SAGA Complex Subunits in Candida albicans Differentially Regulate Filamentation, Invasiveness and Biofilm Formation

Project description:In this study we were interested in studying the role of SAGA subunits in various cellular processes- morphogenetic changes, growth, invasiveness, biofilm formation and to check the role of these subunits under various cellular and genotoxic conditions. In this work, we investigated conditional and null mutants of components of the SAGA complex modules; Ngg1 of the HAT module, Ubp8 of the Dub module, Tra1 of the recruitment module, Spt7 of the architecture module, and Spt8 of the TBP interaction unit to assess their role in processes such as filamentation, invasiveness, and biofilm formation. Spt7 and Spt8 deletion in Candida albicans resulted in ifilamentatation, invasiveness in YPD media at 30 degrees Celsius and also showed sensitivity to antifungal drugs. Wild type SN148 do not make any filaments in YPD at 30 degrees Celsius and didn't show antifungal sensitivity . The aim was to look for transcription profiling of SAGA mutants against wild type to find genes up and down regulated in the mutant especially those ones critical for filamentation, invasiveness and antifungal drug resistance.

Gis2 mutant versus wild-type cells

Project description:To measure the relative changes of gene expression upon Gis2p depletion, 50 ml of delta Gis2 cells or the respective wild-type strain BY4741 were grown in YPD at 30 degrees celsius to an OD600 of 0.5- 0.6. Cells were harvested by centrifugation and washed twice with 5ml of bidistilled water (ddH2O). RNA was isolated by hot phenol extraction for microarray analysis. 20 ug of total RNA derived from wild-type and knock-out cells were reverse transcribed in the presence of 5-(3-aminoallyl)-dUTP and natural dNTPs with a mixture of N9 and dT20V primers, and cDNAs were covalently linked to Cy3 and Cy5 NHS-monoesters (GE HealthSciences Cat# RPN5661), respectively, and competitively hybridized onto yeast oligo arrays at 42 degrees celsius for 14 hours in formamide-based hybridization buffer. Microarrays were scanned with an Axonscanner 4200 (Molecular Devices) and analyzed with GenePix Pro 5.1 (Molecular devices) before uploading into Stanford Microarray Database.

Genome-wide expression analysis of dikaryotic colonies of Schizophyllum commune under various conditions: wild-type, delta-wc2, delta-wc2 hsp3(pr)-wc2

Project description:The blue light receptor WC-2 was shown to be involved in mushroom development in the basidiomycete Schizophyllum commune. When the gene encoding WC-2 was deleted, no mushrooms formed and colony morphology was radial. This phenotype was similar to the wild-type colony grown in the dark. This phenotype could be complemented by transforming the wc-2 deletion strain with a construct encompassing the wc-2 coding sequence under the control of the heat inducible promoter hsp3. A daily heat shock of 1 hour at 42 degrees Celsius resulted in mushroom development and an asymmetrical colony. In this study we performed a genome-wide expression analysis on dikaryons of wild-type (not heat shocked), delta-wc2 (heat shocked or not heat shocked) and the complemented strain delta-wc2 hsp3-wc2 (heat shocked or not heat shocked).
