LSD1 regulates the plasmablast transcriptome

ABSTRACT: To understand the role of LSD1 in marginal zone B cell development, CD93+ transitional B cells were enriched from the spleens of mice with B cell conditional deletion of LSD1, the cells were cultured for 3 days in the presence of OP9-DL1 cells and BAFF, and day 0 B220+CD93+ transitional B cells and day 3 B220+CD21+CD1d+ ex vivo-derived marginal zone B cells were FACS-sorted and RNA-seq was performed to identify LSD1-target regulated genes.

Project description:Splenocytes were FACS-sorted from Wild-type and Mir146a-/- mice to isolate specific B-cell developmental stages. Utilizing high-throughput sequencing, we comparatively analyzed developmental stage-specific splenic B cell transcriptomes, including Transitional-1 (T1), Transitional-2 (T2), Marginal zone (MZ) and Follicular (FO) in both Wild-type and Mir146a-/- B cells. Two replicates of each developmental stage were submitted for high-throughput sequencing.

Project description:We have analyzed miRNA expression profile in FO and MZ B cells to identified differentially expressed miRNAs between this two subsets that could be potencially involved in the regulation of terminal B cell differentiation Spleens from CD19-Creki/+Dicerfl/+ and CD19-Creki/+Dicerfl/fl mice were collected and after erythrocyte lysis, splenocytes were stained with anti-B220, anti-CD21 and anti-CD23 antibodies. B220+CD21brightCD23+ (MZ) and B220+CD21+CD23bright (FO) B cell populations were sorted and total RNA of purified populations was extracted with TRIzol. miRNA microarray hybridations were performed on Mouse miRNA Microarray platform (Agilent Technologies).

The proteome of B cell maturation identifies PDCD4 as limiting the response of marginal zone B cells – B cell subsets. Includes Fasta file.

Project description:During B cell maturation, transitional and mature B cells acquire cell-intrinsic features that determine their ability to exit quiescence and mount effective immune responses. We used high-resolution mass spectrometry to quantify the proteome of B cell subsets from the mouse spleen and map the differential expression of environmental sensing, transcription- and translation initiation-factors that define cellular identity and function. By comparing transcriptome and proteome, we identified mRNAs linked to B cell activation and differentiation that are expressed without detectable protein. These "poised" mRNAs might enable rapid protein production through increased translation or protein stability. In addition, we found that the translational repressor PDCD4 restrains the response of marginal zone B cells to a T-independent antigen. Our molecular characterization of B cell maturation is a valuable resource to further explore the mechanisms underpinning the specialised functions of B cell subsets.
