RNA sequencing of single LN cells sorted from virus-activated CD4 T clusters

ABSTRACT: PA-GFP mice were infected subcutaneously with either rVSV or rLCMV viruses. Two days upon infection, draining popliteal lymph nodes (LN) were photoactivated in areas containing Ag-specific CD4+ T cell clusters. Photoactivated cells were single-cell sorted and RNA sequencing performed to determine cell composition of activated CD4+ T cell clusters.

Memory CD8+ T cells mediate early pathogen-specific protection through localized delivery of antigen-dependent chemokines and IFNgamma to clusters of inflammatory monocytes

Project description:While cognate antigen-driven clonal expansion is a cardinal feature of memory CD8+ T cell effective response, inflammatory cytokines also orchestrates their rapid activation and production of IFNᵧ, contributing to innate mechanisms of protection. Despite antigen-independent secretion, IFNᵧ accounts for a large part of antigen-dependent protection in a multitude of infections, yet how cognate antigen enables memory CD8+ T cell-derived IFNᵧ to achieve host protection remains unknown. Herein, using distinct models of immunization, we establish that cognate antigen (Ag) recognition by memory CD8+ T cells on conventional CD11chi dendritic cells initiates their coordinated production of a burst of chemokines (CCL3, CCL4, XCL1) under the transcriptional control of IRF4. Using intravital microscopy imaging, we further reveal that memory CD8+ T cells undergo Ag-mediated arrest in splenic red pulp clusters of CCR2+ monocytes to deliver them with IFNᵧ- and Ag-dependent chemokine-potentiating microbicidal activities. These results establish that effective memory CD8+ T cell responses require coordinated stepwise processes that quickly restrict pathogen growth and optimize the local delivery of effector molecules before clonal expansion occurs.

Tolerogenic Dendritic Cells Engineered with IL-10 and Antigen Control Pathogenic T Cell Responses and Promote Type 1 T Regulatory Cells

Project description:Dendritic cells (DC) play a critical role in the maintenance of tissue homeostasis and in promoting tolerance, thus acting as regulatory cells. Tolerogenic DC (tolDC) represent the cells of choice to fulfill the goal of restoring antigen (Ag)-specific tolerance since they can dampen Ag-specific T cell responses, induce pathogenic T cell exhaustion, and Ag-specific regulatory T cells. Here we efficiently generate tolDC by genetic engineering of monocytes with bidirectional lentiviral vectors co-encoding for immunodominant Ag-derived peptides (Ovalbumin or Matrix Protein 1, Insulin and Gliadin) in combination with IL-10 (DCIL-10/Ag). DCIL-10/Ag secrete high amounts of IL_x0002_10 in the absence of pro-inflammatory cytokines and efficiently modulate Ag-specific CD4+ and CD8+ T cell activation and proliferation in vitro in healthy subjects and Celiac Disease patients. DCIL-10/Ag promote Ag-specific CD49b+LAG-3 + T cells, which display the type 1 T regulatory (Tr1) cell gene signature in vitro. In vivo administration of DCIL-10/Ag promote Ag-specific Tr1 cells and prevent type 1 diabetes development in two murine models of disease. In conclusion, we generate an innovative approach based on the use of autologous engineered DC, which effectively modulate pathogenic CD4+ and CD8+ T cell responses in vitro and in vivo by promoting Ag_x0002_specific Tr1 cells suitable for cell-based therapy for restoring tolerance in T cell mediated diseases

The expression of miRNAs analyzed by microarray revealed 27 miRNAs exclusively expressed in culture or sorted Th17 Ag-sp cell subset.

Transcriptome analysis of Foxp3- CD25- regulatory T cells

Project description:The concept of immune regulation/suppression has been well-established. With thymus-derived CD4 CD25 regulatory T (TR) cells, it became clear that a variety of additional peripherally induced TR cells play vital roles in protection from many harmful immune responses including intestinal inflammation. In the present study, we have analyzed in vivo-induced Ag-specific CD4 TR cells with respect to their molecular and functional phenotype. By comparative genomics we could show that these Ag-specific TR cells induced by chronic Ag stimulation in vivo clearly differ in their genetic program from naturally occurring thymus-derived CD4 CD25 TR cells. This distinct population of induced TR cells express neither CD25 nor the TR-associated transcription factor Foxp3. Strikingly, CD25 is not even up-regulated upon stimulation. Despite the lack in Foxp3 expression, these in vivo-induced CD25 TR cells are able to interfere with an Ag-specific CD8 T cell-mediated intestinal inflammation without significant increase in CD25 and Foxp3 expression. Thus, our results demonstrate that in vivo-induced Ag-specific TR cells represent a distinct population of Foxp3 CD25 TR cells with regulatory capacity both in vitro and in vivo. Differential Kinetics of Antigen Dependency of CD4+ and CD8+ T Cells

Project description:Ag recognition via the TCR is necessary for the expansion of specific T cells that then contribute to adaptive immunity as effector and memory cells. Because CD4+ and CD8+ T cells differ in terms of their priming APCs and MHC ligands we compared their requirements of Ag persistence during their expansion phase side by side. Proliferation and effector differentiation of TCR transgenic and polyclonal mouse T cells were thus analyzed after transient and continuous TCR signals. Following equally strong stimulation, CD4+ T cell proliferation depended on prolonged Ag presence, whereas CD8+ T cells were able to divide and differentiate into effector cells despite discontinued Ag presentation. CD4+ T cell proliferation was neither affected by Th lineage or memory differentiation nor blocked by coinhibitory signals or missing inflammatory stimuli. Continued CD8+ T cell proliferation was truly independent of self-peptide/MHC-derived signals. The subset divergence was also illustrated by surprisingly broad transcriptional differences supporting a stronger propensity of CD8+ T cells to programmed expansion. These T cell data indicate an intrinsic difference between CD4+ and CD8+ T cells regarding the processing of TCR signals for proliferation. We also found that the presentation of a MHC class II–restricted peptide is more efficiently prolonged by dendritic cell activation in vivo than a class I bound one. In summary, our data demonstrate that CD4+ T cells require continuous stimulation for clonal expansion, whereas CD8+ T cells can divide following a much shorter TCR signal.
