required for their maintenance. Importantly, tumours harbouring M1hot TAMs were highly enriched for TRM cells and were also associated with improved overall survival outcomes. Our findings suggest that immunotherapeutic approaches aimed at inducing the expression of M1hot program in TAMs rather than generically eliminating all TAMs are likely to be beneficial.

Syndecan-3 positively regulates the pro-inflammatory function of macrophages

Project description:The tumour microenvironment (TME) is a highly structured ecosystem that surrounds a tumour and plays a crucial role in tumorigenesis. As one of the most abundant cell types in the TME, tumour-associated-macrophages (TAMs) can promote disease progression and resistance to therapy. Syndecan-3 (SDC3) is a cell-surface heparan sulphate proteoglycan expressed by TAMs, although its functional relevance in these cells remains unknown. Here, we demonstrated that proinflammatory cytokines drive the expression of SDC3 on the cell surface of macrophages. Genetic ablation of SDC3 in macrophages led to aberrant proliferation, adhesion and expression of CD40 and CD86 surface markers. Moreover, SDC3 defective macrophages exhibited distinctive gene expression patterns, leading to impaired tumour cell phagocytosis and increased tumour cell proliferation. Mechanistically, a decrease in the secretion of pro-inflammatory cytokines was observed in SDC3 KO macrophages, concomitant with impaired T cell effector functions. Additionally, a higher angiogenic capacity was observed in endothelial cells when co-cultured with macrophages deficient for SDC3, possibly mediated through an increased release of VEGFA, PECAM-1 and IL-8 by SDC3 KO cells. Collectively, we have identified SDC3 as a modulator of macrophage functions aiming at supporting a pro-inflammatory and anti-tumour phenotype in these cells.

Autophagic adaptation to oxidative stress alters peritoneal residential macrophage survival and ovarian cancer metastasis [macrophages]

Project description:Tumor-associated macrophages (TAMs) affect cancer progression and therapy. Ovarian carcinoma often metastasizes to the peritoneal cavity. Here, we found two peritoneal macrophage subsets in mice bearing ID8 ovarian cancer based on the Tim-4 (T-cell immunoglobulin and mucin domain containing 4) expression. Tim-4+ TAMs were embryonically originated and locally sustained while Tim-4- TAMs were replenished from circulating monocytes. Tim-4+ TAMs, but not Tim-4- TAMs, promoted tumor growth in vivo. Relative to Tim-4- TAMs, Tim-4+ TAMs manifested high oxidative phosphorylation and adapted mitophagy to alleviate oxidative stress. High levels of arginase-1 in Tim-4+ TAMs contributed to potent mitophagy activities via weakened mTORC1 activation due to low arginine resultant from arginase-1-mediated metabolism. Furthermore, genetic deficiency of autophagy element FIP200 resulted in Tim-4+ TAM loss via ROS-mediated apoptosis, and elevated T cell-immunity and ID8 tumor inhibition in vivo. Moreover, human ovarian cancer-associated CRIg (complement receptor of the Immunoglobulin superfamily) positive macrophages were transcriptionally, metabolically, and functionally similar to murine Tim-4+ TAMs. Thus, targeting CRIg+ (Tim-4+) TAMs may potentially treat ovarian cancer patients with peritoneal metastasis.

Tumour-associated microglia/macrophages in Glioblastoma are heterogenous and display higher immunological reactivity under aconitate decarboxylase 1 deficiency

Project description:In Glioblastoma (GBM), tumour-associated microglia/macrophages (TAMs) represent the most abundant cells of the stromal compartment contributing to tumour immune escape mechanisms. Thus, strategies targeting TAMs are emerging as promising objectives for immunotherapy. However, TAMs heterogeneity is only starting to emerge and little is known about their contribution to GBM progression. Here, we comprehensively study the molecular changes of TAMs in the GL261 GBM mouse model by single-cell RNA-sequencing across GBM progression and in the absence of Acod1/Irg1, a key gene involved in macrophage metabolic reprogramming. We identify distinct TAM profiles, mainly based on their ontogeny, which recapitulate microglia- versus macrophage-like features showing key transcriptional differences and rapidly adapting along GBM stages. Notably, we uncover a decreased antigen-presenting cell signature in TAMs along tumour progression that is less prominent in Acod1/Irg1-deficient mice. Overall, our results provide insights into TAMs heterogeneity and highlight a potential role for Acod1/Irg1 in TAMs polarization along GBM progression.
