Project description:High throughput sequencing technology was used to sequence the transcriptome of breast muscle tissue of control group and test group IV, the results of bioinformatics analysis showed that 417 mRNAs were differentially expressed between control group and test group IV (fold change ≥ 2.0, P < 0.05). Among them, 160 mRNAs presented upregulated and 257 were downregulated. The differentially expressed mRNAs were mainly involved in Melanogenesis, Calcium signaling pathway, Wnt signaling pathway, mTOR signaling pathway, and Vascular smooth muscle contraction.

Transcriptome sequencing and bioinformatics analysis of gastrocnemius muscle in type 2 diabetes mellitus rats

Project description:Objective Type 2 diabetes mellitus (T2DM) is one of the high risk factors for sarcopenia. However, the pathogenesis of diabetic sarcopenia has not been fully elucidated. This study obtained transcriptome profiles of gastrocnemius muscle in normal and T2DM rats based on high-throughput sequencing technology, which may provide new ideas for exploring the pathogenesis of diabetic sarcopenia. Methods Twelve adult male Sprague-Dawley rats were randomly divided into Control group and T2DM group, and gastrocnemius muscle tissue was retained for transcriptome sequencing and real-time quantitative polymerase chain reaction (qRT-PCR) 6 months later. Screening differentially expressed genes (DEGs), Cluster analysis, gene ontology (GO) functional annotation analysis and Kyoto Encyclopedia of Genes and Gnomes (KEGG) functional annotation and enrichment analysis were performed for DEGs. Six DEGs related to apoptosis were selected for qTR-PCR verification.

Transcriptome profile analysis of breast muscle tissues from high or low levels of atmospheric ammonia exposed broilers (Gallus gallus)

Project description:Abstract: Atmospheric ammonia is a common problem in poultry industry. High concentrations of aerial ammonia cause great harm to broilers' health and production. For the consideration of human health, the limit exposure concentration of ammonia in houses is set at 25 ppm. Previous reports have shown that 25 ppm is still detrimental to livestock, especially the gastrointestinal tract and respiratory tract, but the negative relationship between ammonia exposure and the tissue of breast muscle of broilers is still unknown. In the present study, 25 ppm ammonia in poultry houses was found to lower slaughter performance and breast yield. Then, high-throughput RNA sequencing was utilized to identify differentially expressed genes in breast muscle of broiler chickens exposed to high (25 ppm) or low (3 ppm) levels of atmospheric ammonia. The transcriptome analysis showed that 163 genes (fold change ≥ 2 or ≤ 0.5; P-value < 0.05) were differentially expressed between Ammonia25 (treatment group) and Ammonia3 (control group), including 96 down-regulated and 67 up-regulated genes. qRT-PCR analysis validated the transcriptomic results of RNA sequencing. Gene Ontology (GO) functional annotation analysis revealed potential genes, processes and pathways with putative involvement in growth and development inhibition of breast muscle in broilers caused by aerial ammonia exposure. This study facilitates understanding of the genetic architecture of the chicken breast muscle transcriptome, and has identified candidate genes for breast muscle response to atmospheric ammonia exposure.

Expression profiles of microRNAs in fetus and lamb muscle tissue.

Project description:We report the application of single-molecule-based sequencing technology for high-throughput profiling of miRNAs in goat muscle tissue.We identified 1120 miRNAs in fetus and lamb muscle tissue; 895 were known miRNAs and 313 were novel miRNAs. According to analysis, 192 miRNAs were up-regulated in fetus samples compared to lamb samples, while 121 miRNAs were down-regulated.To confirmed stage-specific differences in the abundance of certain miRNAs, we focused on the most up-regulated miRNA and found to be predominantly expressed in lamb muscle tissue, suggesting a potential role in muscle development.It has become a molecular marker for breeding programs of mutton production.

Anti-aging effect of olmesartan on miRNA expression profile in smooth muscle cells

Project description:High-throughput sequencing technique was used to screen the miRNAs which has potential role in the aging delaying effect of olmesartan on human smooth muscle cells. We cultured human vascular smooth muscle cells and used the third to fifth generation cells as unaging cells (Zhengchang-1,-2,-3), natural culture to passage 10 to 15 as aging group samples (Bo-1,-2,-3), and cells treated with olmesartan for the same time as aging group cells as treatment group (bo-ao-1,-2,-3). After collecting cells of each group, RNA was extracted and reversed. After the quality inspection was qualified, miRNA sequencing was carried out. Finally, we confirmed that the miRNA expression profile of human vascular smooth muscle cells changed after olmesartan treatment. This study confirmed that miRNA plays a potential role in the aging process of human smooth muscle cells and that olmesartan may play a role in delaying aging by regulating miRNA expression profile.
