INDETERMINATE-DOMAIN 4 (IDD4) coordinates immune responses with plant-growth in Arabidopsis thaliana.

ABSTRACT: We report the transcriptome composition of idd mutant with and without flg22 treatment compared to WT (ColO). Furthermore, we provide transcriptome data of a IDD4ox line and IDD4-phospho-peptide modified versions without treatment compared to WT (ColO), respectively. In order to identify the genomic regions targeted by IDD4 in vivo, Chromatin-Immunoprecipitation (ChIP) was performed followed by deep sequencing on the Illumina High-Sequencing2000 platform (SEQ).

RNA-seq analysis of flg22-induced gene expression changes in wild type, asr3-1 mutant and OX9 overexpression line

Project description:The correlation coefficient (R) for the expression profile of all transcripts between WT and asr3-1, and between WT and OX9 without flg22 treatment is close to linear, suggesting that ASR3 does not affect general gene transcription. Hierarchical clustering analysis flg22-induced genes in any of the three genotypes suggested that the asr3-1 mutant displayed overall enhanced flg22 response whereas OX9 displayed overall reduced responses compared to WT plants. RNA-seq analysis was performed with 10-day-old seedlings of WT, asr3-1 and 35S:ASR3-HA transgenic line OX9 with or without 100 nM flg22 treatment for 30 min. Two independent repeats were performed for RNA-seq analysis. For each repeat, equal amount of RNA from two biological replicates was pooled for RNA-seq library construction. RNA-seq library preparation and sequencing were carried out on an Illumina HiSeq 2500 platform with 100 nt single end reads.

The latter makes co-implanted MSCs visualization possible by using MSCs labeled by GFP under FACS and single cell microscopy. Two-condition experiment, COLO 320 xenograft without rat MSCs [COLO320 MSC(-)] vs. COLO 320 xenograft with rat MSCs [COLO320 MSC(+)]. Biological replicates: 1 control, 1 sample, paired xengraft tumor cells grown and harvested from the same mouse host. One replicate per array.

Self-amplifying Loop of NF-κB and Periostin Initiated by PIEZO1 Accelerates Mechano-induced Senescence of Nucleus Pulposus Cells and Intervertebral Disc Degeneration

Project description:Abnormal mechanical load is a main risk factor of intervertebral disc degeneration (IDD), and cellular senescence is a pathological change in IDD. Additionally, extracellular matrix (ECM) stiffness promotes human nucleus pulposus cells (hNPCs) senescence. However, the molecular mechanism underlying mechano-induced cellular senescence and IDD progression is not yet fully elucidated. First, we demonstrated that mechano-stress promoted hNPCs senescence via NF-κB signaling. Subsequently, we identified periostin as the main mechano-responsive molecule in hNPCs through unbiased sequencing, which was transcriptionally upregulated by NF-κB p65; moreover, secreted periostin by senescent hNPCs further promoted senescence and upregulated the catabolic process in hNPCs through activating NF-κB, forming a positive loop. Both Postn (encoding periostin) knockdown via siRNA and periostin inactivation via neutralizing antibodies alleviated IDD and NPCs senescence. Furthermore, we found that mechano-stress initiated the positive feedback of NF-κB and periostin via PIEZO1. PIEZO1 activation by Yoda1 induced severe IDD in rat tails without compression, and Postn knockdown alleviated the Yoda1-induced IDD in vivo. Here, we reported for the first time that self-amplifying loop of NF-κB and periostin initiated via PIEZO1 under mechano-stress accelerated NPCs senescence, leading to IDD. Furthermore, periostin neutralizing antibodies, which may serve as potential therapeutic agents for IDD, interrupted this loop.
