Project description:Male Sprague-Dawley rats [Crl:CD®(SD)IGS BR], weighing ~250 g at study initiation were obtained from Charles River Laboratories, Inc. (Wilmington, MA). Rats were housed singly in ventilated, stainless steel, wire-bottom hanging cages and fed non-certified Rodent Chow (Harlan Labs, Madison, WI) and water ad libitum and acclimated for at least 5 days after arrival. Rats were randomly assigned to various treatment groups (3 rats/group) and were dosed once daily by oral gavage with vehicle (0.2% hydroxypropylmethylcellulose at a dose volume of 10 ml/kg) or with 30, 100, or 200 mg/kg of A-998679. Project description:Mouse adult female brain’s cortex (C57BL/6, Charles River Laboratories, Inc.) was isolated and stored immediately at -80°C. Subsequently, the mRNA (15µg) was isolated using TRIzol Reagent and MicroFastTrack 2.0 Kit (Invitrogen). A Sample of 5µg was assessed on Affymetrix Mouse 430.2 array. Aliquots from the leftovers of the same cortical mRNA were diluted to single-cell RNA levels (0.1, 1, and 10 pg) and independently aRNA amplified for a total of 2 and 4 rounds and assessed on Affymetrix Mouse 430.2 arrays.

Project description:Microbiome regulation of lipid metabolism Germfree male C57BL/6J mice were purchased at 8 weeks of age from Charles River Laboratories (L'Arbresle, France). Mice were treated with 10^8 CFU/mL E. coli M8 strain (isolated from the feces of an ob/ob mouse) in drinking water for 14 days. At the end of the treatment, mice were subjected to an oral lipid tolerance test (OLTT) (with 6ml/kg of corn oil) after overnight fasting (14h). All mice were sacrificed 6 hours after the lipid tolerance test and ileum samples were collected for further analysis.

Project description:Male Sprague Dawley Crl CD BR rats (190-240g, 6-8 weeks old), were obtained from Charles River, UK. They were given access to R&M No.1 diet and tap water ad libitum. Animals were housed 5/sex in solid bottom cages maintained at 21oC +/- 2oC, 12 h light and 12 h dark. All animals were acclimatised for at least 5 days prior to commencement of the study. Furan (CAS 110-00-9, Sigma-Aldrich Co. Ltd, >99 % pure was prepared in corn oil vehicle on the day of use and stored in sealed brown bottles. Furan was administered orally via gavage in corn oil at 30mg/kg (5 daily doses per week). Animals (n=5) were removed and sampled after 3 months of furan treatment. An off-dose recovery group was included at the 3 month time point (1 month off-dose) to enable assessment of lesion reversibility.

Project description:Five ovariectomized (OVX) Brown Norway rats (Charles Rivers Laboratories, Wilmington, DE, USA) weighing 200-250 g received 10 µL of 17β-estradiol (E2) eye drops once daily in both eyes for three weeks [20]. The eye drops contained 0.1% (w/v) E2 in saline vehicle containing 20% (w/v) 2-hydroxypropyl-β-cyclodextrin. Five OVX control rats received 10 µL of this vehicle as eye drops for the same dosing regimen and duration. After 24 h of the last treatment, the animals were euthanized by CO2 overexposure, and their eyes were immediately enucleated followed by the isolation of the retina. The tissue samples were rinsed with saline and, then, blotted dry for preparation to label-free shotgun proteomic analyses. All procedures involving animals were reviewed and approved by the Institutional Animal Care and Use Committee at the University of North Texas Health Science Center before the initiation of the studies (approval number: 2018-0028). Project description:Detachments were created in adult male Brown-Norway rats (300?400 g; Charles River Laboratories, Wilmington, MA). Briefly, rodents were anesthetized with a 50:50 mix of ketamine/xylazine, and pupils were dilated with topical phenylephrine (2.5%) and tropicamide (1%). A 20-gauge microvitreoretinal blade was used to create a sclerotomy 2 mm posterior to the limbus, carefully avoiding lens damage. A subretinal injector (Glaser, 32-gauge tip; BD Ophthalmic Systems, Franklin Lakes, NJ) was introduced through the sclerotomy into the vitreous cavity and then through a peripheral retinotomy into the subretinal space. Sodium hyaluronate (10 mg/mL, Healon OVD; Abbott Medical Optics, Uppsala, Sweden) was slowly injected to detach the neurosensory retina from the underlying RPE. In all experiments, approximately one third to one half of the neurosensory retina was detached. Detachments were created in the left eye. The right eye served as the control, with all the steps of the procedure performed except for introduction of the subretinal injector and injection of the sodium hyaluronate. At varying intervals after creation of the detachment, the animals were euthanatized, and the eyes were enucleated.The retina was then dissected away from the retinal pigment epithelium taking just the detached portion in those eyes experimentally detached. All experiments were performed in accordance with the ARVO Statement for the Use of Animals in Ophthalmic and Vision Research and the guidelines established by the University Committee on Use and Care of Animals of the University of Michigan. Project description:WT C57BL/6 and TLR2-/- mice were used in this study. TLR2-/- mice were on a C57BL/6 background and were kindly provided by Dr S. Akira (Osaka University, Osaka, Japan). WT C57BL/6 mice were purchased from Charles River Laboratories (Wilminton, MA, USA). All the mice used in this study were males and more than 7 weeks old. Animals were kept and bred at Karolinska Institutet animal facility in a specific pathogen-free environment with standard room temperature and a 12-12 hour light/dark cycle. Food and water were provided ad libitum. Mice were killed by rapid neck disarticulation and the flexor digitorum brevis muscles were removed.
