Small RNA Sequencing of mouse prefrontal cortex after chronic social defeat stress (CSDS)


ABSTRACT: Chronic stress lastingly affects brain physiology but the underlying mechanisms remain mostly elusive. Here, we profiled synapse-specific, long-term posttanscriptional gene expression regulation in the mouse prefrontal cortex after chronic social defeat stress (CSDS). To this end, synaptosomes from the prefrontal cortex were enriched from mice that underwent 3 weeks of CSDS plus 1 week of recovery, selecting only mice with high susceptibility to the stress and pooling each 15 mice per sample. The synaptosome material was analyzed for protein expression by in vivo 15N metabolic labeling and quantitative mass spectrometry, miRNA expression (Small RNA Seq presented here) and circRNA expression. Overall design: Total RNA from synaptosomes enriched from BL6 prefrontal cortex, 3 replicates of CSDS mice, 3 replicates of Control mice (standard housing). Each sample is pooled from 15 individual animals.
