Gene expression profiling of Trop2+ and Trop2- luminal epithelial cells and stromal cells from young adult and old mouse prostates

ABSTRACT: We set out to determine if distinct prostate luminal epithelial cell-types exhibited age-related differences in their gene expression profiles. We used fluorescence activated cell sorting to isolate Trop2+ and Trop2- luminal cells and EpCAM- CD49f- stromal cells from dissociated preparations of prostate tissue taken from young adult (3 months) and old (24 months of age) mice. RNA was extracted from sorted cells to evaluate differences in gene expression.

Project description:We set out to determine if prostate epithelial cell-types exhibited age-related differences in their gene expression profiles. We used fluorescence activated cell sorting to isolate basal and luminal cells from dissociated preparations of prostate tissue taken from young adult (3 months) and old (24 months of age) mice. RNA was extracted from sorted cells to evaluate differences in gene expression. We found significant enrichment for progenitor genes in old luminal cells including an immune/inflammatory profile that overlapped with a previously identified CD38-lo human prostate luminal progenitor signature. This study demonstrates a luminal progenitor signature in old mouse prostate.

Project description:To investigate the molecular reprogramming of epithelial cells of the prostate during basal to luminal differentiation in vivo, we took advantage of the spatial restriction of multipotent basal cells at the distal region (tip - 100μm) of the developing prostate at postnatal day (P)10 (Tika et al., Development, 2019). The distal tips (100μm) of P10-P13 prostate glands were manually dissected from the main ducts under the stereoscope in order to enrich for multipotent basal cells and basal-derived luminal cells. Cell populations were isolated for RNA sequencing based on EpCAM and Cd49f expression via FACS.

Gene expression profiles of normal human mammary cell subpopulations FACS sorted based on expression of CD49f and EpCAM

Project description:To delineate epithelial subpopulations in human mammary tissue, hematopoietic and endothelial cells were depleted from freshly isolated cell suspensions derived from reduction mammoplasties by fluorescence-activated cell sorting. The resultant Lin- population was fractionated into four distinct subpopulations using CD49f (α6-integrin) and epithelial cell adhesion molecule (EpCAM; also referred to as CD326 and ESA). Based on the immunohistochemical phenotype, and in vivo and in vitro functional assays, these subpopulations were identified as fibroblast-enriched stromal (CD49f -EpCAM-), mammary stem cell (MaSC)-enriched (CD49f hiEpCAM-), luminal progenitor (CD49f +EpCAM+), and mature luminal (CD49f –EpCAM+) cell subpopulations. Microarray profiling was used to derive gene expression signatures representative of these subpopulations using freshly sorted cells (>90% purity) from normal breast tissue. The four mammary cell subpopulations were found to have distinct gene expression profiles.

Gene expression profiles of epithelial and mesenchymal cell populations extracted from normal human breast tissue

Project description:Stromal-epithelial interactions play a fundamental role in tissue homeostasis, controlling cell proliferation and differentiation. The goals and objectives of this study were 1.) To generate gene expression profiles and identify signatures of freshly isolated, non-cultured, luminal epithelial or basal epithelial and CD10 positive or negative stromal cell populations from the human breast. 2.) Use the generated signatures to characterize and validate the molecular identity of human primary epithelial and stromal/mesenchymal breast cells maintained long-term in novel ex vivo culture conditions in serum free medium (previous study in our lab) Breast tissue from three different reduction mammoplasties was dissociated and single cells were labeled with antibodies against lineage markers (CD31, CD45 CD235ab), CD10, EpCAM, CD49f and DAPI. Different cell populations were identified, separated and sorted with a fluorescence activated cell sorter. Gene expression profiling of luminal epithelial (EpCAM+, CD49f+/-), basal epithelial (EPCAM-, CD49f+), CD10 negative and CD10 positive stromal cells was performed and distinct gene expression signatures for the four cell populations were identified.

Project description:Chromosomal rearrangements involving ETS factors, ERG and ETV1, occur frequently in prostate cancer. We here examine mouse prostate cells from WT mice with s with T-ETV1 mice, which contains express the truncated human ETV1 under the endogenous Tmprss2 promoter. ETV1 expression can be tracked by GFP expression. Primary prostate cells were isolated and luminal prostate cells were FACS-sorted by CD49F+ Sca1- from the lineage (CD31, CD45, Ter119) negative fraction. Total RNA was extracted from three biological replicates, and amplified using NuGEN V2cDNA amplification kit. This was used to hybridize to Affymetrix expression arrays using the Mouse Genome 430 2.0 platform.
