Project description:End-stage heart failure has long been regarded as a terminal state of cardiac pathological remodeling that is almost impossible to be reversed by any available therapies.We discovered that short term estrogen (E2) treatment rescues preexisting severe heart failure (HF) induced by trans-aortic constriction (TAC) in mice by restoring ejection fraction (EF) from 30% to 55% and cardiac fibrosis (Iorga A, Li J, Sharma S, Umar S, Bopassa JC, Nadadur RD, et al. Rescue of Pressure Overload‐Induced Heart Failure by Estrogen Therapy. Journal of the American Heart Association [Internet]). Given the role of microRNAs in wide spectrum of physiological and pathological processes in HF and fibrosis, we explored whether E2 rescues HF and fibrosis by controlling the expression of specific microRNAs (miRs). RNA isolated from cardiac left ventricle of mice in sham (n=4), HF (n=5) and E2-rescued (n=5) groups was sent to Ocean Ridge Biosciences for miR microarray analysis. Comparison of E2-Rescued group versus HF and sham groups revealed a subset of 158 miRs significantly regulated by E2.

Project description:Estrogen clearly prevents osteoporotic bone loss by attenuating bone resorption. The molecular basis of how this is accomplished, however, remains elusive. Here we report a critical role of osteoclastic ERa in mediating estrogen action on bone in females. We selectively ablated ERa in differentiated osteoclasts (ERa dOc/dOc). ERa dOc/dOc females, but not males, exhibited clear trabecular bone loss, similar to the osteoporotic bone phenotype in post-menopausal women. Recovery of bone loss by estrogen treatment of the ovariectomized ERa dOc/dOc females was ineffective in the trabecular areas of the long bones and lumbar vertebral bodies. Osteoclastic apoptosis, induced by estrogen, occurred simultaneously with up-regulation of Fas ligand (FasL) expression in intact trabecular bones of ERa +/+mice, but not in ERa dOc/dOc mice. ERa was also required for similar effects of estrogen and tamoxifen in cultured osteoclasts. These findings suggest that the osteoprotective actions of estrogen and SERMS are mediated at least in part through osteoclastic ERa in trabecular bone; and the life span of mature osteoclasts is regulated through activation of the Fas/FasL system. Project description:Estrogen, specifically estradiol, has been shown to mediate DNA methylation changes at mutiple locations downstream of estrogen response elements (EREs). We identified differentially methylated regions associated with long-term estradiol exposure in the hippocampus of C57BL/6J mice. Hippocampus was collected from ovariectomized (OVX) C57BL/6J mice treated with estradiol (n=5) and vehicle controls (n=5). Genomic DNA was extracted and treated with methylation sensitive restriction enzymes HpaII and HinP1I to enrich for the unmethylated fraction. This fraction was interrogated using the Affymetrix GeneChip® Mouse Tiling Promoter 1.0R Array

Project description:This laboratory studies the structural basis of carbohydrate function in terms of adhesion and other biological processes. The Fisher lab is testing the hypothesis that the mucin-coated oral and uterine cavities present similar carbohydrate receptors that specify the bacterial ecology of both regions and the repertoire of these oligosaccharide species is hormonally regulated. This theory also suggests that certain individuals express carbohydrate receptors that make them susceptible to both periodontal disease and preterm labor. Experimental procedure: 40 mature female mice were ovariectomize . The mice were allowed to rest for two weeks to eliminate any remaining endogenous estrogen (E2) and progesterone (P4). Then the mice were separated into four groups (10 each) that received the treatment indicated. Group 1: ovariectomized mice with no supplement, only sesame oil vehicle. Group 2: ovariectomized mice with only P4 supplement (2 mg/day/mouse). Group 3: ovariectomized mice with E2 supplement (100 ng/day/mouse). Group 4: ovariectomized mice with both P4 (2 mg/day/mouse) and E2 (100 ng/day/mouse) supplement. The treatment lasted for 4 days; the steroids were dissolved in sesame oil and injected subcutaneously. After treatment, salivary glands (parotid, submandibular, and sublingual gland), submandibular lymph nodes and uterine horns with cervix were collected. Total RNA was extracted from these samples using Trizol.
