Vehicle or Riociguat oral administation was performed at every 9 hours. 24h later, lungs were dissociated and endothelial cells and pericytes were isolated by flow cytometry, then directly processed to RNA extraction and library construction Results: Riociguat can restore LPS-induced lung injury by inhibiting pericytes activation and maintain vascular homostatsis

Project description:Cullin proteins are scaffolds that coordinate assembly of cullin-RING E3 ubiquitin (Ub) ligases (CRL), complexes that control post-translational ubiquitin modification and degradation of cellular proteins. Cullin-5 (Cul5) coordinates assembly of CRL complexes containing the RING E3 ligase Rbx1/2, the adapter proteins Elongins B and C, and a Suppressor of Cytokine Signalling (SOCS) box-containing substrate recognition protein. To explore potential roles for Cul5, we generated mice lacking Cul5 in the in hematopoietic system. Analyses included biological and molecular studies including proteomic analysis of differential expression of proteins in primary purified hematopoietic stem/progenitor (LSK) cells lacking Cul5.

Influence of the Soluble Guanylate Cyclase Stimulator Riociguat on Myocardial Left Ventricular Proteome and microRNA expression after Experimental Aortic Stenosis in Mice

Project description:We analyzed global proteomic adaptations during heart failure (HF) progression in a mouse model, suffering from left ventricular pressure overload due to transverse aortic constriction (TAC) and in response to riociguat (RIO) treatment, to gain deeper insights in beneficial effects due to sGC stimulation on cardiac remodeling and HF. TAC and sham animals were treated with either riociguat (RIO; 3 mg/kg/day) or vehicle (VEH; Transcutol®/Cremophor®/water: 10%/20%/70%) for five weeks, starting three weeks post-op when cardiac hypertrophy was established, resulting in four treatment groups ( n=5-6 per group). At the end of the study, hearts were dissected and proteomes of the left ventricles (LV) were analyzed by mass spectrometry (MS).

RIOK2-dependent transcriptional changes in primary human hematopoietic stem and progenitor cells (HSPCs)

Project description:Purpose: Illumina next-generation sequencing (NGS) has been used to interrogate the transcriptome profiling (bulk RNA-seq) of primary human HSPCs in the presence and absence of RIOK2. Primary human hematopoietic stem and progenitor cells (HSPCs) isolated from 3 different donors were genome edited to obtain knockdown (KD) and knockout (KO) of RIOK2. The genome edited HSPCs were then differentiated for 48 hours and their total RNA was isolated to perform cDNA synthesis and bulk RNA sequencing. The overall goal of this study was to investigate the global alterations in gene expressions with dose-dependent loss of RIOK2 in primary human HSPCs, that would expand our understanding of RIOK2-dependent transcriptomic changes involved in hematopoietic differentiation.
