Profiling of Granzyme B Secreting B cells

ABSTRACT: B cells with regulatory functions have been described both in immune homeostasis in healthy volunteers and in different pathological conditions. Whereas Il-10 secreting B cells represent the main subset described, recently, a subset of B cells secreting granzyme B (GZMB+) was evidenced in a growing numbers of immunological contexts, autoimmunity, chronic infections and neoplasias. Until now, no phenotype has been evidenced for these GZMB+ B cells and their regulatory functions as well are still to clarify.

Mast cells decrease efficacy of anti-angiogenic therapy by secreting matrix-degrading granzyme B

Project description:Resistance towards anti-angiogenic therapy (AAT) still represents a substantial clinical challenge. We report here that tumor-infiltrating mast cells (MC) are powerful mediators decreasing efficacy of AAT in mice and cancer patients. They act in a cell-extrinsic manner by secreting granzyme B, which liberates pro-angiogenic mediators from the extracellular matrix. In addition, MC also diminish efficacy of anti-angiogenic agents in a cell-autonomous way, which can be blocked by the mast cell degranulation inhibitor cromolyn. Our findings are relevant in humans because patients harboring higher numbers of MC in their tumors have an inferior outcome after anti-angiogenic treatment in the Gepar Quinto randomized Phase 3 clinical trial. Thus, MC-targeting might represent a novel promising approach to increase efficacy of AAT.

Fibroadipogenic Progenitor Regulates Neuromuscular Junction by Secreting Novel Protein Granzyme E

Project description:Muscle fibroadipogenic progenitor (FAP) cells, which are muscular mesenchymal cells that originate from lateral plate mesoderm have been proposed to act as a critical regulator for adult muscle homeostasis1–7, including the maturation and proper functioning of the neuromuscular junction (NMJ)3, Prx-Bap1 paper. However, the mechanism and intercellular crosstalk by which FAPs regulate the stability and functionality of neuromuscular system remains unknown. Here we show that FAPs not only locally but also systemically regulate the neuromuscular system through the secretion of a serine-type endopeptidase Granzyme E which may imply the previously unidentified endocrine function of FAPs. Local transplantation of wild-type FAPs into the neuromuscular disease model (Prrx1Cre;Bap1f/f, hereafter, cKO) can readily prevent neuromuscular defects, including degeneration of the neuromuscular junction and loss of motor neurons. These effects are found not only in transplanted hindlimb muscles but also in the contralateral hindlimb and even forelimb muscles. Notably, subcutaneous administration of microparticles encapsulating FAP-conditioned media into cKO mice was sufficient to restore normal neuromuscular functions. By analyzing the transcriptomic and secretomic profiles of FAPs, we identified a novel protein, Granzyme E, which is specifically expressed in and secreted by FAPs, and which indispensably regulates the structure and function of NMJ and motor neuron survival. Our study has defined a unique mechanism of Granzyme E-dependent, systemic control of the neuromuscular system by FAPs, which would provide a comprehensive understanding on the neuromuscular systems and their crosstalk with non-neuronal cells. These findings may provide a therapeutic benefit to treat NMJ-related diseases.

Project description:The therapeutic potential for human type 2 innate lymphoid cells (ILC2s) has been underexplored. While not observed in mouse ILC2s, we have found that human ILC2s secrete granzyme B (GZMB) and directly lyse tumor cells by inducing pyroptosis and/or apoptosis, which is governed by a DNAM-1CD112/CD155 interaction that inactivates FOXO1. Over time, the high surface density expression of CD155 in AML impairs expression of DNAM-1 and GZMB, thus allowing for immune evasion. We describe a reliable platform capable of up to 2,000-fold expansion of human ILC2s within 4 weeks, whose molecular and cellular ILC2 profile has been validated by single-cell RNA sequencing. In both AML and solid tumor models, exogenously administered expanded human ILC2s show significant antitumor effects in vivo. Collectively, we demonstrate previously unreported properties of human ILC2s and identify this innate immune cell subset as a new member of the cytolytic immune effector cell family.

A novel and divergent role of granzyme A and B in resistance to helminth infection

Project description:Granzyme (gzm) A and B, proteases of NK cells and T killer cells, mediate cell death, but also cleave extracellular matrices, inactivate intracellular pathogens and induce cytokines. Moreover, macrophages, T helper 2 cells, regulatory T cells, mast cells and B cells can express gzms. We recently reported gzm induction in human filarial infection. Here we show that in rodent filarial infection with Litomosoides sigmodontis worm loads were significantly reduced in gzmAxB and gzmB knock out (ko) mice, but enhanced in gzmA ko compared to wildtype (wt) mice. GzmA/B deficiency was associated with a defense-promoting Th2 cytokine and antibody shift and enhanced early inflammation gene expression, whereas gzmA deficiency was linked with alternatively activated macrophages and reduced inflammation. This suggests a novel and divergent role for gzms in helminth infection with gzmA contributing to resistance and gzmB promoting susceptibility. Keywords: knock out vs wild type
