Project description:Actin cytoskeleton has multiple functions in cancer cell invasion and metastasis; yet how these functions are coordinated remains unclear. In the cytoplasm, actin is necessary for the formation of specialized membrane protrusions, termed invadopodia, that concentrate and secrete extracellular matrix-degrading enzymes, particularly the matrix metalloproteinases (MMPs), facilitating tumor cell invasion. We recently identified cysteine- and glycine-rich protein 2 (CRP2) as an actin-crosslinking protein whose activity is required for the assembly and elongation of the invadopodium actin backbone. Here, we report that CRP2 is also located in the nucleus of breast cancer cells to induce a complementary pro-metastasis gene expression program mostly related to breast cancer cell invasion and metastasis. We showed in this present study that CRP2 interacts with the transcription factors SRF and GATA3 to regulated the expression of MMP-9 and MMP-13 respectively. Overall, our data indicate that CRP2 act a key mediator of tumor cell invasion.

Project description:The process of tumor invasion requires degradation of extracellular matrix by proteolytic enzymes. Cancer cells form protrusive invadopodia, which produce and release matrix metalloproteinases (MMPs) to degrade the basement membrane and thereby allowing metastasis. We investigated the effect of LASP1, a newly identified protein in invadopodia, on expression, secretion and activation of MMPs in invasive breast tumor cells. By analyzing and verifying microarray data sets and performing reporter assays, we demonstrate that LASP1 positively modulates the expression of c-Fos and AP-1 activity, and thereby up-regulates expression of MMP1, -3 and -9 in breast cancer cells. Furthermore, zymography assays and Western blot analysis revealed cytosolic LASP1 promotion of MMP vesicle secretion into the extracellular matrix, thus altering the microenvironment during cancer progression. The newly identified role of LASP1 in regulating matrix degradation by affecting MMP transcription and secretion elucidated the migratory potential observed in studies that investigated the upregulation of LASP1 in aggressive cancers.

Project description:Invadopodia are actin-based proteolytic membrane protrusions required for invasive behavior and tumor growth. In this study, we used our high-content screening assay to identify kinases whose activity impacts invadopodia formation. Among the top hits selected for further analysis was TAO3, a STE20-like kinase of the GCK subfamily, for further analysis. TAO3 was overexpressed in many human cancers, and regulated invadopodia formation in melanoma, breast and bladder cancers. Furthermore, TAO3 catalytic activity facilitated melanoma growth in 3-dimensional matrices and in vivo. A novel, potent catalytic inhibitor of TAO3 was developed that inhibited invadopodia formation and function, and tumor cell extravasation, and growth. Treatment with these inhibitors demonstrated that TAO3 activity is required for endosomal trafficking of TKS5-alpha, an obligate invadopodia scaffold protein. A phosphoproteomics screen for TAO3 substrates revealed the dynein subunit protein LIC2 as a relevant substrate. Knockdown of LIC2 or expression of a phosphomimetic form promoted invadopodia formation. Thus, TAO3 is a new therapeutic target with a distinct mechanism of action.

Project description:Invadopodia are adhesive, actin - rich pro trusions , formed by metastatic cancer cells , that degrade the extracellular matrix and facilitate invasion . They support the metastatic cascade by a spatially and temporally coordinated process where by invading cells bind to the matrix , degrade it by speci fic metalloproteinases , and mechanically penetrate diverse tissue ba rriers by forming actin - rich extensions . However, despite the apparent involvement of invadopodia in the metasta t i c process , the molecular mechanisms that regulate invadopodia formation an d function are still largely unclear. In this study, we have explored the involvement of the key Hippo pathway co - regulators , namely YAP , and TAZ , in invadopodia formation and matrix degradation. Towards that goal, we tested the effect of depletion of YAP, TAZ , or both on invadopodia formation and activity in multiple human cancer cell lines. We report that knockdown of YAP and TAZ or their inh ibit ion by verteporfin indu ce a significant elevation in matrix degradation and invadopodia formation in several ca ncer cell lines . Conversely , o verexpression of these proteins strongly suppress es invadopodia formation and matrix degradation . Proteomic and transcriptomic profiling of MDA - MB - 231 cells , following co - knockdown of YAP and TAZ , revealed a significant change in the levels of key invadopodia - associated proteins, including the crucial proteins Tks5 and MT1 - MMP (MMP14). Collectively, our findings show that YAP and TAZ act as negative regulators of invadopodia formation in diverse cancer lines, most likely by red ucing the levels of essential invadopodia components. Dissecting the molecular mechanisms of invadopodia formation in cancer invasion may eventually reveal novel targets for therapeutic applications against invasive cancer

Project description:The therapeutic efficacy of radiotherapy and chemotherapy with temozolomide (TMZ) for glioblastoma (GBM) is currently limited by the augmented invasive abilities of GBM cells that survive treatment, however the underlying mechanisms remain poorly understood. Due to their ability of transporting oncogenic material between cells, small extracellular vesicles (sEVs) have emerged as key mediators of tumour progression and resistance to treatment. Here, we report that sEVs secreted by GBM cells may facilitate tumour invasion through their ability to promote the formation and activity of actin-rich protrusions known as invadopodia, and that this may be further enhanced following exposure to radio-chemotherapeutic treatment. Furthermore, proteomic analysis of GBM cells exposed to this treatment revealed an increased abundance of invasion-related proteins (including adhesion, cytoskeletal, and membrane trafficking proteins), whilst sEVs were instead enriched in invadopodia-related signalling cargo proteins. Collectively, this work highlights the contributing role of invadopodia and sEVs to the pro-invasive abilities of GBM cells that survive radio-chemotherapeutic treatment.

Project description:Deregulated cell migration and invasion, which are hallmarks of metastatic cancer cells, are correlated to structural and functional alterations of the actin cytoskeleton associated to a large panel of actin-binding proteins. Among these, the actin-bundling protein L-plastin, initially detected in leukocytes, is ectopically expressed in several solid tumours and is often considered as a metastatic marker. Phosphorylation of L-plastin on residue serine 5 (Ser5) has been shown to activate L-plastin and to be crucial for invasion and metastasis formation. Here, we investigate the signalling pathway leading to L-plastin Ser5 phosphorylation in four breast cancer cell lines. Whole genome microarray analysis comparing cell lines with different invasive capacities and corresponding L-plastin Ser5 phosphorylation levels revealed that genes of the MAPK/ERK pathway are differentially expressed. In line, in vitro kinase assays showed that MAPK/ERK pathway downstream kinases RSK1 and RSK2 are able to directly phosphorylate L-plastin on Ser5. In parallel to a knockdown approach, activation and inhibition studies of signalling molecules of the MAPK/ERK pathway, followed by computational modelling analysis, confirmed that RSK is an important activator of L-plastin in all four studied cell lines. Finally and rounding up our study, RSK knockdown significantly impaired migratory and invasive capacities of a selected invasive cell line, thus consolidating RSK as a promising therapeutic target in certain invasive carcinomas. Altogether, our data provide substantial evidence that the MAPK/ERK pathway is involved in L-plastin Ser5 phosphorylation in breast cancer cells with its downstream kinases RSK1 and RSK2 being able to directly phosphorylate L-plastin on Ser5.

Project description:In a variety of organisms, including mammals, caloric restriction improves metabolic status and lowers the incidence of chronic-degenerative diseases, ultimately leading to increased lifespan. Here we show that knockout mice for Eps8, a regulator of actin dynamics, display reduced bodyweight, partial resistance to age- or diet-induced obesity, and overall improved metabolic status. We present evidence that these phenotypes, which are associated to increased lifespan in the Eps8 null mice, are due to caloric restriction. This, in turn, is caused by reduced intestinal fat absorption, due to altered morphogenesis of microvilli in intestinal enterocytes. In the nematode, genetic removal of Eps8, causes a microvillar phenotype, indistinguishable from that observed in mice, which leads to early larval lethality. By exploiting the nematode model system, we demonstrate here that the actin bundling activity of Eps8 is indispensable for viability and proper intestinal morphogenesis. This result links a precise molecular function of Eps8 to proper microvillar morphogenesis, and therefore to the phenotype of Eps8-null mice. Project description:Cell-cell fusion is a frequent and essential event during development, and its dysregulation causes diseases ranging from infertility to muscle weakness. Fusing cells repeatedly need to remodel their plasma membrane by orchestrated formation and disassembly of cortical actin filaments, but how the dynamic reorganization of the actin cytoskeleton control is still poorly understood. Here, we identified a ubiquitin- dependent toggle switch that establishes reversible actin bundling during mammalian cell fusion. We found that EPS8-IRSp53 complexes stabilize cortical actin bundles at sites of cell contact, which in part pushes cells towards each other. Conversely, EPS8 monoubiquitylation by CUL3KCTD10 displaces EPS8-IRSp53 from membranes and counteracts actin bundling, a dual activity that allows apposed cells to progress with fusion. We conclude that cytoskeletal rearrangements during development are precisely controlled by ubiquitylation, raising the possibility to modulate the efficiency of cell-cell fusion for therapeutic benefit.

Project description:The mechanisms underlying cancer metastasis remain poorly understood. Here, we report that TFAM deficiency rapidly and stably induced spontaneous lung metastasis in mice with liver cancer. Interestingly, unexpected polymerization of nuclear actin was observed in TFAM-knockdown HCC cells when cytoskeleton was examined. Polymerization of nuclear actin is causally linked to the high-metastatic ability of HCC cells by modulating chromatin accessibility and coordinating the expression of genes associated with extracellular matrix remodeling, angiogenesis, and cell migration. Mechanistically, TFAM deficiency blocked the TCA cycle and increased the intracellular malonyl-CoA levels. Malonylation of mDia2, which drives actin assembly, promotes its nuclear translocation. Importantly, inhibition of malonyl-CoA production or nuclear actin polymerization significantly impeded the spread of HCC cells in mice. Moreover, TFAM was significantly downregulated in metastatic HCC tissues and was associated with overall survival and time to tumor recurrence of HCC patients. Taken together, our study connects mitochondria to the metastasis of human cancer via uncovered mitochondria-to-nucleus retrograde signaling, indicating that TFAM may serve as an effective target to block HCC metastasis.
