Project description:Arabidopsis is a host to the fungal powdery mildew pathogen, Erysiphe cichoracearum, and a nonhost to Blumeria graminis f.sp. hordei, the powdery mildew pathogenic on barley. A screen for mutants that allowed increased entry by this inappropriate or nonhost pathogen on Arabidopsis led to the identification of PEN3. While pen3 mutants permitted both increased penetration and increased hyphal growth by B. g. hordei, they were unexpectedly resistant to E. cichoracearum. This resistance was correlated with the appearance of chlorotic patches and was salicylic acid-dependent. Consistent with this observation, microarray analysis revealed that the salicylic acid defense pathway was hyper-induced in pen3 relative to wild type following inoculation with either E. cichoracearum or B. g. hordei. The pen3 phenotypes result from a loss of function of AtPDR8, a ubiquitously and highly expressed ATP binding cassette transporter. PEN3 protein tagged with green fluorescent protein localized to the plasma membrane in uninfected cells. In infected leaves, the protein concentrated to high levels at infection sites and surrounded fungal penetration pegs. We hypothesize that PEN3 may be involved in exporting toxic substrates to sites of infection and that accumulation of these substrates intracellularly in the pen3 mutant may secondarily activate the salicylic acid pathway. Experiment Overall Design: Three week-old wild-type Col and mutant pen3 Arabidopsis thaliana plants were inoculated with Erysiphe cichoracearum, Blumeria graminis hordei, or not inoculated. 1 day post inoculation 16 rosettes were harvested per replicate. 4 replicates were perfomerd per treatment.

Project description:Arabidopsis does not support the growth and asexual reproduction of the barley pathogen, Blumeria graminis f. sp. hordei Bgh). A majority of germlings fail to penetrate the epidermal cell wall and papillae. To gain additional insight into this interaction, we determined whether the salicylic acid (SA) or jasmonate (JA)/ethylene (ET) defence pathways played a role in blocking barley powdery mildew infections. Only the eds1 mutant and NahG transgenics supported a modest increase in penetration success by the barley powdery mildew. We also compared the global gene expression patterns of Arabidopsis inoculated with the non-host barley powdery mildew to those inoculated with a virulent, host powdery mildew, Erysiphe cichoracearum. Genes repressed by inoculations with non-host and host powdery mildews relative to non-inoculated control plants accounted for two-thirds of the differentially expressed genes. A majority of these genes encoded components of photosynthesis and general metabolism. Consistent with this observation, Arabidopsis growth was inhibited following inoculation with Bgh, suggesting a shift in resource allocation from growth to defence. A number of defence-associated genes were induced during both interactions. These genes likely are components of basal defence responses, which do not effectively block host powdery mildew infections. In addition, genes encoding defensins, anti-microbial peptides whose expression is under the control of the JA/ET signalling pathway, were induced exclusively by non-host pathogens. Ectopic activation of JA/ET signalling protected Arabidopsis against two biotrophic host pathogens. Taken together, these data suggest that biotrophic host pathogens must either suppress or fail to elicit the JA/ET signal transduction pathway.

Project description:Attack by the host powdery mildew Erysiphe cichoracearum usually results in successful penetration and rapid proliferation of the fungus on Arabidopsis. By contrast, the nonhost barley powdery mildew Blumeria graminis f. sp. hordei (Bgh) typically fails to penetrate Arabidopsis epidermal cells. In both instances the plant secretes cell wall appositions or papillae beneath the penetration peg of the fungus. Genetic screens for mutations that result in increased penetration of Bgh on Arabidopsis have recently identified the PEN1 syntaxin. Here we examine the role of PEN1 and of its closest homologue, SYP122, identified as a syntaxin whose expression is responsive to infection. pen1 syp122 double mutants are both dwarfed and necrotic, suggesting that the two syntaxins have overlapping functions. Although syp122-1 and the cell wall mur mutants have considerably more pronounced primary cell wall defects than pen1 mutants, these have relatively subtle or no effects on penetration resistance. Upon fungal attack, PEN1 appears to be actively recruited to papillae, and there is a 2-h delay in papillae formation in the pen1-1 mutant. We conclude that SYP122 may have a general function in secretion, including a role in cell wall deposition. By contrast, PEN1 appears to have a basal function in secretion and a specialized defense-related function, being required for the polarized secretion events that give rise to papilla formation.

Project description:Powdery mildew caused by Erysiphe cruciferarum, is an epidemic of oil rapeseed (Brassica napus) growing worldwide, but resistant germplasm is rare in this species. We obtained the hybrid seeds of distant hybridization between powdery-mildew-immune Brassica carinata cultivar ‘White flower’ and susceptible B. napus cultivar ‘Zhongshuang11’. Five lines in the BC1F3 generation (F3 after backcross to 'Zhongshuang11') were identified to be resistant or moderately resistant. In order to identify the important biological responses to powdery mildew, the foliar transcriptomes of the resistant and susceptible plants in these progenies after powdery mildew inoculation were compared by using Illumina RNA-seq. We identified 10,454 differential expression genes (DEGs) and 1050 genes out of them are related to disease resistance. There were 271 DEGs in Group Resistance expressed at least two fold higher than in group S, while 779 DGEs expressed two fold lower. The genes highly expressed in Group Resistance are those encoding the proteins: (1) related to wax, chloroplast and cell wall metabolism, such as KCS6, CSP41B, RWA, callose synthetase 3, pectinase 9, fructosidase 2, 9s-lipoxygenase LOX2, etc.; (2) kinases including RKL, ERECTA, BAK1, BAM2, LysM receptor like kinase, and lipid transfer protein kinase ERl1 and ERl2; (3) broad spectrum powdery mildew resistance proteins RPW8, calmodulin MLO2, PMR5, MLP328, EDR2, RPS4 and RPS6, etc. In group susceptible, pectinesterase, cytochrome CYP81f2, LOX1, cysteine rich receptor protein kinases and serine / threonine protein kinases such as MEKK, RLK6, CRK45, APK1, BRl3, WAK1, WAK10, etc., and TIR-NB-LRR receptor like proteins R1M1, DSC1, DSC2 and pathogenesis-related protein PR-1 etc. were the most activated genes. The results provide the preliminarily knowledge about molecular mechanism in rapeseed defense response to powdery mildew.
