Project description:Time course of early development of peripheral nerve, from embryonic day 9.5 to postnatal day 0. Origin of samples: mouse embryos expressing green fluorescent protein (GFP) in neural crest stem cells and later glial cells under the control of the proteolipid protein gene (Plp) promoter. At E9.5 the trunk region was dissected out. At E12, E14, E16, E18 and P0 the sciatic nerve was taken. GFP-expressing cells were isolated by FACS sorting. Three or 4 separate batches of embryos analyzed at each stage.

Single cell RNA sequencing analysis using embryonic day 9.5 placenta from control and GATA DKO mice

Project description:The placenta establishes a maternal–fetal exchange interface which ensures transportation of nutrients and gases between the mother and the fetus. A critically conserved step in the establishment of this exchange surface in mammals is the fusion of trophoblast cells into a multinucleated syncytiotrophoblasts (SynT). In mice, SynTs develop via differentiation of the labyrinth trophoblast progenitors (LaTP) of the developing placenta, and in humans, SynTs develop via differentiation of villous cytotrophoblast (CTB) progenitors. Despite being an indispensable step for mammalian development in utero, conserved molecular mechanisms that ensure SynT development are poorly understood. Herein, we show that GATA2 and GATA3 (GATA factors) plays an essential role in labyrinth trophoblast progenitors (LaTPs) thereby committing these cells towards the SynT differentiation program. Mature syncytial layer specific immunostaining with established markers, and ultrastructure analysis of the labyrinth revealed that the loss of GATA factors was impairing the formation of mature SynT cells. We therefore focused our attention on the status of the labyrinth progenitor trophoblast cells in the GATA DKO vs the control placentae. To do that we performed single cell RNA Sequencing (scRNA-Seq) with E9.5 control and Gcm1Cre GATA KO placentae.

Project description:We employed RNA-seq to transcriptionally profile a pure population of hand-dissected polyploid TGCs from embryonic day 9.5. These data provide a set of polyploid-specific TGCs transcripts that will aid in the understanding of TGCs differentiation and endoreplication. TGCs were micro-dissected from day E9.5 nine implantation sites from C57BL/6J mice. The portion of the TGCs in direct contact with the spongiotrophoblast layer and the labyrinth layer were manually removed to avoid collecting any polyploid cells from the former or multi-nucleated syncytiotrophoblast cells from the latter.

RNA sequence of embryonic dorsal skin and AGM (Aorta-gonad-mesonephros) tissues

Project description:Purpose: The goals of this study are to identify the different gene expresion level in dorsal skin cells or AGM region at different embryonic stages Methods: Dorsal skin cells were collected from different-staged embryo: E9.5, E10.5 E12.5 and E14.5. AGM were collected from different-staged embryo: E9.5, E10.5 and E11.5. In duplicate. RNA were extrated and analyzed using RNA sequencing. For dorsal skine tissues, we screened the genes were highly expressed at early embryonic stage E9.5, but not later stages (E10.5, E12.5, and E14.5). For embryonic AGM tissues, we selected the genes were highly expressed at early embryonic stage E9.5, but not later stages (E10.5 and E11.5). The gene list were filtered based on secreted potein candidates. These gene candidates might function as novel embryonic anti-tumor factors Results: Using an optimized data analysis workflow, we mapped the gene list for potential secretory anti-tumor factors. Conclusions: One of the top gene from the gene lists is NEPN which we hypotherze it has unique anti-tumor function in the early embryonic microenvirnemt and might be used as a novel cancer therapeutic drug

Project description:Transcriptional profiling of E9.5 mouse embryo tissue from the presomitic mesoderm (PSM) and somites I-IV. Tissue from embryos lacking a functional Paraxis gene (Paraxis-/-) was compared to identical tissue from E9.5 Wild Type embryos. The goal was to identify genes that had become deregulated in the absence of the transcription factor, Paraxis. Two-condition experiment: WT vs Paraxis-/- tissue. Biological replicates: 3 pools of 5 WT samples, 3 pools of 5 Paraxis-/- samples. Technical replicates: 3 dye swaps.
