We observed that caspase-4 targeting dampens the mRNA expression induction of critical senescence-associated secretory phenotype factors while increasing the transcription of E2F target genes and genes related to the G2/M checkpoint.

Project description:In the present study, we analyse the effect of knocking down TLR2 and TLR10 during oncogene induced senescence in IMR90 cells expressing a inducible version of oncogenic RAS (ER:RAS) in the transcriptome using Ampliseq RNA sequencig. We observed that TLR2 and TLR10 regulate the expression of many pro-inflammatory cytokines and chemokines that constitute the senescence associated secretory phenotype. There is also significant regulation of genes of the acute phase response.

Project description:We compared the profile of miRNAs expressed in HEK293 and MRC5 cells that overexpressed KRASG12V to those expressed in parental cells that harbored wild-type KRAS. The results indicated that a subset of miRNAs was significantly down-regulated in KRASG12V transfected two cells. To address the functional relevance of miRNAs in KRAS mutant cancers, we transfected exogenous KRASG12V into HEK293 and MRC5 cells with wild type KRAS genes, and we comprehensively profiled the dysregulated miRNAs.

HepaCAM knockdown impairs learning and memory in mice by regulating cholesterol synthesis pathway

Project description:Astrocytes engage in metabolic interactions with nearby neurons by supplying various substances. However, how they regulate learning and memory through changes in the metabolic state of neurons is still unclear. In this study, we show that knocking down astrocytic hepaCAM silences genes related to cholesterol biosynthesis in astrocytes, leading to a decrease in cholesterol levels. Additionally, astrocytes play a crucial role in synapse development and function by releasing signals that guide neurons. Consequently, without enough cholesterol, the level of neuronal synaptic proteins and the density of dendritic spines decrease. Then we demonstrate that knocking down hepaCAM leads to impaired learning and memory in mice. Collectively, hepaCAM, a cell adhesion molecules specifically expressed in astrocytes, regulates learning and memory by regulating the cholesterol synthesis pathway.

Senescence-induced vascular remodeling creates therapeutic vulnerabilities in pancreas cancer

Project description:KRAS mutant pancreatic ductal adenocarcinoma (PDAC) is characterized by a desmoplastic response that promotes hypovascularity, immunosuppression, and resistance to chemo- and immunotherapies. We show that a combination of MEK and CDK4/6 inhibitors that target KRAS-directed oncogenic signaling can suppress PDAC proliferation through induction of retinoblastoma (RB) protein-mediated senescence. In preclinical mouse models of PDAC, this senescence-inducing therapy produces a senescence-associated secretory phenotype (SASP) that includes pro-angiogenic factors that promote tumor vascularization, which in turn enhances drug delivery and efficacy of cytotoxic gemcitabine chemotherapy. In addition, SASP-mediated endothelial cell activation stimulates the accumulation of CD8+ T cells into otherwise immunologically “cold” tumors, sensitizing tumors to PD-1 checkpoint blockade. Therefore, in PDAC models, therapy-induced senescence can establish emergent susceptibilities to otherwise ineffective chemo- and immunotherapies through SASP-dependent effects on the tumor vasculature and immune system.
