NLRP2 promotes proinflammatory and chemotactic responses in proximal tubular epithelial cells

ABSTRACT: In this study, we investigated the functional role of NLRP2 in proximal tubular epithelial cells by overexpressing it in human conditionally immortalized cells obtained from urine of a healthy subject. We found that NLRP2 markedly upregulates proinflammatory and chemotactic responses as well as downregulates interferon-mediated signaling pathways.

Oxysterols Modify NLRP2 in Epithelial Cells, Identifying a Mediator of Ozone-induced Inflammation.

Project description:Ozone (O3) is a prevalent air pollutant that causes lung inflammation following exposure. Previous studies demonstrate that O3 oxidizes lipids, such as cholesterol, in the airway to produce oxidized cholesterol products (oxysterols), such as secosterol-A (secoA), which form covalent linkages preferentially with lysine residues and consequently modify protein function. The breadth of proteins modified by this oxysterol as well as the biological consequences in the lung are unknown. Using an alkynyl-tagged form of secosterol-A and shotgun proteomics, we identified NLR Family Pyrin Domain Containing 2 (NLRP2) to be adducted at lysine (K1019) in the terminal leucine-rich-repeat, a known regulatory region for NLR proteins. We characterized NLRP2 expression in airway epithelial cells and used CRISPR-Cas9 knockout and shRNA knockdown of NLRP2 for analysis of inflammasome complex activity and pro-inflammatory mediator production following O3 exposure. We observed an increase in caspase-1 activity in response to O3, which was not altered by knocked down NLRP2 expression. O3-induced pro-inflammatory gene expression for CXCL1, CXCL2, and IL8 was further enhanced in NLRP2 knockout cells. Together, our findings uncover NLRP2 as a highly abundant, key component of pro-inflammatory signaling pathways in airway epithelial cells and formation of oxysterol-NLRP2 adduct as a novel mediator of O3-induced inflammation.

Loss of the Maternal Effect Gene Nlrp2 Impairs Embryonic and Extra-Embryonic Development, Unveiling a Novel Cause of Congenital Anomalies

Project description:Maternal effect genes (MEGs) are expressed in the oocyte and early mammalian preimplantation embryo. Impaired MEG function can impact offspring development and health. We investigated the consequences of postimplantation embryonic and extra-embryonic development in mice with maternal loss-of-function of Nlrp2, an MEG that encodes a protein of the subcortical maternal complex (SCMC). We employed three mating schemes, wildtype (WT) x WT, maternal Nlrp2-knock-out (KO) x WT, and WT x paternal Nlrp2-KO, to study the embryonic and extraembryonic phenotypes at embryonic day 11.5. Concepti of maternal Nlrp2-KO matings have striking abnormalities, including aberrant yolk sac vasculature, lower embryo to yolk sac area ratios, increased embryo resorption, and embryos with variably severe craniofacial abnormalities, and congenital heart defects. The placentas have significant morphological abnormalities, particularly in the junctional zone and labyrinth layers. This indicates that Nlrp2 is important for trophoblast development and placental architecture, supported by significantly altered gene expression profiles in the placentas of pregnant Nlrp2-KO mice. Taken together, these findings highlight the importance of maternal Nlrp2 for embryonic and placental development, and for the first time confirm that inactivating maternal effect mutations can cause disrupted placental development along with congenital cardiac and other anomalies. In conclusion, maternal loss of Nlrp2 profoundly impacts mouse embryonic and extra-embryonic development, highlighting a new role of MEGs. These findings pave the way for future research into genetic mechanisms underlying congenital anomalies and developmental disorders.

Rg1 improves motor recovery after spinal cord injury by suppressing astrocytic NLRP2 inflammasome via the TLR4–MyD88–NF-κB pathway

Project description:Spinal cord injury (SCI)-induced neuroinflammation and astrocyte pyroptosis severely impede motor functional recovery. Here, we report that ginsenoside Rg1 attenuates astrocyte pyroptosis by regulating the TLR4‑MyD88‑NF‑κB signaling axis, thereby promoting functional recovery in SCI rats. RNA‑seq combined with RT‑qPCR analysis revealed that Rg1 treatment significantly downregulated the transcriptional levels of Tlr4, Myd88, and Map3k7 in primary astrocytes, and reduced the expression and nuclear translocation of downstream NF‑κB pathway phosphoproteins (p‑IKKβ, p‑p65). Collectively, our data provide a molecular framework for the neuroprotective effects of Rg1 after SCI: Rg1 inhibits TLR4‑MyD88‑IKKβ/NF‑κB signaling, downregulates NLRP2 inflammasome‑mediated astrocyte pyroptosis, thereby alleviating neuroinflammation and promoting motor functional recovery. Importantly, the RNA‑seq analysis offers global molecular evidence identifying the key transcriptional pathway (TLR4/MyD88/NF‑κB) targeted by Rg1.

Losing the maternal effect gene Nlrp2 alters the ovulated mouse oocytes transcriptome and impacts histone demethylase KDM1B expression

Project description:We performed RNA sequencing on pools of oocytes after inducing ovarian stimulation in wild-type and Nlrp2-null female mice. We found 231 differentially expressed genes (DEGs) in Nlrp2-null COCs compared to WT COCs (123 up- and 108 downregulated; adjusted p<0.05). The DEGs we identified were enriched for processes involved in neurogenesis, gland morphogenesis, and protein metabolism and for post-translationally methylated proteins. When we compared our RNA sequencing results to an oocyte-specific reference transcriptome that contains many previously unannotated transcripts, we found 228 DEGs, including genes not identified with the first analysis. Intriguingly, 68% and 56% of DEGs from the first and second analyses, respectively, overlap with oocyte-specific hyper- and hypomethylated domains.

Proximal tubule cells in blood and urine have potential as biomarkers for kidney disease biopsies

Project description:Early diagnosis and treatment is pivotal to the management of kidney disease, whereas the pathological mechanisms and minimally invasive diagnostic method still need to be investigated. In the present study, single-cell RNA sequencing (scRNA-seq) was used to evaluate the heterogeneity of kidney diseases in single cell level. Cellular gene expression profiles of cells of renal tissue, peripheral blood mononuclear cells (PBMCs) and urine from four nephritis patients were performed. Our analysis revealed 12 subsets of renal cells and leukocytes, including fibroblast cells, mesangial cells, epithelial cells, proximal tubule cells (PTCs), and 6 types of immune cells, CD8+ T cell, macrophages (MC), nature killer cells (NK), dendritic cells (DC), B cell and neutrophils. PTCs were detected in both PBMC and urine, while PTC was negative in healthy blood sample. Multiple populations of fibroblast cells, mesangial cells and PTCs demonstrated pro-inflammatory or pro-apoptotic responses. Gene expression analysis suggested that chemotactic and activating effect of inflammatory PTCs and fibroblasts on neutropils were critical for the development and progress of nephritis, which was supported by the widely overexpressed pro-inflammatory genes in these cells. Gene expression profiles of inflammatory PTCs in PBMC, urine and kidney are highly correlated, indicating the high possibility of urine and PBMC PTCs in serving as a surrogate for kidney biopsies.
