Project description:MiR-30e represses the osteogenic program in mesenchymal stem cells (MSCs) and smooth muscle cells (SMCs) by targeting IGF2, and drives their differentiation into adipogenic or smooth muscle lineage, respectively. In Experiment 1, SMCs were cultured from mouse aortas and transduced with miR-30e or control lentivirus. In Experiment 2, MSCs were extracted from mouse bone marrow and transduced with miR-30e or control lentivirus. In Experiment 3, the MSCs were transfected with a scrambled oligo or anti-miR-30e oligo. In all 3 experiments, N=3 per group. RNA was extracted from each experiment and run on Affymetrix arrays.

Next Generation Sequencing of the transcriptomes of prostate cancer cells with or without ectopic expression of microRNA miR-30e

Project description:MicroRNAs (miRNAs) function as regulators of cancer progression as they modulate different cellular processes. The objective of this study is to demonstrate a multi-dimensional function of miR-30e through regulating genes involved in various signaling pathways including androgen receptor signaling. miR-30e targets androgen receptor mRNA and functions as a tumor suppressor. We noted loss of miR-30e expression in prostate tumors and restored expression of miR-30e led to cell cycle arrest, induction of apoptosis, improved drug sensitivity in prostate cancer cells, and reduced tumor progression in prostate cancer xenograft models. To understand the tumor suppressor function of miR-30e on a global scale, we performed total RNA-sequence analysis upon overexpression of miR-30e in a prostate cancer cell line 22Rv-1, which expresses a splice variant of androgen receptor and represents an androgen-independent and drug-resistant form of prostate cancer.qRT-PCR validation of RNA-sequencing data supports our in vitro study showeing differential expression of genes involved in cell cycle progression, apoptosis and ubiquitination upon expression of miR-30e in prostate cancer cells.

Network-based integration of multi-omics data identifies the determinants of miR-491-5p effects

Project description:The identification of miRNAs’ targets and associated regulatory networks might allow the definition of new strategies using drugs whose association might mimic a given miRNA’s effects. Based on this assumption our group devised a multi-omics approach in an attempt to precisely characterize miRNAs’ effects. We combined the analysis of miR-491-5p direct targets, and effects at the transcriptomic and proteomic levels. We thus constructed an interaction network which enlightened highly connected nodes, being either direct or indirect targets of miR-491-5p effects: the already known EGFR and BCL2L1, but also EP300, CTNNB1 and several small-GTPases. By using different combinations of specific inhibitors of these nodes, we could greatly enhance their respective cytotoxicity and mimic miR-491-5p-induced phenotype. Our methodology thus constitutes an interesting strategy to comprehensively study the effects of a given miRNA. Also, we identified targets for which pharmacological inhibitors are already available for a clinical use, or in clinical trial phases. This study might thus enable innovative therapeutic options for ovarian cancer, which remains the first cause of death from gynecological malignancies in developed countries.

Synthetic double stranded RNA reliably induces innate immunity similar to a live viral vaccine in humans

Project description:Adjuvants are critical for the success of vaccines, and agonists for microbial pattern recognition receptors are promising new candidates. A mechanism for the immune enhancing role of adjuvants is to stimulate innate immunity. We studied the innate immune response in humans to synthetic double stranded RNA (poly ICLC), a ligand for TLR3 and MDA-5 cytosolic RNA helicase. Transcriptional analysis of blood samples from eight volunteers, after subcutaneous administration of poly ICLC showed upregulation of genes involved in multiple innate immune pathways in all subjects, including interferon and inflammasome signaling. Blocking of type I interferon receptor ex vivo significantly dampened the response to poly IC. Comparative transcriptional analysis showed that several innate pathways were similarly induced in volunteers immunized with the highly efficacious Yellow Fever Vaccine. Therefore a chemically defined microbial agonist like poly ICLC can be a reliable and authentic microbial mimic for inducing innate immunity, here for a live attenuated viral vaccine in humans.
