Outer Membrane Vesicles Secreted by Bacteroides fragilis Inhibit CFTR Chloride Secretion by Human Colon Organoids

Project description:The goals of this study were to develop a model to study host pathogen interactions in primary human colon organoids and to test the hypothesis that Bacteroides fragilis toxin (BFT-2) secreted in outer membrane vesicles (OMVs) modulates mucosal immunity and CFTR Cl- secretion. Since Bacteroides species often resides in mucus, OMVs are likely to represent a mechanism of communication between Bacteroides and the host. Two strains of Bacteroides were studied, Enterotoxigenic Bacteroides fragilis (ETBF), which produces BFT-2, and the non-toxigenic Bacteroides fragilis strain NCTC 9343 (NTBF) that does not produce BFT-2. We also utilized two additional strains of Bacteroides fragilis: one in which bft-2 was knocked out (ETBF Δbft), and one that was engineered to contain bft-2 (NTBF+bft). We report that most Bacteroides fragilis OMVs reduced CFTR Cl- secretion but had no effect on tight junction or cell adhesion proteins, transepithelial resistance (TER) or cytokine secretion by primary human colon organoids. We conclude that OMVs secreted by Bacteroides can be an important mechanism of host pathogen interactions in the colon by reducing CFTR Cl- secretion.

Bacteroides fragilis toxin promotes gallbladder cancer in mice

Project description:Enterotoxigenic Bacteroides fragilis (ETBF), which secretes B. fragilis toxin (BFT) and has been associated with colorectal cancer, can be enriched in patients with gallbladder cancer (GBC). Whether and how ETBF contributes to GBC is unclear. Here we confirm using analysis of patient samples that ETBF is enriched in GBC tumors, while experiments in mice show that ETBF colonises the gallbladder. In vitro, and in patient-derived organoids, ETBF promoted GBC proliferation. ETBF also promoted tumor growth in a BFT-dependent manner in a mouse GBC allograft model. Mechanistically, the tumorigenic activity of BFT was dependent upon the ETBF surface protein, MltD, which interacts with the host receptor protein, TMPRSS13. This interaction activated JAK2-STAT3 signaling to promote tumorigenesis. BFT also activated NF-κB signaling, which increased CXCL1 secretion, leading to myeloid-derived suppressor cells recruitment and angiogenesis in the tumor microenvironment. Taken together, these findings uncover mechanisms through which ETBF facilitates GBC development, with potential promise as therapeutic targets to limit GBC progression.

Bacteroides Fragilis enterotoxin engagement of the colonic epithelium is associated with early YAP1 activation and colon tumorigenesis

Project description:Enterotoxigenic Bacteroides fragilis (ETBF) promotes colonic inflammation and tumorigenesis, but the epithelial signaling events linking toxin (BFT2)-induced junctional injury to tumor development remain incompletely defined. Here, wild-type BFT2, but not catalytically inactive BFT2, induced E-cadherin loss and nuclear accumulation of active YAP1 in colonic epithelial cells. Deletion of BFT receptors, CLAUDINs 3 and 4 mitigated BFT-associated E-cadherin loss and YAP1 nuclear translocation, supporting a model in which BFT epithelial targeting is linked to early YAP1 activation. In ETBF-colonized Min mice, epithelial YAP1 activation was rapid and transient, whereas STAT3 and NF-kB activation persisted after colonization. Nevertheless, conditional deletion of epithelial Yap1 was sufficient to abrogate colon tumor formation without altering ETBF colonization or abolishing IL-17 colitis. Transcriptomic analyses further associated epithelial Yap1 deficiency with reduced regenerative epithelial programs and increased secretory-cell differentiation signatures. Together, these findings support a model in which BFT engagement of CLAUDINs and toxin catalytic activity are associated with epithelial YAP1 activation, and epithelial YAP1 contributes to ETBF-associated colon tumorigenesis in Min mice
