Project description:Perfluorobutanoic acid (PFBA) is a short-chain polyfluoroalkyl substance (PFAS) that is measurable in fish tissues and environmental matrices. Previous studies suggest that there are adverse effects on animal and human health following exposure to PFBA, however the mechanisms underlying the toxicity are not well characterized. This study measured biological responses (developmental, transcriptome, and behavioral responses) of zebrafish (wild-type AB strain, Danio rerio) following exposure to a range of PFBA concentrations (0.1-100 µl/L). To further elucidate putative mechanisms related to locomotor abnormalities by PFBA, RNA-seq was conducted. These data shed new mechanistic light onto the sublethal effects of lesser studied short chain perfluorinated chemicals.

Project description:Perfluoroalkyl substances (PFAS) are a class of synthetic chemicals that are persistent in the environment. Due to concerns linked with longer chain PFAS, shorter chain chemicals were used as replacements, but developmental toxicity assessments of the shorter chain chemicals are limited. The aim of this study was to compare the developmental toxicity of three perfluoroalkyl acids (PFAAs): perfluorooctanoic acid (PFOA), composed of 8 carbon (C8), perfluorohexanoic acid (PFHxA, C6), and perfluorobutanoic acid (PFBA, C4) using the zebrafish (Danio rerio). LC50s at 120 hour post fertilization (hpf) were determined to assess the potency of each PFAA by exposing developing zebrafish (1-120 hpf) to a range of concentrations. In sublethal assessments, zebrafish were exposed to 0, 4, 40, or 400 parts per billion (ppb; µg/L) of the PFAAs throughout embryonic development (1-72 hpf). Effects of the embryonic exposure on locomotor activities was completed with the visual motor response test at 120 hpf. At 72 hpf, morphological changes including total body length, head length, and head width were assessed. Additionally, transcriptomic profiles of PFOA, PFHxA, and PFBA were determined at 72 hpf to compare altered molecular and disease pathways. The LC50 ranking was PFOA > PFHxA > PFBA, which followed trend as expected based on chain length. PFOA caused hyperactivity and PFBA hypoactivity, while PFHxA did not change behavior. PFOA, PFHxA, and PFBA caused morphological and transcriptomic alterations that were unique for each chemical and were concentration dependent. Cancer was a top enriched disease for PFOA, while FXR/RXR activation was a top canonical pathway in all PFBA exposures. Overall, an embryonic exposure to PFOA, PFHxA, or PFBA resulted in morphological alterations in zebrafish eleuthero-embryos; however, only PFOA and PFBA induced neurobehavioral alterations. The transcriptomic profile of each chemical was unique indicating different toxicity mechanisms and were aligned with human adverse health outcomes supporting predictive value.

Project description:This study investigated the toxicological effects of two short-chain per- and polyfluoroalkyl substances (PFAS), including perfluorobutanoic acid (PFBA) and perfluorohexanoic acid (PFHxA), on the transcriptome of human nasal RPMI2650 cells. Gene annotation enrichment analysis highlighted neurofactor, synaptic, solute carrier, and ribosome signaling pathways in common pathways of widespread transcriptional changes in RPMI2650 cells. Systematic analysis of RNA-seq data revealed that PFAS-treated RPMI2650 cells are encoded to participate in cell proliferation (H3C14), nerve regeneration (L1CAM, P2RY1), and solute transport (SLC17A7, LRRK2). TOP 10 GO analysis showed that DEGs was mainly concentrated in synaptic regulation, cell division correlation and solute carrier transport. For example, in the PFBA treatment group, typical terms include synaptic vesicular membrane (GO:0030672), synaptic transmission, glutaminergic energy (GO:0035249), and interneuronal synaptic transmission (GO:0007270), while in the PFHxA treatment group, DEGs is mainly enriched in nucleosomes (GO:0000786). Microtubule binding (GO:0008017) and neuronal differentiation in the central nervous system (GO:0021953). In KEGG enrichment pathway, PFBA-treated differential genes were mainly concentrated in synaptic related pathways, such as serotonergic synapses (hsa04726), glycerophospholipid metabolism (hsa00564), and GABAergic synapses (hsa04727). In addition to neuron-related pathways, PFHxA processing enrichedneurotransmitter related pathways such as sphingolipid biosynthesis-Globo and isoglobo series (hsa00603), GABaergic synapses (hsa04727), and neuroactive ligand-receptor interactions (hsa04080).

Project description:Streptococcus Pneumoniae capsule is a major virulence factor. The capsule is known to inhibit complement fixation and phagocytes, but its impact on innate immune responses is not known. To address this question, we compared transcriptional responses by human monocyte derived macrophages in responses to wild type S. pneumoniae with isogenic mutant bacteria lacking their capsule. We conducted these experiments with and without macrophage pretreatment with cytochalasin D to evaluate the impact of phagocytosis, and we also made comparisons to mutant S.pneumoniae lacking bacterial lipoprotein which is known to be necessary for maximal innate immune responses.

However the diverse array of phagocytes residing within different tissues combined with the necessarily prompt nature of apoptotic cell clearance has made it difficult to study this process in situ. Thus, the full spectrum of functions executed by tissue resident phagocytes in response to homeostatic apoptosis remains unclear. We used microarrays to characterize the transcriptome within murine intestinal dendritic cells and macrophages both before and after in situ phagocytosis of apoptotic intestinal epithelial cells.

Project description:Invertebrates lack the adaptive immune system homologous to that of vertebrates, thus depend exceptionally on the innate immune system for the pathogen recognition and clearance. Phagocytes play a pivotal role in the innate immunity of invertebrates, but the molecular basis underlying their phagocytic killing is still far from well understood. In the present study, phagocytes from the Pacific oyster Crassostrea gigas were efficiently sorted based on the phagocytosis of FITC-labeled latex beads by fluorescence-activated cell sorting. The differentially expressed proteins were revealed by quantitative proteome and validated by qRT-PCR and flow cytometry.

Project description:Per- and polyfluoroalkyl substances (PFAS) are a group of organic chemicals that contain multiple carbon-fluorine bonds, which make them highly stable and widespread in our environment, even in the human body. They have been detected in cord blood and showed correlation with neonatal health abnormalities such as weight loss. Here, we utilized human embryonic stem cell-based model to assess the early developmental toxicity of six PFAS (PFOA, PFHxA, PFBA, PFOS, PFHxS, and PFBS) and their skin developmental toxicity. We employed global monolayer differentiated model to mimic the early embryonic development and performed non-neural ectoderm (NNE) differentiation which can give rise to keratinocytes to imitate early skin development. Transcriptomic analysis revealed that PFOS caused the most differentially expressed genes (DEGs) both in day 16 global monolayer samples and NNE samples to making it the most toxic of the six PFAS. Further validation of gene expression levels showed that the TGF-β signaling pathways were involved in the early global developmental toxicity and in the early skin developmental toxicity of PFOS. Interestingly, our results showed that PFOS inhibited the ciliogenesis of NNE cells, which may interfere with the transduction of TGF-β, Notch, Hedgehog, and Hippo signaling pathways, ultimately inhibiting early embryonic skin development and posing a potential threat to the occurrence of congenital skin diseases. Overall, our study suggests a potential toxicity mechanism by which PFAS inhibit ciliogenesis to interfere with the transduction of important signaling pathways, leading to early global developmental toxicity as well as early embryonic skin developmental toxicity and potential congenital skin diseases.
