In search for materials able to be colonized by a normal endothelium: сharacterization and NGS gene expression profiling of human primary endotheliocytes cultivated on electrospun 3D matrices

ABSTRACT: Endothelization of the luminal surface of vascular grafts is required for their long-term functioning. In order to select the best surface we have cultivated HUVEC on different 3D matrices and assessed the efficacy of cell proliferation and the changes in the gene expression profiles. 3D matrices were produced by electrospinning from solutions of poly(D,L-lactide-co-glycolide) (PLGA), polycaprolactone (PCL) and blends of PCL with gelatin (PCL-Gl) and glutaraldehyde-treated PCL-GL (PCL-Gl-glu) in hexafluoroisopropanol. Gene expression profiling was executed using Illumina HiSeq2500 platform.

Plasma functionalization of PCL nanofibers changes protein interactions with cells resulting in increased cell viability

Project description:The surface properties of electrospun scaffolds can greatly influence protein adsorption and thus strongly dictate cell-material interactions. In this study, we aim to investigate possible correlations between the surface properties of argon, nitrogen and ammonia/helium plasma-functionalized polycaprolactone (PCL) nanofibers and their cellular interactions by examining the protein corona patterns of the plasma-treated nanofibers as well as the cell membrane proteins involved in cell proliferation. As a result of the performed plasma treatments, PCL nanofiber morphology was preserved while wettability was improved profoundly after all treatments because of the incorporation of polar surface groups. Depending on the discharge gas, different types of groups are incorporated which influenced the resultant cell-material interactions. Argon plasma-functionalized PCL nanofibers, only enriched by oxygen-containing functional groups, were found to show the best cell-material interactions, followed by N2 and He/NH3 plasma-treated samples. SDS-PAGE and LC-MS clearly indicated an increased protein retention compared to non-treated PCL NFs. The most differential proteins surface a distinct molecular machinery mediating the increased proliferation and viability on plasma treated NF. Finally, differential protein analysis between the different plasma’s gives a first hint into the molecular underpinning of the superior performance of Argon NF.

Human Bone Marrow Stromal Cells cultured on 3D nanofiber scaffolds or 2D films

Project description:Microarray analysis was used to evaluate expression differences from a single donor human bone marrow stromal cells (hBMSCs) as a function of varied polymer-based tissue engineering scaffolds. These scaffolds include polycaprolactone (PCL) nanofibers (PCL_NF), films (PCL_SC), poly D,L-lactic acid (PDLLA) nanofibers (PDLLA_NF), films (PDLLA_SC), tissue culture polystyrene (TCPS) and TCPS with osteogenic supplements (TCPS_OS). The results revealed that scaffold structure was able to significantly affect gene expression, with nanofiber scaffolds inducing similar gene expression patterns to hBMCSs cultured with osteogenic media. A library of scaffolds prepared from polycaprolactone or poly D,L-lactic acid was sythesized and cultured with hBMSCs for 14 days with RNA extracted from cells on Day 1 and Day 14. Gene expression analysis was performed using BRB ArrayTools. SC = spun coat, BNF = big nanofiber, TCPS = tissue culture polystyrene, TCPS+OS = tissue culture polystyrene with osteogenic supplements. Transcriptome analysis revealed the symbiosis niche of 3D scaffolds to accelerate bone defect healing

Project description:3D printed scaffolds have been shown to be superior in promoting tissue repair, but the cell-level specific regulatory network activated by 3D printing scaffolds with different material components to form a symbiosis niche have not been systematically revealed. Here, three typical 3D printed scaffolds, including natural polymer hydrogel (Gelatin-methacryloyl, GelMA), synthetic polymer material (Polycaprolactone, PCL) and bioceramic (β-tricalcium phosphate, β-TCP), were fabricated to explore the regulating effect of the symbiotic microenvironment during bone healing. Enrichment analysis showed that hydrogel promotes tissue regeneration and reconstruction by improving blood vessel generation by enhancing oxygen transport and red blood cell development. The PCL scaffold regulates cell proliferation and differentiation by promoting cellular senescence, cell cycle and DNA replication pathways, accelerating the process of endochondral ossification and the formation of callus. The β-TCP scaffold can specifically enhance the expression of osteoclast differentiation and extracellular space pathway genes to promote the differentiation of osteoclasts and promote the process of bone remolding. In these processes, specific biomaterial properties can be used to guide cell behavior and regulate molecular network in the symbiotic microenvironment to reduce the barriers of regeneration and repair.

Cohesin HiChIP datasets of pluripotent and surface ectoderm cells with and without p63

Project description:Purpose: Assess changes in the three-dimensional (3D) chromatin architecture during surface ectoderm commitment and evaluate changes in 3D chromatin architecture in human embryonic stem cells and surface ectoderm cells with and without p63. Methods: Cohesin HiChIP profiles for human embryonic stem cells and surface ectoderm cells with and without p63 were generated in triplicate with deep sequencing on the Illumina HiSeq 4000 sequencer. Using HiC-Pro, paired end reads were aligned to hg19, duplicate reads were removed, and then were assigned to MboI restriction fragments, filtered for valid interactions, and used to generate binned interaction matrices of 10 kb resolution. Contacts were called using FitHiC and filtered for counts >= 10 and FDR < 0.001. Results: The greatest change in 3D chromatin architecture was observed between human embryonic stem cells and surface ectoderm cells, while moderate changes were observed in human embryonic stem cells with and without p63, and limited changes were observed in surface ectoderm cells with and without p63. Conclusion: Both p63 and the surface ectoderm morphogens direct changes in chromatin folding to establish the 3D chromatin landscape in the surface ectoderm.

Transcriptome analysis of rMSCs cultured for 4 hours in differerent substrate (TCP, PCL film, ISB-P fIlm)

Project description:We developed new isosorbide derivatives (propoxylated or ethoxylated isosorbide) which have more reactivity in PCL-PU copolymer synthesis than the original isosorbide. PCL-PU copolymer incorporated propoxylated isosorbide (ISB-P) exhibited notable mechanical performance (50 MPa tensile strength and 1150% elongation with hyperelasticity under cyclic load), ultra-cell-adhesive property, and enhanced osteogenic differentiation. There was no significant difference in surface characteristics between ISB-P and PCL), however, mRNA profiles showed higher expressions of heat shock proteins which have been reported to promote osteogenic differentiation at 4 h after cell seeding in rMSC cultured on ISB-P compared to PCL. In conclusion, the engagement of heat shock protein to the ISB-P surface during cell attachment may promote osteogenic differentiation in ISB-P.
