RNA-Seq from zmknl1 and WT kernels at 14 DAP (day after pollination) with each two independent biological replicates

ABSTRACT: To further understand the basis of the phenotypic defects in the zmknl1 mutant, we performed RNA-Seq from zmknl1 and WT kernels at 14 DAP with each two independent biological replicates. Totally 555 significantly differentially expressed genes (DEGs) were identified according to these RNA-Seq data (q-value <0.05, log2(fold change) >= 1.5), with 317 down- and 238 up-regulated genes.

Project description:A customized targeted oligoarray was used to monitor the expression levels of 1000 genes, representative of the immature kernel transcriptome. Using this oligoarray we compared transcripts from 10 DAP kernels of two susceptible and two drought tolerant genotypes. These four genotypes were extracted from our RIL population (B73xH99) and grown under water stress and well watered field conditions. Keywords: Stress response Two-condition experiment: water stress field condition vs. well watered field condition. Transcriptional profiling of 10 DAP kernel (two susceptible and two drought tolerant genotypes) in the first condition vs 10 DAP kernel (two susceptible and two drought tolerant genotypes) in the second condition. Project description:At 35 DAP whole kernels (pericarp + endosperm + embryo) without glumes of green house grown ears of heterozygous (+/bt2-H2328), self-pollinated plants were visually divided into pools of phenotypically normal looking kernels (small indentation, slightly smaller than mutant kernels, genotype +/+ or +/bt2-H2328) and pools of phenotypically mutant kernels (plump, round kernels, slightly larger than normal kernels, genotype bt2-H2328/bt2-H2328). Developmental changes in transcriptomes of two key transport tissues in maize kernels: The Maternal Transfer Zone (MATZ) and Basal Endosperm Transfer Layer (BETL)

Project description:Transcriptome profiles of MATZ and BETL tissues are compared across three stages of development. Sugars and other nutrients unloaded from vascular tissues in the MATZ are imported by the BETL for utilization by the developing endosperm. Pronounced changes in gene expression occur in both tissues during kernel development. RNAseq data were obtained from duplicate tissue samples isolated by cryomicrodissection of developing maize kernels at three developmental time points; 8 days post-pollination (DAP), 14 DAP and 20 DAP.

Project description:Transcriptional response of Bacillus subtilis to daptomycin in wild-type and in a daptomycin resistant mutant. Bacillus subtilis 168, WT (-DAP) vs. DapR1 (-DAP), WT (+DAP) vs. DapR1 (+DAP), DapR1 (+DAP) vs. DapR1 (-DAP). Each experiment was conducted at least twice using two independent total RNA preparations. For daptomycin untreated comparison between 168 WT and DapR1 mutant, DapR1 was labeled with Alexa Fluor 647 and WT was labeled with Alexa Fluor 555. For daptomycin treated experiments between WT and DapR1, DapR1 was labeled with Alexa Fluor 647 and WT with Alexa Fluor 555. For treated vs. untreated DapR1, the DAP treated samples were labeled with Alexa Fluor 647 and the untreated with Alexa Fluor 555. For dye swap, untreated DapR1 was labeled with Alexa Fluor 647 and DAP treated with Alexa Fluor 555.

Transcriptional profiles uncover Aspergillus flavus induced resistance in mature maize kernels

Project description:Aflatoxin contamination caused by the opportunistic pathogen A. flavus is a major concern in maize production prior to harvest and during storage. Previous studies indicate that both constitutive and induced resistance are involved in maize kernel defense against A. flavus infection, little is known about molecular mechanisms of mature kernels in response to fungal infection. The purpose of this study is to determine gene expression differences in maize kernels between resistant and susceptible lines in response to A. flavus challenge. To avoid the environmental effects in the field inoculation, a laboratory based inoculation technique Kernel Screening Assay (KSA) was used to challenge kernels with A. flavus. After 72 hours incubation of inoculated and noninculated mature kernels, gene expression profiles of two Near Isogenic Lines (NIL) of Eyl25 (A. flavus resistant) and Eyl31 (A. flavus susceptible) were compared using oligonucleotide array.
