Comparison of transcriptome in gonads and somatic tissues in mice

ABSTRACT: RNA from female gonads (C57/BL6xDBA/2 background) at different developmental stages (11.5dpc, 12.5dpc, 14.5dpc, 16.5dpc, 18.5dpc, 20.5dpc – dpc: days post coitum) together with RNA from adult testes and 17 different somatic tissues was used for transcriptome comparison.

Project description:Following sex determination, XY and XX gonads develop into a testis and an ovary, respectively. Depending on the sex of the gonad, resident germ cells will subsequently be committed to either spermatogenesis or oogenesis. In this study we took advantage of the Wv/Wv mouse genetic model, in which gonads are almost devoided of germ cells, to uncover gene expression underlying fetal germ cell development. Male and Female gonads were collected at 12, 14 and 16 days post coitum (dpc) from wild-type and Wv/Wv mouse embryos. Total RNAs were extracted and hybridized on Affymetrix microarrays. Expression signals from male and female gonads or from wild-type and mutant gonads were compared to identify sexually dimorphic genes as well as genes expressed in germ cell during fetal gonad development.

Project description:Meiosis is a specialized division that is specifically engaged in germ cells. This program is finely and dichotomically regulated: germ cells enter meiosis at fetal life in ovaries (13.5 dpc) and only at post-natal life in testes (8 dpp). We used microarrays to determine the gene expression modifications in Meioc mutant gonads displaying early meiotic defects. 14.5 dpc ovaries and 8 dpp testes from Meioc+/+ and Meioc-/- mice (NMRI) were dissected and processed for RNA extraction and hybridization on Affymetrix microarrays. Two pools were analysed for each genotype and each sex. We sought to constitutes pairs (A and B) of wild type and knock-out pools from embryos and pups that were homogenous for developmental stages and from matched littermates. 10 ovaries from 5 embryos composed each pool for females and 4 testes from 4 pups composed each pool for males.

Microarray analysis of the Asian seabass (Lates calcarifer) gonads

Project description:In this study, we attempt to characterize the transcriptomic profile of the Asian seabass gonads at various developmental stages. The protandric Asian seabass or barramundi (Lates calcarifer) typically matures as a male at approximately 2M-bM-^@M-^S4 years of age and then changes sex to a female in later years. For this experiment, Asian seabass of several ages were collected from the Marine Aquaculture Center of the Agri-Food & Veterinary Authority of Singapore and from farms around Singapore. There were no treatments carried out in this experiment. The gonads were examined by histology and classified according to sexual maturation status as described by Guiguen and colleagues (Guiguen et al. Environmental Biology of Fishes, 1994). Altogether, we analyzed 22 gonadal samples that could be classified into six different types of gonads. Total 22 samples: Adult Ovaries (F3-stage; 5 years old fish) : 4 Adult Testes (M3-stage; 5 years old fish) : 4 Early Testes (M3-stage; 8-9 months old fish) : 3 Early Transforming Gonads (>2 years old fish) : 3 Late Transforming Gonads (>2 years old fish) : 4 Undifferentiated Gonads (4.5 months old fish) : 4
