RNA-seq of primary human aortic smooth muscle cells treated with AEA

ABSTRACT: Primary human aortic smooth muscle cells (HAoSMC) were starved over night in 1% FCS. On the next day cells were treated with 100nM Anandamide (AEA) or EtOH as control for 4h in presence of 10µM diclofenac. Subsequently cells were treated with or without 10ng/mL IL-1β for last 1.5h.

Profiling the anandamide reuptake inhibitor WOBE437

Project description:Anandamide, or N-arachidonoylethanolamine (AEA), is a signaling lipid that modulates neurotransmitter release via activation of the cannabinoid receptor type 1 (CB1R) in the brain. Termination of anandamide signaling is thought to be mediated via a facilitated cellular reuptake process utilizing an unknown transporter protein. Recently, WOBE437 has been reported as a novel, natural product-based inhibitor of AEA reuptake that is active in cellular and in vivo models. To profile its target interaction landscape, we synthesized pac-WOBE, a photoactivatable probe derivative of WOBE437, and performed chemical proteomics in mouse neuroblastoma Neuro-2a cells. Surprisingly, WOBE437, but not OMDM-1, was found to increase AEA uptake in Neuro-2a cells. In line, WOBE437 reduced cellular levels of AEA and related N-acylethanolamines (NAEs). Using pac-WOBE, we identified saccharopine dehydrogenase-like oxidoreductase (SCCPDH), vesicle amine transport 1 (VAT1) and ferrochelatase (FECH) as WOBE437-interacting proteins in Neuro-2a cells

2-arachidonoyl glycerol (2-AG) modulated miRNAs expression in human cholangiocarcinoma

Project description:The endocannabinoid system (ECS) is dysregulated in various liver diseases. Biliary tract cancers (BTCs) encompass gallbladder carcinoma, intrahepatic, perihilar and distal cholangiocarcinoma. Previously we have shown that the two major endocannabinoids (ECs), anandamide (AEA) and 2-arachidonoyl glycerol (2-AG) exerted opposing effects on BTCs cell lines in which 2-AG promoted tumor proliferation. The underlying mechanism and regulation of 2-AG on miRNAs expression in BTCs remain inconclusive.

Cannabinoids modulate proliferation, differentiation, and migration signaling pathways in oligodendrocytes

Project description:To elucidate the specific molecular signals associated with the activation or blockade of CB1 and CB2 receptors in this glial cell, mass spectrometry-based proteomics and in silico biology tools were used to determine which signaling pathways and molecular mechanisms are triggered in a human oligodendrocytic cell line (MO3.13) by several pharmacological stimuli: the phytocannabinoid cannabidiol (CBD); CB1 and CB2 agonists WIN55,212-2, ACEA, and HU308; CB1 and CB2 antagonists AM251 and AM630; and endocannabinoids anandamide (AEA) and 2-arachidonoylglycerol (2-AG).

Transcriptional profiling of antiestrogen-sensitive and -resistant MCF-7 cells

Project description:Antiestrogen-responsive (#Wt), tamoxifen-(#TamR1/8) and fulvestrant-(#FulvR6/7) cells were routinely maintained in DMEM/F12 (phenolred-free) with 1% FCS, glutamine (2mM), penicillin/streptomycin (1%) and insulin (6ng/ml). Selection was done in 1ÂµM tamoxifen or 100nM fulvestrant, respectively. For microarray studies, cells were in parallel cultured under steroid-depleted conditions: DMEM/F12 [1% charcoal-stripped, steroid-depleted FCS [CCS: (phenolred-free) with 1% FCS, glutamine (2mM), penicillin/streptomycin (1%) and insulin (6ng/ml)) for 3d and under steroid-depleted conditions treated for 1h with vehicle (DMSO), 4OHT (1ÂµM) or fulvestrant (100nM), respectively.

Effect of cabozantinib on transcriptome and signaling pathways in leukemic cell line Kasumi-1

Project description:To identify molecular effects of the tyrosine kinase inhibitor cabozantinib (XL184), we applied gene expression profiling on human acute leukemic cell lines Kasumi-1, which harbors t(8;21) coupled with KIT mutation, by Whole Transcriptome Shotgun Sequencing (RNA-seq). After Kasumi-1 cells were treated with 100nM cabozantinib for 4h or 24h, differentially expressed genes (DEGs) of 124 genes upregulated and 416 were downregulated in 4h, and 184 genes upregulated and 69 genes were downregulated in 24h treatment, compared with vehicle group.

Genome-wide changes in OGT-depleted mouse adipose tissue

Project description:Appetite control is key to combat obesity in an over-nutritious environment. Whether and how over-nutrition induces hyperphagia and obesity through the adipose-to-brain axis is unclear. O-linked beta-D-N-acetylglucosamine (O-GlcNAc) transferase (OGT) couples nutrient cues to O-GlcNAcylation of intracellular proteins at serine/threonine residues. Here we show that adipocyte OGT is essential for high fat diet-induced hyperphagia, but is dispensable for baseline food intake. Adipocyte OGT stimulates hyperphagia by transcriptional activation of de novo lipid desaturation and accumulation of anandamide (AEA), an endogenous appetite-inducing cannabinoid (CB). Pharmacological manipulation of peripheral CB1 signaling regulates hyperphagia in an adipocyte OGT-dependent manner. These findings define adipocyte OGT as a fat sensor that regulates peripheral lipid signals, and uncover an unexpected adipose-to-brain axis that induces hyperphagia and obesity.
