Gene expression profiles of Ecrg4-treated mouse microglia

Project description:Esophageal cancer-related gene 4 (Ecrg4), a hormone-like peptide, is thought to be a tumor suppressor, however little is known about the mechanism of how Ecrg4 suppresses tumorigenesis. Using the mouse glioma–initiating cell models, we identified Ecrg4 acts as a tumor suppressor in vivo. To characterize the function of Ecrg4 towards microglia, an innate immune cell in central nervous system, we performed gene expression microarray analysis for primary microglia treated with or without recombinant Fc-fused Ecrg4 fragments. We demonstrate here that Ecrg4 fragments, amino acid residues 71-132 and 133-148, which are produced by the proteolitic cleavage, induced the expression of pro-inflammatory cytokines in microglia. Thus, Ecrg4 acts as a cytokine/chemokine inducer, which activates the immune cells to eradicate tumor.

ECRG4 as a novel diagnostic plasma marker detected in the early phase of Alzheimer’s disease

Project description:Current therapeutic methods for Alzheimer’s disease (AD) aim to delay its progression; therefore, developing a reliable method for diagnosing the early phases of the disease (EAD) is crucial. Based on the finding that the secretory protein Esophageal Cancer-Related Gene 4 (ECRG4) is elevated in the hippocampus of AD patients, we developed a new ECRG4 ELISA system. Here, we show that ECRG4 peptides, particularly those containing amino acids 108 to 132, are increased in the plasma of approximately 25% of mild cognitive impairment and half of AD patients, compared to non-dementia individuals. RNA sequencing analysis of the plasma revealed decreased levels of CHST3 mRNA, which is highly expressed in oligodendrocyte-lineage cells, in ECRG4-positive patients. We demonstrate that amino acids 71 to 107 exert cytotoxicity to oligodendrocytes in culture and decrease the intensity of the oligodendrocyte marker in the corpus callosum when injected into the brain. This results in extravascular IgG leakage due to the accumulation of oligodendrocyte precursor cells (OPCs), which regenerate myelin, around the blood vessels. Given that similar phenomena were detected in the brains of EAD patients, these results suggest that ECRG4 is involved in the progression of AD through vascular destruction in a manner dependent on oligodendrocyte defects.

Gene expression profile of Ba/F3 variant (#BEB1) cells that have binding ability to Ecrg4

Project description:Esophageal cancer-related gene 4 (Ecrg4) encodes a hormone-like peptide, and is thought to be involved in a variety of physiological phenomena including tumor suppressor function. Recent progress of Ecrg4 reveals that Ecrg4 peptides are pro-inflammatory and induce the expression of several cytokines and chemokines. However, detailed molecular mechanisms of Ecrg4 signaling, especially the receptors for Ecrg4 peptides, remain poorly understood. Here, using retrovirus-mediated expression cloning, we identified lectin-like oxidized low-density lipoprotein receptor-1 (LOX-1) as a membrane protein bound for amino acid residues 71-132 of Ecrg4, Ecrg4(71-132). Moreover, not only LOX-1 but also several scavenger receptors such as Scarf1, Cd36 and Stabilin-1 efficiently internalized Ecrg4(71-132) into cells. The broad scavenger receptor competitive inhibitor polyinosinic acid reduced both the internalization of Ecrg4(71-132) and activation of NF-kB in microglia. In addition, this activation is dependent on Myd88, an adaptor protein to recruit signaling proteins to TLR and IL-1R receptors with induction of several immune response. These data suggest that multiple scavenger receptors recognize Ecrg4(71-132) and transduce its signals along with TLR/IL-1R receptors in microglia.

Gene expression data from mouse ES cell-derived primitive NSCs and definitive NSCs

Project description:Primitive neural stem cells (NSCs) could be derived from pluripotent mouse embryonic stem (ES) cells, and then differentiate into definitive-type neural stem cells which resemble NSCs obtained from the central nervous system. Hence, primitive NSCs define an early stage of neural induction and provide a model to understand the mechanism that controls initial neural commitment. In this study, we performed microarray assay to analyze the global transcriptional profiles in mouse ES cell-derived primitive and definitive NSCs and to depict the molecular changes during the multi-staged neural differentiation process. Primitive NSCs derived directly from ESCs in Lif (p-NSC_L), primitive NSCs that were sub-cultured in the presence of Lif and FGF (p-NSC_LF), as well as definitive NSCs derived from primitive NSCs in medium containing FGF and EGF, were collected for RNA extraction and hybridization on Affymetrix microarrays. Mouse ESCs and NSCs obtained from mouse embryonic brain (E11.5) were included for controls. For each cell type, we collected two biological replicate samples for microarray analysis.
