IL15 Deficiency in Rats

Project description:NK cells are a major immune cell in the uterus during pregnancy. IL15 is required for the maturation of these cells, therefore a mutation in the IL15 gene results in a lack of NK cells. In this study, implantation sites were collected from gestational day 9.5 pregnant IL15 deficient or WT Sprague-Dawley Holtzman rats. RNA was collected, and a Clariom S microarray was performed to elucidate differences in gene expression at the developing placenta and uterus between mutants and WT dams.

RNA-seq identifies IL15-induced adipogenesis and fatty acid metabolism pathways in human adipocytes

Project description:IL15 has been suggested to reduce WAT mass in obese mice models. However, the underlying molecular mechanism is largely unknown. We determined the gene expression profile of human adipocytes treated with vehicle or IL15. Adipocytes isolated from healthy donors exhibited reduced expression of gene signatures related to adipogenesis and fatty Acid metabolism upon IL15 stimulation, which is consistent with the previous notion that IL15 exhibits an anti-obesity effect. Taken together, our findings suggested that SNORD46 physically interact with IL15 and acts as a natural IL15 antagonist in the serum of donors with obesity to hinder the anti-obese effect of IL15.

Multinucleated Tartrate Resistant Acid Phosphatase Positive Cells with Macrophage and Osteoclast Phenotypes in Bone

Project description:Prothrombin (PT) and osteopontin (OPN) promotes adhesion of different TRAP-positive multinucleated cells isolated from rat long bone (Hu et al. Exp Cell Res. 2008; 314: 638-50). The PT-adhering cell could represent either an immature precursor to the OPN-adherent osteoclast or constitute a distinct multinucleated cell population with a unique role in bone. Herein, phenotypic differences between PT- and OPN- cells were investigated with microarray- expression analysis. Characteristic for PT-cells was expression of innate immune response genes and scavenger receptors whereas OPN-cells expressed typical osteoclast proteins such as collagenases implicated in bone degradation.

Regulatory role of multinucleated giant cells derived from placental CD14+ macrophages: Pathophysiological implications

Project description:The role of placental macrophages is largely ignored in the success of pregnancy. We found that CD14+ macrophages purified from at-term placentas spontaneously matured into multinucleated giant cells (MGCs). MGCs lost the expression of CD14 and co-inhibitory molecules, such as Programmed cell Death-Ligands 1 and 2. Although MGCs kept phagocytosis and property to produce ROS, their inflammatory potential measured by TNF/IL-10 imbalance and activation of p38 MAPK and NF-M-NM-:B was blunted without being associated with M2 phenotype. The investigation of gene expression revealed the enrichment with categories related to inflammation, apoptosis and canonical functions of macrophages in MGCs. Whereas most of the genes associated with inflammation and immune responses were down-modulated, those such as VEGFC or associated with matrix remodeling were specifically up-regulated in MGCs. Importantly, we found that patients with preeclampsia or chorioamnionitis, two inflammatory and infectious pathologies, respectively, that affect placentas, were unable to generate MGCs. Taken together, these results suggest that MGCs represent a new way to regulate the inflammatory and cytocidal activity of placental macrophages in a context that imposes contradictory constraints, such as semi-allograft acceptance and defense against aggression. The dysregulation of MGC formation may be associated with placental inflammatory and infectious pathologies.
