Comparative RNA-seq analysis of murine lung infected with A.fumigatus

ABSTRACT: We report the application of RNA-seq analysis for high-throughput profiling of murine lungs infected with Aspergillus fumigatus. We compared the lung transcription of wildtype murine lungs and lungs from mice deficient in metabolic cytokine adiponectin.

Project description:In lung diseases caused by the major mould pathogen Aspergillus fumigatus the pulmonary epithelium is destroyed by invasive growth of fungal hyphae, a process thought to require fungal proteases. Here we show that the A. fumigatus pH-responsive transcription factor PacC governs expression of secreted proteases during invasive lung infections and is required for epithelial invasion and pathogenicity. In addition, A. fumigatus M-NM-^TpacC mutants aberrantly remodel the fungal cell wall during infection. In order to characterise the basis of differential Olorofim susceptibility, and distinguish which metabolic pathways these transcription factors regulate, RNA-seq analysis was used to compare these transcription factor knockout mutants to wildtype A. fumigatus in both the presence and absense of Olorofim.

Differential gene expression in human myeloid dendritic cells interacting with different knockout strains of Aspergillus fumigatus in vitro

Project description:Dendritic cells (DC) are the most important antigen presenting cells and play a pivotal role in host immunity to infectious agents by acting as a bridge between the innate and adaptive immune systems. Myeloid DCs (mDC) were isolated from peripheral blood and co-cultured with conidia (con) and germlings (ks) of Aspergillus fumigatus knockout (mitA, mnt1, rodA) and wildtype (wt) strains for 6 hours at an MOI of 1. RNA was extracted and hybridised to microarrays. Microarrays contain probes for approximately 110 selected human immune-relevant genes, including cytokine and chemokine genes, their receptors and downstream innate immunity genes. Effect of adiponectin deficiency on pulmonary responses to subacute ozone exposure in mice

Project description:Adiponectin is an adipose-derived hormone with anti-inflammatory activity. Following subacute ozone exposure (0.3 ppm for 24-72 h), pulmonary neutrophilic inflammation is augmented in adiponectin deficient mice. The purpose of this study was to use microarrays to examine the impact of adiponectin deficiency on changes in pulmonary gene expression induced by ozone, a common air pollutant. Lungs were harvested from wildtype and mice that were genetically deficient in adiponectin. Mice were exposed either to room air or to ozone (0.3 ppm) for 72 h. RNA was extracted and microarray analysis of gene expression performed. Both male and female mice were used.

Protein kinase A regulated genes of Aspergillus fumigatus -FEBIT transcriptome analysis

Project description:Aspergillus fumigatus is a ubiquitous mould but also an opportunistic human pathogen causing life-threatening infections in immunocompromised patients. Survival of the fungus in different habitats depends on effective mechanisms of signal perception and transduction such as the cAMP dependent protein kinase A (PKA) pathway, which is involved in virulence of A. fumigatus. Here, by transcriptome analysis putative targets of this important signaling cascade were identified, revealing 632 differently regulated genes including 23 putative transcriptional regulators. The highest up-regulated transcription factor gene was located in the until now unknown fmp secondary metabolite gene cluster encoding an incomplete non-ribosomal peptide synthetase as core enzyme. Overexpression of the fmp cluster resulted in formation of fumipyrrole, which was not described as natural product yet. Although genes of the fmp cluster are transcribed in infected mouse lungs, deletion of its regulatory gene fmpR resulted in wild-type virulence in a murine infection model.
