PHD inhibition upregulates glycolytic pathway in OGD

ABSTRACT: To elucidate the mechanism of enarodustat pretreatment-induced protection against oxygen-glucose deprivation (OGD), we have we have employed whole genome microarray expression profiling as a discovery platform to identify genes upregulated by enarodustat exposure. HK2 cells were pretreated with either vehicle (DMSO) or enarodustat for 24 hours prior to OGD for 16 hours. Pathway analysis revealed upregulated glysolysis pathway.

Project description:Oxygen-glucose deprivation (OGD) is a cellular phenomenon consistently observed upon occurrence of ischemic stroke which eventually results in neuronal death. Neuronal cell death after ischemia takes place via two distinct processes, necrosis and apoptosis. Apoptosis, programmed cell death, is denoted by chromatin condensation, nuclear blebbing, cellular shrinkage, and DNA fragmentation. Unlike apoptosis, cellular swelling and lysis is suggestive of necrosis. However, these two processes are related not only to the severity but also to the duration of ischemia. Microarray analysis was carried out using 20 Illumina mouse Ref8V1.1 genechip arrays. The assignment of the arrays was as follows: Controls (n=5); exposure to OGD for 10min, 5h, 8h, 15h and 24 h (n=3 respectively).

RNA‑seq transcriptomic analysis of HL‑1 cardiomyocytes under OGD/R injury with or without Phillyrin treatment

Project description:Ischemia‑reperfusion (I/R) injury is a key pathological process in myocardial infarction, for which effective therapies are still lacking. Phillyrin has been reported to exert cardioprotective effects, but its molecular mechanisms remain unclear. HL‑1 mouse cardiomyocytes were subjected to oxygen‑glucose deprivation/reoxygenation (OGD/R) to mimic I/R injury. Cells were divided into two groups: OGD/R alone (IR) and OGD/R with Phillyrin treatment (IR+Phil). Total RNA was extracted from 3 biological replicates per group and sequenced using Illumina NovaSeq 6000. Transcriptomic profiling identified differentially expressed genes and pathways regulated by OGD/R and Phillyrin treatment. The processed data file contains raw read counts and FPKM values for all genes across all samples. This dataset provides a comprehensive transcriptomic resource to explore the molecular mechanisms of Phillyrin‑mediated cardioprotection against I/R injury.

Effects of imiquimod on macrophages by transcriptomic analysis

Project description:Imiquimod is a small-molecule immunomodulator with anti-tumor effects, and its anti-tumor activity mainly relies on its immunomodulatory capacity, including activating dendritic cells， T cells, natural killer cells, and promoting the migration of antigen-presenting cells. In this study, we pretreated mouse bone marrow-derived macrophages (BMDMs) with imiquimod for 24 hours and conducted transcriptomic analysis to explore the mechanism by which imiquimod regulates macrophages to exert anti-tumor effects. We found that imiquimod pretreatment of macrophages upregulated multiple signaling pathways, with the most significant being immune regulation-related signaling pathways. Notably, pathways such as the NF-κB pathway, Toll-like receptor (TLR) pathway were significantly upregulated. We infer that imiquimod activates TLR pathway and the downstream NF-κB pathway, promoting the polarization of macrophages to a pro-inflammatory phenotype and exerting anti-tumor effects.

The Atypical Antidepressant Tianeptine Confers Neuroprotection against Oxygen-Glucose Deprivation

Project description:Proregenerative and neuroprotective effects of antidepressants are an important topic of inquiry in neuropsychiatric research. Oxygen-glucose deprivation (OGD) mimics key aspects of ischemic injuryin vitro. Here, we studied the effects of 24-h pretreatment with serotonin (5-HT), citalopram (CIT), fluoxetine (FLU), and tianeptine (TIA) on primary mouse cortical neurons subjected to transient OGD. 5-HT (50 μM) statistically significantly enhanced neuron viability as measured by MTT assay and reduced cell death and LDH release. CIT (10 μM) and FLU (1 μM) did not increase the effects of 5-HT and neither antidepressant conferred neuroprotection in the absence of supplemental 5-HT in serum-free cell culture medium. By contrast, pre-treatment with TIA (10 μM) resulted in robust neuroprotection, even in the absence of 5-HT. Furthermore, TIA inhibited mRNA transcription of candidate genes related to cell death and hypoxia and attenuated lipid peroxidation, a hallmark of neuronal injury. Finally, deep RNA sequencing of primary neurons subjected to OGDdemonstrated that OGD induces many pathways relating to cell survival, the inflammation-immune response, synaptic dysregulation and apoptosis, andthatTIA pretreatment counteracted theseeffects of OGD. In conclusion, this study highlights the comparative strength of the 5-HT independent neuroprotective effects of TIA and identifies the molecular pathways involved.

Immunomodulatory Effects of Yeast Cell Wall Compounds on mouse BMDM

Project description:Since few years, yeast cell wall components and especially beta-glucans (BG) have been identified as potential stimulators of the innate immune system in murine and human species. A large screening of crude and BG-enriched components from Saccharomyces cerevisiae has been performed on murine macrophages. A priming effect with bacterial ligands was revealed in terms of pro-inflammatory cytokines release. To further characterize the responsiveness of bone marrow derived-macrophages which express high levels of dectin-1, the major receptor of BG, transcriptional analysis would provide more hypotheses about the genes and signaling pathways triggered by yeast cell wall compounds in macrophages. Gene expression profiling was performed in murine BMDM pretreated with a set of three BG-containing cell wall compounds and then stimulated with LPS. Conditions with BG-enriched pretreatment clustered together in contrast to the crude compound and mock pretreatment conditions that remained separated whatever the time point analyzed, with a greater number of differentially-expressed genes following the crude compound treatment compared to BG-enriched pretreatment. BG-enriched pretreatment induced a specific set of genes in mouse macrophages, involving the PI3K/AKT signaling pathway. Among them, the two main upregulated genes by BG-enriched priming condition were considered for further analysis and their associated-protein production were consistently increased in BMDM pretreated with BG-enriched compound. Microarray results were confirmed using RT-qPCR on a different set of samples.
