Escherichia coli exposed to biocides (antiseptics and disinfectants)

ABSTRACT: We compared the exposure of E. coli MG1655 to ten commonly used antiseptics and disinfectants for short (30min) and long (7-12h) term, looking for common and unique stress response elements.

Project description:Disinfection is very critical for the removal of pathogens that constitute public health concerns. However, a rounded understanding of the antimicrobial action of different classes of disinfectants is still lacking. In this work, the physiological and transcriptomic responses of Escherichia coli to two different disinfectants namely: potassium ferrate (K2FeO4) and more conventional disinfectant, sodium hypochlorite (NaOCl), were examined. The study aimed to examine E. coli genetic response to ferrate and compare and contrast it to the genetic response of E. coli to hypochlorite.

Using quantitative RNA-seq analysis, we compared the transcriptome of naïve MG1655 and the lysogens carrying the Stx2 phage of the 2011 E. coli O104:H4 outbreak strain or of the E. coli O157:H7 strain PA8, which share high degree of sequence similarity.

Project description:Our focus of this study is to determine the gene expression changes in E. coli colonizing the mouse intestine as compared to E. coli grown in minimal medium in vitro. We colonized CD-1 male mice with E. coli MG1655 StrR wild type and extracted the total RNA from mouse cecal mucus. We also extracted E. coli MG1655 StrR RNA from in vitro culture and sequenced and compared to determine the genes important for E. coli colonization of the mammalian intestine. Our results indicate that OmpC is important for E. coli to colonize the mouse intestine as it is upregulated as compared to in vitro culture. OmpF is not important; it is detrimental for E. coli colonization of the mouse intestine and is highly downregulated in the colonized E. coli

We also mapped the binding of the ß subunit of RNA Polymerase under both aerobic and anaerobic growth conditions. As a control, we also performed ChIP-chip on FNR in a ∆fnr mutant strain of Escherchia coli MG1655 K-12. We also examined FNR immunoprecipitation at various FNR concentrations using IPTG and Ptac::fnr (PK8263). The ∆hns/∆stpA strains were also used. Project description:The gene expression profile of E. coli K-12 MG1655 grown in minimal medium supplemented with elevated copper concentrations (as copper-glycine) has been analysed using whole genome oligonucleotide microarrays. “Pool” RNA was isolated from five independently grown 50ml DMA cultures of E. coli K-12 MG1655. “Test” RNA was isolated from three independently grown 50ml DMA cultures of E. coli K-12 MG1655, for each growth condition (no added CuSO4, or supplemented with either 0.75mM or 2mM copper glycine). A sample of each culture was taken at mid to late exponential growth phase at OD600 0.8 (measured using a Pharmacia Biotech Ultrospec 2000 in a cuvette with a path length of 10mm).
